Dry period plane of energy: Effects on glucose tolerance in transition dairy cows
Overfeeding energy in the dry period can affect glucose metabolism and the energy balance of transition dairy cows with potential detrimental effects on the ability to successfully adapt to early lactation. The objectives of this study were to investigate the effect of different dry cow feeding strategies on glucose tolerance and on resting concentrations of blood glucose, glucagon, insulin, nonesterified fatty acids (NEFA), and β-hydroxybutyrate (BHB) in the peripartum period. Cows entering second or greater lactation were enrolled at dry-off (57 d before expected parturition) into 1 of 3 treatment groups following a randomized block design: cows that received a total mixed ration (TMR) formulated to meet but not exceed energy requirements during the dry period (n = 28, controlled energy); cows that received a TMR supplying approximately 150% of energy requirements during the dry period (n = 28, high energy); and cows that were fed the same diet as the controlled energy group for the first 28 d, after which the TMR was formulated to supply approximately 125% of energy requirements until calving (n = 28, intermediate energy). Intravenous glucose tolerance tests (IVGTT) with rapid administration of 0.25 g of glucose/kg of body weight were performed 28 and 10 d before expected parturition, as well as at 4 and 21 d after calving. Area under the curve for insulin and glucose, maximal concentration and time to half-maximal concentration of insulin and glucose, and clearance rates were calculated. Insulin resistance (IR) indices were calculated from baseline samples obtained during IVGTT and Spearman rank correlations determined between IVGTT parameters and IR indices. Treatment did not affect IVGTT parameters at any of the 4 time points. Correlation between IR indices and IVGTT parameters was generally poor. Overfeeding cows energy in excess of predicted requirements by approximately 50% during the entire dry period resulted in decreased postpartum basal plasma glucose and insulin, as well as increased glucagon, BHB, and NEFA concentrations after calving compared with cows fed a controlled energy diet during the dry period. In conclusion, overfeeding energy during the entire dry period or close-up period alone did not affect glucose tolerance as assessed by IVGTT but energy uptake during the dry period was associated with changes in peripartal resting concentrations of glucose, as well as postpartum insulin, glucagon, NEFA, and BHB concentrations
Intraspecific comparative genomics of isolates of the Norway spruce pathogen (Heterobasidion parviporum) and identification of its potential virulence factors
Background: Heterobasidion parviporum is an economically most important fungal forest pathogen in northern Europe, causing root and butt rot disease of Norway spruce (Picea abies (L.) Karst.). The mechanisms underlying the pathogenesis and virulence of this species remain elusive. No reference genome to facilitate functional analysis is available for this species. Results: To better understand the virulence factor at both phenotypic and genomic level, we characterized 15 H. parviporum isolates originating from different locations across Finland for virulence, vegetative growth, sporulation and saprotrophic wood decay. Wood decay capability and latitude of fungal origins exerted interactive effects on their virulence and appeared important for H. parviporum virulence. We sequenced the most virulent isolate, the first full genome sequences of H. parviporum as a reference genome, and re-sequenced the remaining 14 H. parviporum isolates. Genome-wide alignments and intrinsic polymorphism analysis showed that these isolates exhibited overall high genomic similarity with an average of at least 96% nucleotide identity when compared to the reference, yet had remarkable intra-specific level of polymorphism with a bias for CpG to TpG mutations. Reads mapping coverage analysis enabled the classification of all predicted genes into five groups and uncovered two genomic regions exclusively present in the reference with putative contribution to its higher virulence. Genes enriched for copy number variations (deletions and duplications) and nucleotide polymorphism were involved in oxidation-reduction processes and encoding domains relevant to transcription factors. Some secreted protein coding genes based on the genome-wide selection pressure, or the presence of variants were proposed as potential virulence candidates. Conclusion: Our study reported on the first reference genome sequence for this Norway spruce pathogen (H. parviporum). Comparative genomics analysis gave insight into the overall genomic variation among this fungal species and also facilitated the identification of several secreted protein coding genes as putative virulence factors for the further functional analysis. Separable and non-separable multi-field inflation and large non-Gaussianity
In this paper we provide a general framework based on formalism to
estimate the cosmological observables pertaining to the cosmic microwave
background radiation for non-separable potentials, and for generic \emph{end of
inflation} boundary conditions. We provide analytical and numerical solutions
to the relevant observables by decomposing the cosmological perturbations along
the curvature and the isocurvature directions, \emph{instead of adiabatic and
entropy directions}. We then study under what conditions large bi-spectrum and
tri-spectrum can be generated through phase transition which ends inflation. In
an illustrative example, we show that large and
can be obtained for the case of separable and

A multivariate dataset on water–energy–food nexus:Multi-actor governance for social justice
The article presents and validates an extensive multivariate dataset that offers insights into water–energy–food (WEF) nexus governance for social justice at the intrahousehold, household, and community levels. The lack of insights in the WEF nexus debate that take social justice and governance into account is what spurred the data collection. The initial process involved scoping the originally selected investigated sites and their suitability. Once the research areas were identified, the data were collected from 1184 households in the Matatiele, Magareng, and Greater Taung Local Municipalities in South Africa, using a semi-structured questionnaire and KoboCollect software. The freely available software was installed on Android Tablets which were used by the enumerators. The questionnaires were initially piloted in Matatiele Local Municipality, testing for internal validity and skip patterns, as well as time to complete the questionnaire. The reliability of the Likert responses from the questionnaire was evaluated using Cronbachʼs alpha. The questionnaire was then refined for data collection and utilized a total of twenty-two (22) locally trained enumerators who were employed at the investigated sites. These enumerators were trained in administering the questionnaire and the use of the KoboCollect software used in data collection. The enumerators also received training on how to conduct the survey ethically, including informed permission, confidentiality, and the option to withdraw from the interview. The design of the data collection process was a cross-sectional survey that was conducted between 6 June and 4 August 2022, using purposive sampling. At the end of each data collection day, the enumerators uploaded their collected data into the KoboTool cloud, which allowed the lead in the survey to assess the data and effect any correctional measures on the questionnaire if the need arose. The enumerators also used a WhatsApp chat group to communicate real time opportunities and challenges in the questionnaire, which allowed the lead in the survey to constantly update the questionnaire. The multivariate questionnaire was divided into sections of socioeconomic and demographic characteristics, community-level governance, decision-making, food, energy, and water security, social justice, legal knowledge, and rights to utilize these resources. The dataset will be of significance to multi-disciplinary researchers focusing on WEF security, governance, and social justice in Southern Africa. Furthermore, environmental and sustainability practitioners can find valuable insights from the provided data. The employed methodology is replicable and adaptable, enabling real-time monitoring of social justice and governance in the context of food, energy, and water security. The real time monitoring of governance and social justice in water, energy and food allows for the possibility of continual data collection and updating, and if a longitudinal design is adopted, it can be used for impact inquiry of any interventions or policies.</p
