    Comparative Synthesis: Learning Near-Optimal Network Designs by Query

    When managing wide-area networks, network architects must decide how to balance multiple conflicting metrics, and ensure fair allocations to competing traffic while prioritizing critical traffic. The state of practice poses challenges since architects must precisely encode their intent into formal optimization models using abstract notions such as utility functions, and ad-hoc manually tuned knobs. In this paper, we present the first effort to synthesize optimal network designs with indeterminate objectives using an interactive program-synthesis-based approach. We make three contributions. First, we present comparative synthesis, an interactive synthesis framework which produces near-optimal programs (network designs) through two kinds of queries (Propose and Compare), without an objective explicitly given. Second, we develop the first learning algorithm for comparative synthesis in which a voting-guided learner picks the most informative query in each iteration. We present theoretical analysis of the convergence rate of the algorithm. Third, we implemented Net10Q, a system based on our approach, and demonstrate its effectiveness on four real-world network case studies using black-box oracles and simulation experiments, as well as a pilot user study comprising network researchers and practitioners. Both theoretical and experimental results show the promise of our approach

    CCL21/CCR7 Promotes G2/M Phase Progression via the ERK Pathway in Human Non-Small Cell Lung Cancer Cells

    C-C chemokine receptor 7 (CCR7) contributes to the survival of certain cancer cell lines, but its role in the proliferation of human non-small cell lung cancer (NSCLC) cells remains vague. Proliferation assays performed on A549 and H460 NSCLC cells using Cell Counting Kit-8 indicated that activation of CCR7 by its specific ligand, exogenous chemokine ligand 21 (CCL21), was associated with a significant linear increase in cell proliferation with duration of exposure to CCL21. The CCL21/CCR7 interaction significantly increased the fraction of cells in the G2/M phase of the cell cycle as measured by flow cytometry. In contrast, CCL21/CCR7 had no significant influence on the G0/G1 and S phases. Western blot and real-time PCR indicated that CCL21/CCR7 significantly upregulated expression of cyclin A, cyclin B1, and cyclin-dependent kinase 1 (CDK1), which are related to the G2/M phase transition. The expression of cyclin D1 and cyclin E, which are related to the G0/G1 and G1/S transitions, was not altered. The CCL21/CCR7 interaction significantly enhanced phosphorylation of extracellular signal-regulated kinase (P-ERK) but not Akt, as measured by Western blot. LY294002, a selective inhibitor of PI3K that prevents activation of the downstream Akt, did not weaken the effect of CCL21/CCR7 on P-ERK. Coimmunoprecipitation further confirmed that there was an interaction between P-ERK and cyclin A, cyclin B1, or CDK1, particularly in the presence of CCL21. CCR7 small interfering RNA or PD98059, a selective inhibitor of MEK that disrupts the activation of downstream ERK, significantly abolished the effects of exogenous CCL21. These results suggest that CCL21/CCR7 contributes to the time-dependent proliferation of human NSCLC cells by upregulating cyclin A, cyclin B1, and CDK1 potentially via the ERK pathway

    Overexpression of CARMA3 in Non-Small-Cell Lung Cancer Is Linked for Tumor Progression

    We aimed to investigate the clinical significance of the expression of novel scaffold protein CARMA3 in non-small-cell lung cancer (NSCLC) and the biological function of CARMA3 in NSCLC cell lines. We observed moderate to high CARMA3 staining in 68.8% of 141 NSCLC specimens compared to corresponding normal tissues. The overexpression of CARMA3 was significantly correlated with TNM stage (P = 0.022) and tumor status (P = 0.013). CARMA3 upregulation also correlated with a shorter survival rate of patients of nodal status N0 (P = 0.042)as well as the expression of epidermal growth factor receptor (EGFR) (P = 0.009). In EGFR mutation positive cases, CARMA3 expression was much higher (87.5%) compared to non-mutation cases (66.1%). In addition, we observed that knockdown of CARMA3 inhibits tumor cell proliferation and invasion, and induces cell cycle arrest at the boundary between the G1 and S phase. We further demonstrated a direct link between CARMA3 and NF-κB activation. The change of biological behavior in CARMA3 knockdown cells may be NF-κB-related. Our findings demonstrated, for the first time, that CARMA3 was overexpressed in NSCLC and correlated with lung cancer progression, EGFR expression, and EGFR mutation. CARMA3 could serve as a potential companion drug target, along with NF-kB and EGFR in EGFR-mutant lung cancers

    Cellular function of osteocytes in normal and αklotho-deficient mice

    During the last decade, osteocyte-derived factors i.e., sclerostin, dentin matrix protein-1, fibroblast growth factor 23 (FGF23) that reduces serum phosphate concentration by mediating FGF receptor 1c/αklotho in the kidney, have been highlighted for osteocytes’ fine-turned regulation on bone remodeling and phosphate homeostasis. Osteocytes are interconnected through gap junctions between their cytoplasmic processes, and thereby, build upon the functional syncytia, referred to as the osteocytic lacunar-canalicular system (OLCS). Osteocytes appear to communicate surrounding osteocytes and osteoblasts by means of two possible pathways of molecular transport throughout the OLCS : One is a passageway of their cytoplasmic processes, and the other is a pericellular space in the osteocytic canaliculi. The regularly-oriented OLCS in mature compact bone appears to efficiently serve for molecular transport, mechanosensing and targeted bone remodeling that would erase microdamages in bone. In a disrupted signaling state of FGF23/αklotho, serum concentration of phosphate would be markedly-elevated. Despite highly-elevated serum phosphate, αklotho -deficient mice revealed defective mineralization in bone matrix. OLCS in αklotho -deficient mice were irregularly-distributed and the connectivity of cytoplasmic processes of osteocytes was very poor, so that osteocytes did not seem to form functional syncytia. Therefore, osteocytes’function cooperated with other bone cells, rather than serum concentration of calcium/phosphate, and this seems to play a central role in maintaining bone mineralization. In this review, the biological function of the regularly-arranged OLCS in a normal state will be introduced, as well as dysfunctional osteocytes in αklotho-deficient state, using animal models

    YOLOv8-ACU: improved YOLOv8-pose for facial acupoint detection

    IntroductionAcupoint localization is integral to Traditional Chinese Medicine (TCM) acupuncture diagnosis and treatment. Employing intelligent detection models for recognizing facial acupoints can substantially enhance localization accuracy.MethodsThis study introduces an advancement in the YOLOv8-pose keypoint detection algorithm, tailored for facial acupoints, and named YOLOv8-ACU. This model enhances acupoint feature extraction by integrating ECA attention, replaces the original neck module with a lighter Slim-neck module, and improves the loss function for GIoU.ResultsThe YOLOv8-ACU model achieves impressive accuracy, with an [email protected] of 97.5% and an [email protected]–0.95 of 76.9% on our self-constructed datasets. It also marks a reduction in model parameters by 0.44M, model size by 0.82 MB, and GFLOPs by 9.3%.DiscussionWith its enhanced recognition accuracy and efficiency, along with good generalization ability, YOLOv8-ACU provides significant reference value for facial acupoint localization and detection. This is particularly beneficial for Chinese medicine practitioners engaged in facial acupoint research and intelligent detection

    CCL21/CCR7 Prevents Apoptosis via the ERK Pathway in Human Non-Small Cell Lung Cancer Cells

    Previously, we confirmed that C-C chemokine receptor 7 (CCR7) promotes cell proliferation via the extracellular signal-regulated kinase (ERK) pathway, but its role in apoptosis of non-small cell lung cancer (NSCLC) cell lines remains unknown. A549 and H460 cells of NSCLC were used to examine the effect of CCL21/CCR7 on apoptosis using flow cytometry. The results showed that activation of CCR7 by its specific ligand, exogenous chemokine ligand 21 (CCL21), was associated with a significant decline in the percent of apoptosis. Western blot and real-time PCR assays indicated that activation of CCR7 significantly caused upregulation of anti-apoptotic bcl-2 and downregulation of pro-apoptotic bax and caspase-3, but not p53, at both protein and mRNA levels. CCR7 small interfering RNA significantly attenuated these effects of exogenous CCL21. Besides, PD98059, a selective inhibitor of MEK that disrupts the activation of downstream ERK, significantly abolished these effects of CCL21/CCR7. Coimmunoprecipitation further confirmed that there was an interaction between p-ERK and bcl-2, bax, or caspase-3, particularly in the presence of CCL21. These results strongly suggest that CCL21/CCR7 prevents apoptosis by upregulating the expression of bcl-2 and by downregulating the expression of bax and caspase-3 potentially via the ERK pathway in A549 and H460 cells of NSCLC

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    Application of a Continuous Terrestrial Photogrammetric Measurement System for Plot Monitoring in the Beijing Songshan National Nature Reserve

    Monitoring sample areas is the basis of ecological management. Songshan National Nature Reserve is one of the most important components of the ecosystem of the central metropolitan area of Beijing, and has fallen behind in its monitoring technology and methods. So, updating the existing equipment and technology is necessary. The current system suffers from high equipment costs and is not convenient to carry, so the work efficiency is low. Furthermore, the data cannot be visualized in three dimensions (3D), and complex terrain conditions cannot be measured. Therefore, this study researched and developed a continuous terrestrial photogrammetric measurement system that is theoretically based on the principles of photogrammetry, image processing technology, and dendrometry. The system applies a self-developed personal digital assistant (PDA) photogrammetry-based dendrometer and software to continuously evaluate stand sampling areas. Through experimental verification, the relative root mean square error (RMSE) of the trunk diameter measurements was found to be 5.59%, and the relative RMSE of hypsometrical measurements was 3.93%, which are both higher than the accuracy required for traditional forestry surveys. Furthermore, the advantages of this system include its low cost, lightweight equipment, easy operation, high measurement efficiency, 3D visualization, and applicability under complex terrain conditions. Since it is highly accurate and efficient, the continuous terrestrial photogrammetric system can be easily applied to monitor stand sampling areas in Songshan National Nature Reserve. In addition, it can be applied to second-class forest surveys in China, thus guaranteeing the monitoring of big data for the ecological environment of China
