Gastric cancer is one of the most prevalent cancers in Korea, which has been studied in many different ways to improve the therapeutic outcome, but it is not yet satisfactory. It is clearly important to develop new therapeutic modalities such as gene therapy for improving the therapuetic outcome of gastric cancer besides the conventional modalities such as operation, chemotherapy or radiation therapy. This study was conducted to evaluate the effectiveness of retroviral-mediated p53 tumor suppressor gene therapy in gastric cancer.
Ten human gastric cell lines were cultured and their DNAs were analysed with PCR-SSCP(polymerase chain reaction - single strand conformation polymorphism) and DNA sequencing. After construction of retroviral vector LNC/p53 containing human p53 cDNA and master packaging cell line PA 317/LNC/p53, the titer of recombinant retrovirus was measured. To investigate the effect of retroviral-mediated p53 gene transduction in human gastric cancer cell lines, the growth rates of broth N-87 cell line with mutant p53 and YCC-S-2 cell line with normal p53 were compared before and after infection with LNC/p53 retrovirus in vitro and in vivo.
The following results were obtained.
1) In PCR-SSCP, 6 out of 10 human gastric cancer cell lines had the mutant p53 gene, which was reconfirmed by nucleotide sequencing.
2) The growth inhibition of N-87 cells after p53 transduction using retroviral vector was significant when compared to the parent N-87 cells, which was dependent on the retroviral titer.
3) The growth of both p53 transduced YCC-S-2 cells and the parent YCC-S-2 cells were not different.
4) In nude mice, growth of the tumors formed by N-87 cells were slightly inhibited after retroviral-mediated p53 gene transduction. Apoptotic changes were more prominent in tumors which were infected with p53 containing retrovirus as compared to those of p53 non-containing retrovirus under light and electron microscopes.
5) The growth of tumors formed by YCC-S-2 cells were not inhibited by p53 transduction in nude mice.
6) The expression rate of p53 protein after p53 containing retroviral infection in KATO-โ
ข cell line, which showed the deletion of p53 gene, was dependent on LNC/p53 retroviral titer. However, the expression rate was less than 5% with the titer of 100 m.o.i.(multiplicity of infection)/cell upon immunohistochemical analysis.
In conclusion, the growth inhibition by retroviral-mediated p53 transduction in human gastric cancer cell lines was significant in gastric cancer cell line with mutant p53 in vitro, and the growth of turners formed by gastric cancer cell line with mutant p53 was slightly inhibited after p53 transduction using retroviral vector in nude mice. Only slight inhibition of tumor growth in nude mouse was thought to be due to low transduction efficiency.restrictio

The relationship between family function and sexual behavior among the unmarried

We examined 288 unmarried medical college students to assess the relationship
between family function and sexual behavior including sexual knowledge, attitude
and experience, using questionnaries of family APGAR score, FACES โ
ข, amount of sex
communication held between parents and students, and family attitudes. The results
are as the following:
1. There were no statistically significant relationship between family APGAR
score and sexual behavior, but the sexual behavior and the APGAR score were related
when considering the family attitudes. In conservative families, the students
showed more conservative attitudes and less sexual expriences if they belonged to
healthy families with high family APGAR scores. If the family attitudes were
liberal and they have a higher APGAR score, they showed more liberal sexual
attitudes and also had more sexual experiences.
2. The sexual experiences and attitudes were not directly influenced by the
amount of sex communication held within the family. But, when considering the
family attitudes, they showed the relationship. In the conservative families, the
sexual behavior seemed more conservative if sex communications were held in the
families. In the families with liberal attitudes, the sexual behavior seems more
liberal if sex communications were held among family members.
3. The families with higher scores on family cohesion showed less sexual
experience and a more conservative sexual attitude.
4. The family adaptability and sexual behavior showed no relationship, but only
in liberal families, there existed more sexual experiences if they had higher
scores on family adaptability.
