Progesterone Administration Modulates Cortical TLR4/NF-κB Signaling Pathway after Subarachnoid Hemorrhage in Male Rats
Our previous study concerning brain trauma has shown that progesterone could regulate toll-like receptor 4 (TLR4) and nuclear factor-kappa B (NF-κB) signaling pathway in the brain, which also has been proved to play important roles in early brain injury (EBI) after subarachnoid hemorrhage (SAH). The aim of the current study was to investigate whether progesterone administration modulated TLR4/NF-κB pathway signaling pathway in the brain at the early stage of SAH. All SAH animals were subjected to injection of 0.3 ml fresh arterial, non-heparinized blood into prechiasmatic cistern in 20 seconds. Male rats were given 0 or 16 mg/kg injections of progesterone at post-SAH hours 1, 6, and 24. Brain samples were extracted at 48 h after SAH. As a result, SAH could induce a strong up-regulation of TLR4, NF-κB, pro-inflammatory cytokines, MCP-1, and ICAM-1 in the cortex. Administration of progesterone following SAH could down-regulate the cortical levels of these agents related to TLR4/NF-κB signaling pathway. Post-SAH progesterone treatment significantly ameliorated the EBI, such as the clinical behavior scale, brain edema, and blood-brain barrier (BBB) impairment. It was concluded that post-SAH progesterone administration may attenuate TLR4/NF-κB signaling pathway in the rat brain following SAH
Transcriptome sequencing and phylogenomic resolution within Spalacidae (Rodentia)
BACKGROUND: Subterranean mammals have been of great interest for evolutionary biologists because of their highly specialized traits for the life underground. Owing to the convergence of morphological traits and the incongruence of molecular evidence, the phylogenetic relationships among three subfamilies Myospalacinae (zokors), Spalacinae (blind mole rats) and Rhizomyinae (bamboo rats) within the family Spalacidae remain unresolved. Here, we performed de novo transcriptome sequencing of four RNA-seq libraries prepared from brain and liver tissues of a plateau zokor (Eospalax baileyi) and a hoary bamboo rat (Rhizomys pruinosus), and analyzed the transcriptome sequences alongside a published transcriptome of the Middle East blind mole rat (Spalax galili). We characterize the transcriptome assemblies of the two spalacids, and recover the phylogeny of the three subfamilies using a phylogenomic approach. RESULTS: Approximately 50.3 million clean reads from the zokor and 140.8 million clean reads from the bamboo ratwere generated by Illumina paired-end RNA-seq technology. All clean reads were assembled into 138,872 (the zokor) and 157,167 (the bamboo rat) unigenes, which were annotated by the public databases: the Swiss-prot, Trembl, NCBI non-redundant protein (NR), NCBI nucleotide sequence (NT), Gene Ontology (GO), Cluster of Orthologous Groups (COG), and Kyoto Encyclopedia of Genes and Genomes (KEGG). A total of 5,116 nuclear orthologous genes were identified in the three spalacids and mouse, which was used as an outgroup. Phylogenetic analysis revealed a sister group relationship between the zokor and the bamboo rat, which is supported by the majority of gene trees inferred from individual orthologous genes, suggesting subfamily Myospalacinae is more closely related to subfamily Rhizomyinae. The same topology was recovered from concatenated sequences of 5,116 nuclear genes, fourfold degenerate sites of the 5,116 nuclear genes and concatenated sequences of 13 protein coding mitochondrial genes. CONCLUSIONS: This is the first report of transcriptome sequencing in zokors and bamboo rats, representing a valuable resource for future studies of comparative genomics in subterranean mammals. Phylogenomic analysis provides a conclusive resolution of interrelationships of the three subfamilies within the family Spalacidae, and highlights the power of phylogenomic approach to dissect the evolutionary history of rapid radiations in the tree of life
The role of quality of care and attitude towards disability in the relationship between severity of disability and quality of life: findings from a cross-sectional survey among people with physical disability in China
Background: People with physical disability (PWPD) is the largest subgroup of people with disability (PWD) in China, but few studies have been conducted among this vulnerable population. The objective of this study was to investigate the level of quality of life (QoL), self-perceived quality of care and support (QOCS), severity of disability and personal attitude towards disability among people with physical disability in China, as well as to identify how QoL can be affected by severity of disability through QOCS and personal attitude towards disability among PWPD. Methods: A cross-sectional study was conducted among 1,853 PWPD in Guangzhou, China. Data were collected on participants’ QoL, QOCS, personal attitude towards disability and severity of disability. Structural equation modeling was used to examine the effects of the other variables on QoL. Results: Even with a mild disability (mean score:1.72), relatively low levels of QoL (mean score: 2.65- 3.22) and QOCS (mean score: 2.95 to 3.28), as well as unfavorable personal attitude towards disability (mean score: 2.75 to 3.36) were identified among PWPD. According to SEM, we found that the influence of severity of physical disability on QoL is not only exerted directly, but is also indirectly through QOCS and their personal attitudes towards disability, with QOCS playing a more important mediating role than PWPD’s attitudes towards their own disability. Conclusions: Unfavorable health status was identified among PWPD in China. Focusing on improvement of assistance and care services has the potential to substantially improve PWPD’s QoL. Further research should focus on understanding the needs and their current state of health care of PWPD in China thus being able to develop better interventions for them
Isospin Effect on the Process of Multifragmentation and Dissipation at Intermediate Energy Heavy Ion Collisions
In the simulation of intermediate energy heavy ion collisions by using the
isospin dependent quantum molecular dynamics, the isospin effect on the process
of multifragmentation and dissipation has been studied. It is found that the
multiplicity of intermediate mass fragments for the neutron-poor
colliding system is always larger than that for the neutron-rich system, while
the quadrupole of single particle momentum distribution for the
neutron-poor colliding system is smaller than that of the neutron-rich system
for all projectile-target combinations studied at the beam energies from about
50MeV/nucleon to 150MeV/nucleon. Since depends strongly on isospin
dependence of in-medium nucleon-nucleon cross section and weakly on symmetry
potential at the above beam energies, it may serve as a good probe to extract
the information on the in-medium nucleon-nucleon cross section. The correlation
between the multiplicity of intermediate mass fragments and the total
numer of charged particles has the behavior similar to , which
can be used as a complementary probe to the in-medium nucleon-nucleon cross

Cancer-induced bone pain sequentially activates the ERK/MAPK pathway in different cell types in the rat spinal cord
<p>Abstract</p> <p>Background</p> <p>Previous studies have demonstrates that, after nerve injury, extracellular signal-regulated protein kinase (ERK) activation in the spinal cord-initially in neurons, then microglia, and finally astrocytes. In addition, phosphorylation of ERK (p-ERK) contributes to nociceptive responses following inflammation and/or nerve injury. However, the role of spinal cells and the ERK/MAPK pathway in cancer-induced bone pain (CIBP) remains poorly understood. The present study analyzed activation of spinal cells and the ERK/MAPK pathway in a rat model of bone cancer pain.</p> <p>Results</p> <p>A Sprague Dawley rat model of bone cancer pain was established and the model was evaluated by a series of tests. Moreover, fluorocitrate (reversible glial metabolic inhibitor) and U0126 (a MEK inhibitor) was administered intrathecally. Western blots and double immunofluorescence were used to detect the expression and location of phosphorylation of ERK (p-ERK). Our studies on pain behavior show that the time between day 6 and day 18 is a reasonable period ("time window" as the remaining stages) to investigate bone cancer pain mechanisms and to research analgesic drugs. Double-labeling immunofluorescence revealed that p-ERK was sequentially expressed in neurons, microglia, and astrocytes in the L4-5 superficial spinal cord following inoculation of Walker 256 cells. Phosphorylation of ERK (p-ERK) and the transcription factor cAMP response element-binding protein (p-CREB) increased in the spinal cord of CIBP rats, which was attenuated by intrathecal injection of fluorocitrate or U0126.</p> <p>Conclusions</p> <p>The ERK inhibitors could have a useful role in CIBP management, because the same target is expressed in various cells at different times.</p

The interactome was further validated by the expression
pattern analysis in nodules, gene ontology term semantic
similarity, co-expression analysis, and luciferase complementation image assay. In the G. max−B. diazoefficiens
interactome, bacterial proteins are mainly ion channel and
transporters of carbohydrates and cations, while G. max
proteins are mainly involved in the processes of metabolism,
signal transduction, and transport. We also identified the top 10 highly interacting proteins (hubs) for each species. Kyoto Encyclopedia of Genes and Genomes pathway analysis for each hub showed that a pair of 14-3-3 proteins (SGF14g and SGF14k) and 5 heat shock proteins in G. max are possibly involved in symbiosis, and 10 hubs in

SDSS J013127.34032100.1: A newly discovered radio-loud quasar at with extremely high luminosity
Only very few z>5 quasars discovered to date are radio-loud, with a
radio-to-optical flux ratio (radio-loudness parameter) higher than 10. Here we
report the discovery of an optically luminous radio-loud quasar, SDSS
J013127.34-032100.1 (J0131-0321 in short), at z=5.18+-0.01 using the Lijiang
2.4m and Magellan telescopes. J0131-0321 has a spectral energy distribution
consistent with that of radio-loud quasars. With an i-band magnitude of 18.47
and radio flux density of 33 mJy, its radio-loudness parameter is ~100. The
optical and near-infrared spectra taken by Magellan enable us to estimate its
bolometric luminosity to be L_bol ~ 1.1E48 erg/s, approximately 4.5 times
greater than that of the most distant quasar known to date. The black hole mass
of J0131-0321 is estimated to be 2.7E9 solar masses, with an uncertainty up to
0.4 dex. Detailed physical properties of this high-redshift, radio-loud,
potentially super-Eddington quasar can be probed in the future with more
dedicated and intensive follow-up observations using multi-wavelength

Complete sequence and organization of Antheraea pernyi nucleopolyhedrovirus, a dr-rich baculovirus
<p>Abstract</p> <p>Background</p> <p>The completion and reporting of baculovirus genomes is extremely important as it advances our understanding of gene function and evolution. Due to the large number of viral genomes now sequenced it is very important that authors present significantly detailed analyses to advance the understanding of the viral genomes. However, there is no report of the <it>Antheraea pernyi </it>nucleopolyhedrovirus (AnpeNPV) genome.</p> <p>Results</p> <p>The genome of AnpeNPV, which infects Chinese tussah silkworm (<it>Antheraea pernyi</it>), was sequenced and analyzed. The genome was 126,629 bp in size. The G+C content of the genome, 53.4%, was higher than that of most of the sequenced baculoviruses. 147 open reading frames (ORFs) that putatively encode proteins of 50 or more amino acid residues with minimal overlap were determined. Of the 147 ORFs, 143 appeared to be homologous to other baculovirus genes, and 4 were unique to AnpeNPV. Furthermore, there are still 29 and 33 conserved genes present in all baculoviruses and all lepidopteran baculoviruses respectively. In addition, the total number of genes common to all lepidopteran NPVs is sill 74, however the 74 genes are somewhat different from the 74 genes identified before because of some new sequenced NPVs. Only 6 genes were found exclusively in all lepidopteran NPVs and 12 genes were found exclusively in all Group I NPVs. AnpeNPV encodes <it>v-trex</it>(Anpe115, a 3' to 5' repair exonuclease), which was observed only in CfMNPV and CfDEFNPV in Group I NPVs. This gene potentially originated by horizontal gene transfer from an ancestral host. In addition, AnpeNPV encodes two <it>conotoxin</it>-like gene homologues (<it>ctls</it>), <it>ctl1 </it>and <it>ctl2</it>, which were observed only in HycuNPV, OpMNPV and LdMNPV. Unlike other baculoviruses, only 3 typical homologous regions (<it>hr</it>s) were identified containing 2~9 repeats of a 30 bp-long palindromic core. However, 24 perfect or imperfect direct repeats (<it>dr</it>s) with a high degree of AT content were found within the intergenic spacer regions that may function as non-<it>hr</it>, <it>ori</it>-like regions found in GrleGV, CpGV and AdorGV. 9 <it>dr</it>s were also found in intragenic spacer regions of AnpeNPV.</p> <p>Conclusion</p> <p>AnpeNPV belongs to Group I NPVs and is most similar to HycuNPV, EppoNPV, OpMNPV and CfMNPV based on gene content, genome arrangement, and amino acid identity. In addition, analysis of genes that flank <it>hr</it>s supported the argument that these regions are involved in the transfer of sequences between the virus and host.</p
