    Wells, fields and the emergence of early-to-middle Holocene villages in the Huai River region, China: Contesting regional similarities and differences on prehistoric water management with new archaeological and palaeo-environmental evidence

    The history of water management is closely intertwined with evolution of social structures. In particular, the emergence of rice farming and transition to sedentary life contributed significantly to the development of water management practices, with increasing labour and economic investments to build and operate water-management infrastructures. Here we present archaeological, archaeobotanical and geoarchaeological evidence of early water management obtained from the excavations of three early-to-middle Holocene sites in the upper and middle Huai River region (UMHR), including Hanjing from Sihong County of Jiangsu Province and Xielaozhuang and Zhangwangzhuang from Luohe City and Zhumadian City of Henan Province. Our 14C dates suggest multiple episodes of regional inhabitation in the UMHR region. Our results also reveal that the wells at Xielaozhuang represent some of the earliest attempts to ensure water consumption in the village, and importantly, the rice-field-like structures at Hanjing and Zhangwangzhuang are evidence of a more systematic effort to modify and transform local landscapes for rice cultivation and related economic activities. In line with the results from published phytolith studies, we found that the early experiment of rice cultivation altered local soil and hydrological conditions at these sites. These early forms of rice fields share some similarities in micro-and-macro- morphologies and represent successful adaptations to the low-lying plains that continued to have fluctuating hydrological regimes. The construction and operation of these wells, moats and rice-field-like structures stimulated collaboration beyond individual households and groups in these early-to-middle Holocene villages in the UMHR region and thus would have had profound social and economic ramifications

    Modelling arterial pressure waveforms using Gaussian functions and two-stage particle swarm optimizer

    Changes of arterial pressure waveform characteristics have been accepted as risk indicators of cardiovascular diseases. Waveform modelling using Gaussian functions has been used to decompose arterial pressure pulses into different numbers of subwaves and hence quantify waveform characteristics. However, the fitting accuracy and computation efficiency of current modelling approaches need to be improved. This study aimed to develop a novel two-stage particle swarm optimizer (TSPSO) to determine optimal parameters of Gaussian functions. The evaluation was performed on carotid and radial artery pressure waveforms (CAPW and RAPW) which were simultaneously recorded from twenty normal volunteers. The fitting accuracy and calculation efficiency of our TSPSO were compared with three published optimization methods: the Nelder-Mead, the modified PSO (MPSO), and the dynamic multiswarm particle swarm optimizer (DMS-PSO). The results showed that TSPSO achieved the best fitting accuracy with a mean absolute error (MAE) of 1.1% for CAPW and 1.0% for RAPW, in comparison with 4.2% and 4.1% for Nelder-Mead, 2.0% and 1.9% for MPSO, and 1.2% and 1.1% for DMS-PSO. In addition, to achieve target MAE of 2.0%, the computation time of TSPSO was only 1.5 s, which was only 20% and 30% of that for MPSO and DMS-PSO, respectively

Against the background of China’s new normal, combined with the present economic situations and market demands, the local universities shall start from their own problems and find out ways of adjusting specialties scientifically and then put forward some countermeasures and suggestions to promote a higher and more sustainable development

    Mask Propagation for Efficient Video Semantic Segmentation

    Video Semantic Segmentation (VSS) involves assigning a semantic label to each pixel in a video sequence. Prior work in this field has demonstrated promising results by extending image semantic segmentation models to exploit temporal relationships across video frames; however, these approaches often incur significant computational costs. In this paper, we propose an efficient mask propagation framework for VSS, called MPVSS. Our approach first employs a strong query-based image segmentor on sparse key frames to generate accurate binary masks and class predictions. We then design a flow estimation module utilizing the learned queries to generate a set of segment-aware flow maps, each associated with a mask prediction from the key frame. Finally, the mask-flow pairs are warped to serve as the mask predictions for the non-key frames. By reusing predictions from key frames, we circumvent the need to process a large volume of video frames individually with resource-intensive segmentors, alleviating temporal redundancy and significantly reducing computational costs. Extensive experiments on VSPW and Cityscapes demonstrate that our mask propagation framework achieves SOTA accuracy and efficiency trade-offs. For instance, our best model with Swin-L backbone outperforms the SOTA MRCFA using MiT-B5 by 4.0% mIoU, requiring only 26% FLOPs on the VSPW dataset. Moreover, our framework reduces up to 4x FLOPs compared to the per-frame Mask2Former baseline with only up to 2% mIoU degradation on the Cityscapes validation set.     Quantitative Proteomics Reveals Antibiotics Resistance Function of Outer Membrane Proteins in Aeromonas hydrophila

    Outer membrane proteins (OMPs) play essential roles in antibiotic resistance, particularly in Gram-negative bacteria; however, they still have many unidentified functions regarding their behavior in response to antibiotic stress. In the current work, quantitative tandem mass tag labeling-based mass spectrometry was used to compare the outer membrane related proteins between an oxytetracycline-resistant (OXY-R) and its original control stain (OXY-O) in Aeromonas hydrophila. Consequently, a total of 261 commonly altered proteins in two biological repeats were identified including 29 proteins that increased and 28 that decreased. Gene ontology analysis showed that the expression of transport proteins was significantly reduced, and translation-related proteins were downregulated in the OXY-R strain. After using western blotting to validate selected altered proteins, eight OMP-related genes were knocked out and their roles in antibiotic resistance were further evaluated. The survival assays showed that some mutants such as ΔAHA_4281, ΔAHA_2766, ΔAHA_2282, ΔAHA_1181, and ΔAHA_1280 affected the susceptibility of A. hydrophila to antimicrobials. Moreover, the minimum inhibitory concentration assay showed that these candidate mutants also respond differently to other types of antibiotics. Our results reveal several novel outer membrane related proteins of A. hydrophila that play important roles in antibiotic resistance, and as such, may be helpful for screening studies to identify novel drug targets

    PIM1 Promotes Hepatic Conversion by Suppressing Reprogramming-Induced Ferroptosis and Cell Cycle Arrest

    Protein kinase-mediated phosphorylation plays a critical role in many biological processes. However, the identification of key regulatory kinases is still a great challenge. Here, we develop a trans-omics-based method, central kinase inference, to predict potentially key kinases by integrating quantitative transcriptomic and phosphoproteomic data. Using known kinases associated with anti-cancer drug resistance, the accuracy of our method denoted by the area under the curve is 5.2% to 29.5% higher than Kinase-Substrate Enrichment Analysis. We further use this method to analyze trans-omic data in hepatocyte maturation and hepatic reprogramming of human dermal fibroblasts, uncovering 5 kinases as regulators in the two processes. Further experiments reveal that a serine/threonine kinase, PIM1, promotes hepatic conversion and protects human dermal fibroblasts from reprogramming-induced ferroptosis and cell cycle arrest. This study not only reveals new regulatory kinases, but also provides a helpful method that might be extended to predict central kinases involved in other biological processes

    Repair of Adult Mammalian Heart After Damages by Oral Intake of Gu Ben Pei Yuan San

    Adult mammalian heart repair after myocardial damage is highly inefficient due to the post-mitotic nature of cardiomyocytes. Interestingly, in traditional Chinese medicine (TCM), there are reported effective treatments of myocardial infarction (MI) and heart failure in adult humans by oral intake of a TCM concoction named Gu Ben Pei Yuan San (GBPYS), which is composed of Panax ginseng, velvet antler, Gekko gecko Linnaeus tail, human placenta, Trogopterus dung, Panax notoginseng, and amber. We fed mice with GBPYS after myocardial damages through everyday self-feeding. We then examined the effect of everyday oral intake of GBPYS on improving cardiac function and myocardial repair in adult mice after apical resection or MI. We found that long-term oral intake of GBPYS significantly improved cardiac function after myocardial damages in adult mice. BrdU, phospho-histone 3, and AuroraB staining indicated increased cell proliferation at the border zone of MI after TCM feeding. GBPYS feeding reduced organ inflammation, induced angiogenesis, and is non-toxic to mice after long-term oral intake. Further, serum derived from TCM-fed MI rats promoted division of both neonatal rat cardiomyocytes and human induced pluripotent stem cell (iPSC)-derived cardiomyocytes in vitro. Oral intake of GBPYS improved heart repair after myocardial damages in adult mice. Our results suggest that there are substances present in GBPYS that help improve adult mammalian heart repair after MI. Also, it could be a good choice of non-invasive alternative therapy for myocardial damages and heart failure after rigorous clinical study in the future
