    This article introduces a methodology to evidence the current attractiveness level of Italian high-speed railway stations in a GIS environment, involving station services and fow parameters. The model has been elevant to detect stations with lower attractive capacity, and afterward, to implement the station attractivity, the work proposed employing a coworking spaces strategy as a service in station. Coworking spaces produce enefts both for the traveler and the transport company. These places became part of the services ofered within railway stations since they are fow providers able to change appearance and idea of experience at station. In France, a coworking strategy has been created from the collaboration of Regus, leader company in coworking spaces supply, and the French railway group (SNCF). The Italian railway company (Ferrovie dello Stato) does not consider the attractiveness potential of coworking in the management of station resources; coworking spaces in Italy are placed outside stations. Accordingly, Torino Porta Susa station has been identifed as one of the stations with low attractivity capacity from the methodology implemented, and it has been chosen as the case study to implement the coworking strategy. The choice of Torino Porta Susa is accurate also for showing the value of associating coworking as urban policies support.     Decellularized Human Dermal Matrix as a Biological Scaffold for Cardiac Repair and Regeneration.

    The complex and highly organized environment in which cells reside consists primarily of the extracellular matrix (ECM) that delivers biological signals and physical stimuli to resident cells. In the native myocardium, the ECM contributes to both heart compliance and cardiomyocyte maturation and function. Thus, myocardium regeneration cannot be accomplished if cardiac ECM is not restored. We hypothesize that decellularized human skin might make an easily accessible and viable alternate biological scaffold for cardiac tissue engineering (CTE). To test our hypothesis, we decellularized specimens of both human skin and human myocardium and analyzed and compared their composition by histological methods and quantitative assays. Decellularized dermal matrix was then cut into 600-mm-thick sections and either tested by uniaxial tensile stretching to characterize its mechanical behavior or used as three-dimensional scaffold to assess its capability to support regeneration by resident cardiac progenitor cells (hCPCs) in vitro. Histological and quantitative analyses of the dermal matrix provided evidence of both effective decellularization with preserved tissue architecture and retention of ECM proteins and growth factors typical of cardiac matrix. Further, the elastic modulus of the dermal matrix resulted comparable with that reported in literature for the human myocardium and, when tested in vitro, dermal matrix resulted a comfortable and protective substrate promoting and supporting hCPC engraftment, survival and cardiomyogenic potential. Our study provides compelling evidence that dermal matrix holds promise as a fully autologous and cost-effective biological scaffold for CTE

    Management of Metastatic Endometrial Cancer: Physicians' Choices Beyond the First Line. A MITO Survey

    BACKGROUND: Endometrial cancer (EC) therapeutic and diagnostic approaches have been changed by the development of a new prognostic molecular classification, the introduction of dostarlimab in microsatellite instability (MSI) high pre-treated advanced EC patients with further expected innovation deriving from lenvatinib plus pembrolizumab regardless MSI status. How this is and will be translated and embedded in the clinical setting in Italy is not known; this is why we developed Multicentre Italian Trials in Ovarian cancer and gynaecologic malignancies (MITO) survey on the current practice and expected future changes in EC. METHODS: We designed a self-administered, multiple-choice online questionnaire available only for MITO members for one month, starting in April 2021. RESULTS: 75.6% of the respondents were oncologists with a specific focus on gynaecologic malignancies and 73.3% of the respondents declared the availability of clinical trials in second line treatment for advanced EC. The therapeutic algorithm in second line was heterogeneous, being the most frequent choice administering anthracyclines followed by endocrine therapy or enrolling in clinical trials. While more than half of the clinicians declared that they performed the molecular classification, only six/45 respondents (13.3%) ran all the tests needed for it. On the other hand, 80% of them declared regular assessment of MSI status with IHC as recommended. The therapeutic approach in MSI high advanced EC patients has changed since dostarlimab approval. Indeed the most frequent choice in second line has been chemotherapy (53.3%) before its availability, while dostarlimab has been preferred in more than three-fourths of the cases (75.6%) after its approval. As for MSS patients, 77.8% of clinicians would choose lenvatinib plus pembrolizumab for them in second line once approved. CONCLUSIONS: Despite the selected sample of respondents from Italian MITO centres showing good knowledge of diagnostic and therapeutic innovations in EC, these are not fully implemented in everyday clinics, except for MSI status assessment

    Ecosystem-Based Adaptation Approach and Adaptation Planning Support Tools: Potential Implementation for the Urban Context

    Climate Change (CC) is having a progressively negative impact on natural resources management. It, indeed, worsens existing environmental challenges, placing socio-ecological systems in a situation of new risks. This process invites to reflect about the Ecosystem-based Adaptation (EbA) approach, which combines concepts of sustainable land use and conservation in one integrated adaptation strategy. Challenges related to definition and integration of the EbA process into urban context exist, in particular concerning the necessity of a systemic methodology deployment in urban planning instruments and adaptation processes. As a possible entry point, this work aims to identify the adaptation planning support tools for building an EbA process at city scale, defining a possible methodology for tracing a shortlist of tools supporting EbA principles. The outcomes have demonstrated that tools, albeit not having exhaustive capacities, provide important directions for theoretical investigation and consequent actions. There is a need for tools explicitly addressed to the EbA approach, not still homogeneously supported in the various stages of the adaptation framework. Hence, they offer useful insights to establish a comprehensive adaptation process able to integrate EbA in urban planning practices. To date, this represents one of the triggering points for the implementation of an operational Nature-based Solution (NbS) framework

    Red Wine Consumption and Cardiovascular Health

    Wine is a popular alcoholic beverage that has been consumed for hundreds of years. Benefits from moderate alcohol consumption have been widely supported by the scientific literature and, in this line, red wine intake has been related to a lesser risk for coronary heart disease (CHD). Experimental studies and meta-analyses have mainly attributed this outcome to the presence in red wine of a great variety of polyphenolic compounds such as resveratrol, catechin, epicatechin, quercetin, and anthocyanin. Resveratrol is considered the most effective wine compound with respect to the prevention of CHD because of its antioxidant properties. The mechanisms responsible for its putative cardioprotective effects would include changes in lipid profiles, reduction of insulin resistance, and decrease in oxidative stress of low-density lipoprotein cholesterol (LDL-C). The aim of this review is to summarize the accumulated evidence correlating moderate red wine consumption with prevention of CHD by focusing on the different mechanisms underlying this relationship. Furthermore, the chemistry of wine as well as chemical factors that influence the composition of the bioactive components of red wine are also discussed

    Target Analysis and Retrospective Screening of Multiple Mycotoxins in Pet Food Using UHPLC-Q-Orbitrap HRMS

    A comprehensive strategy combining a quantitative method for 28 mycotoxins and a post-target screening for other 245 fungal and bacterial metabolites in dry pet food samples were developed using an acetonitrile-based extraction and an ultrahigh-performance liquid chromatography coupled to high-resolution mass spectrometry (UHPLC-Q-Orbitrap HRMS) method. The proposed method showed satisfactory validation results according to Commission Decision 2002/657/EC. Average recoveries from 72 to 108% were obtained for all studied mycotoxins, and the intra-/inter-day precision were below 9 and 14%, respectively. Results showed mycotoxin contamination in 99% of pet food samples (n = 89) at concentrations of up to hundreds µg/kg, with emerging Fusarium mycotoxins being the most commonly detected mycotoxins. All positive samples showed co-occurrence of mycotoxins with the simultaneous presence of up to 16 analytes per sample. In the retrospective screening, up to 54 fungal metabolites were tentatively identified being cyclopiazonic acid, paspalitrem A, fusaric acid, and macrosporin, the most commonly detected analytes

    Mapping and Assessing Soil Sealing in Padua Municipality through Biotope Area Factor Index

    Soil sealing is a worldwide phenomenon of covering of natural or seminatural soil with impervious surfaces, such as built-up or paved surfaces. It is widely recognized as a major environmental issue which drives landscape fragmentation and ecosystem services degradation and loss. Italy is one of European countries with the highest extent of soil sealing. The most affected area is northern Italy, especially the Po Valley with more than 12% of sealed surfaces. According to official data, Veneto Region and the city of Padua are seriously affected by this phenomenon. The Biotope Area Factor is a consolidated ecological urban index for mapping soil sealing, adopted in different European cities to support urban planning; it expresses the ratio of the ecologically effective surface area in relation to the total land area according to land cover classes. The general aim of this study is to map and to assess soil sealing in the whole municipal territory of Padua using the Biotope Area Factor (BAF) index. We tested and adopted a digital land cover map together with aerial images to perform a BAF analysis on the whole municipal territory of Padua. By using sample areas previously analyzed, we validated our source data by a double spatial validation process; therefore, soil sealing analysis was scaled-up to the municipality territory. Results show that in the city of Padua, the average BAF index value is 0.6; totally permeable surfaces (BAF = 1) cover 59.5%, whereas totally “sealed” surfaces (BAF = 0) are 40.3% of the municipal territory (93 km2). Most of the sealed soil is located in the east sector and in the historical core of the city, with BAF values ranging from 0 to 0.2. A particularly critical area is identified within the new industrial area of the city, which is strongly affected by soil sealing. BAF maps are useful tools to identify critical areas by geovisualizing surface permeability at a very detailed scale and by enabling further analyses for hydrogeological risk assessment and urban climate regulation. Moreover, the use of BAF maps at urban scale today represent an important tool for urban management, especially for policy makers who are planning mitigation and compensation measures to control soil sealing

    Cognitive Reserve in Early Manifest Huntington Disease Patients: Leisure Time Is Associated with Lower Cognitive and Functional Impairment

    We focused on Cognitive Reserve (CR) in patients with early Huntington Disease (HD) and investigated whether clinical outcomes might be influenced by lifetime intellectual enrichment over time. CR was evaluated by means of the Cognitive Reserve Index questionnaire (CRIq), an internationally validated scale which includes three sections: education, working activity, and leisure time. The clinical HD variables were quantified at three different time points (baseline-t0, 1 year follow up-t1 and 2 years follow up-t2) as per the Unified Huntington’s Disease Rating Scale (UHDRS), an internationally standardized and validated scale including motor, cognitive, functional and behavioral assays. Our sample consisted of 75 early manifest patients, withclinical stage scored according to the Total Functional Capacity (TFC) scale. Our correlational analysis highlighted a significant inverse association between CRIq leisure time (CRIq_LA) and longitudinal functional impairment (namely, the differential TFC score between t2 and t0 or ΔTFC) (p p p < 0.05). Our results suggest that higher is the CRIq_LA, milder is the progression of HD in terms of functional, multidimensional and cognitive outcome
