    Strengthening Chemical Accident Prevention and Preparedness in European Neighbour Countries. Prepared for DG-ECHO by the Major Accident Hazards Bureau and the Natech Project

    The Seveso Directive aims at control of major hazards associated with dangerous substances and has existed as legislation in the European Union since 1982. It was the first comprehensive legislation of its kind in the world and continues to be a leading model for process safety governance globally. In 2014 DG-ECHO and the Joint Research Centre agreed on collaboration the implementation of the Seveso Directive by European Neighbourhood Policy countries in support to the Civil Protection Mechanism 2014-2010. The Year 1 strategy for this project aimed at building profiles of country progress in establishing chemical accident and prevention programmes. It also began the first phase in making a selection of analytical tools accessible to support government and industry in their efforts to perform hazard identification and risk analysis and understand results. This document proposes a strategy for Year 2 of the project. The proposal describes a strategy targeted to establish and maintain visibility and direct communication on the project with EU Neighbour Countries, determine a first subset of countries for in order to begin targeted capacity building, and to complete a first set of online tool prototypes for implementation support as well as identify opportunities for further tool development in future.JRC.G.5-Security technology assessmen

    Lacticaseibacillus rhamnosus CRL1505 Peptidoglycan Modulates the Inflammation-Coagulation Response Triggered by Poly(I:C) in the Respiratory Tract

    Lacticaseibacillus rhamnosus CRL1505 beneficially modulates the inflammation-coagulation response during respiratory viral infections. This study evaluated the capacity of the peptidoglycan obtained from the CRL1505 strain (PG-Lr1505) to modulate the immuno-coagulative response triggered by the viral pathogen-associated molecular pattern poly(I:C) in the respiratory tract. Adult BALB/c mice were nasally treated with PG-Lr1505 for two days. Treated and untreated control mice were then nasally challenged with poly(I:C). Mice received three doses of poly(I:C) with a 24 h rest period between each administration. The immuno-coagulative response was studied after the last administration of poly(I:C). The challenge with poly(I:C) significantly increased blood and respiratory pro-inflammatory mediators, decreased prothrombin activity (PT), and increased von Willebrand factor (vWF) levels in plasma. Furthermore, tissue factor (TF), tissue factor pathway inhibitor (TFPI), and thrombomodulin (TM) expressions were increased in the lungs. PG-Lr1505-treated mice showed significant modulation of hemostatic parameters in plasma (PT in %, Control = 71.3 ± 3.8, PG-Lr1505 = 94.0 ± 4.0, p < 0.01) and lungs. Moreover, PG-Lr1505-treated mice demonstrated reduced TF in F4/80 cells from lungs, higher pro-inflammatory mediators, and increased IL-10 compared to poly(I:C) control mice (IL-10 in pg/mL, Control = 379.1 ± 12.1, PG-Lr1505 = 483.9 ± 11.3, p < 0.0001). These changes induced by PG-Lr1505 correlated with a significant reduction in lung tissue damage. Complementary in vitro studies using Raw 264.7 cells confirmed the beneficial effect of PG-Lr1505 on poly(I:C)-induced inflammation, since increased IL-10 expression, as well as reduced damage, production of inflammatory mediators, and hemostatic parameter expressions were observed. In addition, protease-activated receptor-1 (PAR1) activation in lungs and Raw 264.7 cells was observed after TLR3 stimulation, which was differentially modulated by PG-Lr1505. The peptidoglycan from L. rhamnosus CRL1505 is able to regulate inflammation, the procoagulant state, and PAR1 activation in mice and macrophages in the context of the activation of TLR3 signaling pathways, contributing to a beneficial modulation of inflammation-hemostasis crosstalk.Fil: Zelaya, MarĂ­a Hortensia del Rosario.     USO DE CÍTRICOS COMO SUSTRATO PARA LA CRÍA EN SEMICAUTIVERIO DE Hermetia illucens “MOSCA SOLDADO NEGRO” EN LORETO, PERÚ

    In the rearing of native fish of the Peruvian Amazon such as Colossoma macropomum, Piaractus brachypomus, Brycon amazonicus, Prochilodus nigricans and Arapaima gigas, balanced feed is used, in whose formulation fishmeal and other non‑local inputs predominate. This makes its acquisition more expensive and difficult, mainly by rural Amazonian producers who are far from the urban area. Worldwide, there are experiences of alternatives for raising fish based on the use of vegetables, fruits and insects, taking advantage of the biological diversity of the environment. Thus, the use of insect larvae constitutes a promising alternative for the aquaculture sector on a global scale. The present study shows the semi‑captivity breeding of Hermetia illucens "black soldier fly" using organic waste (citrus) collected from urban and rural areas, highlighting the use of orange bagasse and grapefruit . In this way, using the circular economy principle, live larvae and soldier fly meal were obtained to be used as an alternative to feed commercially important fish. As a final product of the rearing of the larvae, an organic compost ideal for agroforestry systems was obtained. This study shows a cheap, accessible and environmentally friendly methodology for raising black soldier fly in semi‑captivity in the region of Loreto by using citrus from the urban and rural area.En la crianza de peces nativos de la Amazonıá peruana como Colossoma macropomum, Piaractus brachypomus, Brycon amazonicus, Prochilodus nigricans y Arapaima gigas se utiliza alimento balanceado, en cuya formulación predomina la harina de pescado y otros insumos no locales. Esto, encarece y dificulta su adquisición, principalmente por los productores rurales amazónicos que se encuentran alejados del área urbana. Mundialmente, existen experiencias de alternativas para la crianza de peces en base al uso de vegetales, frutas e insectos, aprovechando la diversidad biológica del entorno. AsĂ­, el uso de larvas de insectos constituye una alternativa promisoria para el sector acuıć ola a escala global. El presente estudio muestra la crĂ­a en semicautiverio de Hermetia illucens “mosca soldado negra” utilizando residuos orgánicos (cĂ­tricos) colectados del área urbana y rural, destacando el uso de bagazo de naranjas y toronja. De esta forma utilizando, el principio de economĂ­a circular, se obtuvieron larvas vivas y harina de mosca soldado, las cuales podrán ser utilizadas para alimentar a peces de importancia comercial. Como producto final de la crianza de las larvas se obtuvo un compost orgánico ideal para sistemas agroforestales.     Hypophysation was the first known induced reproduction technique in fish farming and, it continues being applied to this day. The study aimed to evaluate the reproductive response of adult males and females of black-finned pacu Colossoma macropomum, induced with injections of paiche pituitary extract (EPP) and carp pituitary extract (CPE). We used ten females and ten males with an average body weight of 6,81 ± 1,86 kg and 6,59 ± 2,34 kg, respectively. Four females were treated with carp pituitary extract (CPE) at 6,0 mg/kg body weight, while another six received paiche pituitary extract (PPE) at 3,5 mg/kg body weight. In both cases, the dose was administered in two injections (10% at the beginning and 90% twelve hours later). Four males were induced with EPC and six with EPP at 1 mg/kg body weight administered in two 50% doses simultaneously as the females. The females induced with the EPP spawned in their entirety, while with the EPC only 75% of them. The means of total weight of spawned oocytes, number of oocytes per gram, absolute fecundity, hatching rate and the total number of larvae produced from the two hormonal treatments used in the present study did not present significant differences (P&gt;0.05). The present work demonstrated that it is possible to carry out the induced reproduction of Colossoma macropomum using paiche pituitary extract.La hipofizaciĂłn fue la primera tĂ©cnica de reproducciĂłn inducida conocida en piscicultura y sigue siendo una tĂ©cnica aplicada hasta la actualidad. El objetivo del estudio fue evaluar la respuesta reproductiva de machos y hembras adultas de gamitana Colossoma macropomum, inducidas con inyecciones de extracto de pituitaria de paiche (EPP) y extracto de pituitaria de carpa (EPC). Utilizamos diez hembras y diez machos con un peso corporal promedio de 6,81 ± 1,86 kg y 6,59 ± 2,34 kg, respectivamente. Cuatro hembras fueron tratadas con el extracto de hipĂłfisis de carpa (EPC) a 6,0 mg/kg de peso corporal, mientras que otras seis recibieron el extracto de pituitaria de paiche (EPP) a razĂłn de 3,5 mg/kg de peso corporal. En ambos casos la dosis fue administrada en dos inyecciones (10% al inicio y 90% doce horas despuĂ©s). Cuatro machos fueron inducidos con EPC y seis con EPP a razĂłn de 1 mg/kg de peso corporal administrados en dos dosis de 50%, al mismo tiempo que las hembras. Las hembras inducidas con el EPP desovaron en su totalidad, mientras que con el EPC solo el 75% de ellas. La tasa de desove, asĂ­ como las medias de peso total de ovocitos desovados, nĂșmero de ovocitos por gramo, fecundidad absoluta, tasa de eclosiĂłn y el nĂșmero total de larvas producidas de los dos tratamientos hormonales utilizados en el presente estudio no presentaron diferencias significativas (P&gt;0.05).     THE INFLUENCE OF HIV-1 SUBTYPES C, CRF31_BC AND B ON DISEASE PROGRESSION AND INITIAL VIROLOGIC RESPONSE TO HAART IN A SOUTHERN BRAZILIAN COHORT

    Background: Although most HIV-1 infections in Brazil are due to subtype B, Southern Brazil has a high prevalence of subtype C and recombinant forms, such as CRF31_BC. This study assessed the impact of viral diversity on clinical progression in a cohort of newly diagnosed HIV-positive patients. Methods: From July/2004 to December/2005, 135 HIV-infected patients were recruited. The partial pol region was subtyped by phylogeny. A generalized estimating equation (GEE) model was used to examine the relationship between viral subtype, CD4+ T cell count and viral load levels before antiretroviral therapy. Hazard ratio (Cox regression) was used to evaluate factors associated with viral suppression (viral load < 50 copies/mL at six months). Results: Main HIV-1 subtypes included B (29.4%), C (28.2%), and CRF31_BC (23.5%). Subtypes B and C showed a similar trend in CD4+ T cell decline. Comparison of non-B (C and CRF31_BC) and B subtypes revealed no significant difference in the proportion of patients with viral suppression at six months (week 24). Higher CD4+ T cell count and lower viral load were independently associated with viral suppression. Conclusion: No significant differences were found between subtypes; however, lower viral load and higher CD4+ T cell count before therapy were associated with better response

    Taking the pulse of Earth's tropical forests using networks of highly distributed plots

    Tropical forests are the most diverse and productive ecosystems on Earth. While better understanding of these forests is critical for our collective future, until quite recently efforts to measure and monitor them have been largely disconnected. Networking is essential to discover the answers to questions that transcend borders and the horizons of funding agencies. Here we show how a global community is responding to the challenges of tropical ecosystem research with diverse teams measuring forests tree-by-tree in thousands of long-term plots. We review the major scientific discoveries of this work and show how this process is changing tropical forest science. Our core approach involves linking long-term grassroots initiatives with standardized protocols and data management to generate robust scaled-up results. By connecting tropical researchers and elevating their status, our Social Research Network model recognises the key role of the data originator in scientific discovery. Conceived in 1999 with RAINFOR (South America), our permanent plot networks have been adapted to Africa (AfriTRON) and Southeast Asia (T-FORCES) and widely emulated worldwide. Now these multiple initiatives are integrated via ForestPlots.net cyber-infrastructure, linking colleagues from 54 countries across 24 plot networks. Collectively these are transforming understanding of tropical forests and their biospheric role. Together we have discovered how, where and why forest carbon and biodiversity are responding to climate change, and how they feedback on it. This long-term pan-tropical collaboration has revealed a large long-term carbon sink and its trends, as well as making clear which drivers are most important, which forest processes are affected, where they are changing, what the lags are, and the likely future responses of tropical forests as the climate continues to change. By leveraging a remarkably old technology, plot networks are sparking a very modern revolution in tropical forest science.     MORTALIDAD DE BANDA NEGRA Myloplus Schomburgkii DEBIDO A LA POBRE CALIDAD DEL AGUA ASOCIADA CON LA SOBREPOBLACIÓN DE LENTEJAS DE AGUA EN UN ESTANQUE DE CRIANZA EN LA AMAZONÍA PERUANA

    Ponds covered with duckweeds release nutrients rapidly, causing problems of eutrophication of the water which can reduce water clarity and quality, creating a hypoxic or anoxic ‘dead zone' lacking sufficient oxygen to support most organisms. In the present study, the sudden mortality of black-band myleus Myloplus schomburgkii Jardine, 1841 drove the investigate of factors that caused the death of the specimens registered in a fish pond in the Peruvian Amazon. After the tallying of dead fish, it was revealed that sixty individuals (75% of the total population) of M. schomburgkii had died. The taxonomic identification of the aquatic plant revealed the presence of “duckweeds” locally named in Peru as “lenteja de agua” Lemna minor distributed throughout the pond. Physical and chemical water parameters revealed low levels of oxygen, acid water, and high levels of nitrate and phosphate. Since duckweeds cannot be completely eliminated from waters that are conducive to its growth and require periodic harvesting to prevent matting, proper management strategies therefore become critical to guarantee good quality of water in the pond and avoid black-band myleus deaths due to imbalances in physical and chemical parameters.Los estanques cubiertos de lentejas de agua liberan nutrientes de forma rĂĄpida causando problemas de eutrofizaciĂłn del agua que pueden reducir la transparencia y perjudicar su calidad, creando una "zona mortal" hipĂłxica o anĂłxica que carece de oxĂ­geno suficiente para mantener a la mayorĂ­a de los organismos. En el presente estudio se observĂł la mortalidad del pez “banda negra” Myloplus schomburgkii Jardine, 1841, lo que motivĂł a investigar los factores que causaron la muerte de los ejemplares registrados en un estanque de peces en la AmazonĂ­a peruana. Tras el recuento de los peces muertos, se revelĂł que sesenta individuos (75% de la poblaciĂłn total) de M. schomburgkii murieron. La identificaciĂłn taxonĂłmica revelĂł la presencia de la planta acuĂĄtica Lemna minor denominadas localmente en PerĂș como "lentejas de agua" distribuida por todo el estanque. Los parĂĄmetros fĂ­sicos y quĂ­micos del agua revelaron bajos niveles de oxĂ­geno, agua ĂĄcida y altos niveles de nitrato y fosfato. Dado que la lenteja de agua no puede eliminarse por completo del agua, que favorece su desarrollo, hay que cosecharlas periĂłdicamente para evitar que aumenten desmedidamente. Las estrategias de manejo son fundamentales para garantizar la buena calidad del agua del estanque y evitar mortalidades debido a desequilibrio de parĂĄmetros fĂ­sicos y quĂ­micos del agua
