    Static parameter estimation and testability analysis of distribution lines based on multi-source measurement information

    The information monitoring of distribution networks usually relies on multiple measurement systems, and the placement and number of measurement configurations are also restricted by objective factors. It is highly likely that insufficient measurements will affect the parameter estimation of distribution lines. To deal with the aforementioned issue, this paper establishes the measurement equations for static parameters of distribution lines and proposes a method of parameter estimation based on the elitist searching approach. Aiming at different measurement conditions, the testability analysis method is introduced to distinguish clear parameters and uncertain parameters of distribution lines. For the constructed line parameter samples, the estimated values of line parameters are calculated with the consideration of multi-source effective measurement data at different time points. Moreover, the indices of relative errors between the sample values and the estimated values are used to judge the testability of static parameters of distribution lines. The case study on IEEE 33 bus distribution system analyzes the application scenarios with and without measurement errors. The simulation results show that the testability analysis method can effectively identify whether the parameters to be estimated are clear or not, and then the estimated values of clear parameters and the bounds of some disturbing uncertain parameters can be estimated accurately to provide certain decision support for engineering applications

    Metabonomics Combined with UPLC-MS Chemical Profile for Discovery of Antidepressant Ingredients of a Traditional Chinese Medicines Formula, Chaihu-Shu-Gan-San

    This study proposed a new strategy for uncovering the active chemical constituents of a traditional Chinese medicines (TCMs) formula, Chaihu-Shu-Gan-San (CSGS). Metabonomics and chemical profile were integrated in combination with the multivariate statistical analysis (MVA) to discover the chemical constituents which contribute to the antidepressant effect of CSGS. Based upon the difference between CSGS and QZ (CSGS without Zhi-Qiao) extracts in the chemical profiles and the regulations of metabolic disturbances induced by CUMS, synephrine, naringin, hesperidin, and neohesperidin were recognized as the active constituents of CSGS from Zhi-qiao responsible for those missing regulations of CSGS when Zhi-Qiao was subtracted from the whole formula. They participated in the regulations of the deviated metabolites 2–4, 10–14, and 22–25, involved in metabolic pathways of ketone bodies synthesis, phenylalanine, tyrosine and tryptophan biosynthesis, valine, aspartate, glutamate metabolism, and glycolysis/gluconeogenesis. Furthermore, the assay of MAO-A activity confirmed the potential antidepressant effect of naringin and its active sites on the MAO-A was inferred by molecular docking study. The integration of metabonomics and chemical profile was proved to be a useful strategy for uncovering what the active chemical constituents in TCM formula are and how they make contributions for the efficacy of the formula

    The role of Akkermansia muciniphila in inflammatory bowel disease: Current knowledge and perspectives

    Inflammatory bowel diseases, including Crohn’s disease and ulcerative colitis, is a chronic relapsing gastrointestinal inflammatory disease mediated by dysregulated immune responses to resident intestinal microbiota. Current conventional approaches including aminosalicylates, corticosteroids, immunosuppressive agents, and biological therapies are focused on reducing intestinal inflammation besides inducing and maintaining disease remission, and managing complications. However, these therapies are not curative and are associated with various limitations, such as drug resistance, low responsiveness and adverse events. Recent accumulated evidence has revealed the involvement of mucin-degrading bacterium Akkermansia muciniphila (A. muciniphila) in the regulation of host barrier function and immune response, and how reduced intestinal colonisation of probiotic A. muciniphila can contribute to the process and development of inflammatory bowel diseases, suggesting that it may be a potential target and promising strategy for the therapy of inflammatory bowel disease. In this review, we summarise the current knowledge of the role of A. muciniphila in IBD, especially focusing on the related mechanisms, as well as the strategies based on supplementation with A. muciniphila, probiotics and prebiotics, natural diets, drugs, and herbs to promote its colonisation in the gut, and holds promise for A. muciniphila-targeted and -based therapies in the treatment of inflammatory bowel disease

    Novel HYDIN variants associated with male infertility in two Chinese families

    IntroductionInfertility is a major disease affecting human life and health, among which male factors account for about half. Asthenoteratozoospermia accounts for the majority of male infertility. High-throughput sequencing techniques have identified numerous variants in genes responsible for asthenoteratozoospermia; however, its etiology still needs to be studied.MethodIn this study, we performed whole-exome sequencing on samples from 375 patients with asthenoteratozoospermia and identified two HYDIN compound heterozygous variants, a primary ciliary dyskinesia (PCD)-associated gene, in two unrelated subjects. H&E staining, SEM were employed to analyze the varies on sperm of patients, further, TEM was employed to determine the ultrastructure defects. And westernblot and immunostaining were chose to evaluate the variation of structural protein. ICSI was applied to assist the mutational patient to achieve offspring.ResultWe identified two HYDIN compound heterozygous variants. Patient AY078 had novel compound heterozygous splice variants (c.5969-2A>G, c.6316+1G>A), altering the consensus splice acceptor site of HYDIN. He was diagnosed with male infertility and PCD, presenting with decreased sperm progressive motility and morphological abnormalities, and bronchial dilatation in the inferior lobe. Compared to the fertile control, HYDIN levels, acrosome and centrosome markers (ACTL7A, ACROSIN, PLCζ1, and Centrin1), and flagella components (TOMM20, SEPT4, SPEF2, SPAG6, and RSPHs) were significantly reduced in HYDIN-deficient patients. Using intracytoplasmic sperm injection (ICSI), the patient successfully achieved clinical pregnancy. AY079 had deleterious compound heterozygous missense variants, c.9507C>G (p. Asn3169Lys) and c.14081G>A (p. Arg4694His), presenting with infertility; however, semen samples and PCD examination were unavailable.DiscussionOur findings provide the first evidence that the loss of HYDIN function causes asthenoteratozoospermia presenting with various defects in the flagella structure and the disassembly of the acrosome and neck. Additionally, ICSI could rescue this failure of insemination caused by immobile and malformed sperm induced by HYDIN deficiency

    Robust Lifetime Measurement in Large-Scale P2P Systems with Non-Stationary Arrivals

    Characterizing user churn has become an important topic in studying P2P networks, both in theoretical analysis and system design. Recent work [26] has shown that direct sampling of user lifetimes may lead to certain bias (arising from missed peers and round-off inconsistencies) and proposed a technique that estimates lifetimes based on sampled residuals. In this paper, however, we show that under non-stationary arrivals, which are often present in real systems, residual-based sampling does not correctly reconstruct user lifetimes and suffers a varying degree of bias, which in some cases makes estimation completely impossible. We overcome this problem using two contributions: a novel non-stationary ON/OFF churn model and an unbiased randomized residual sampling technique for measuring user lifetimes. The former allows correlation between ON/OFF periods of the same user and exhibits different join rates during the day. The latter spreads sampling points uniformly during the day and uses a novel estimator to reconstruct the underlying lifetime distribution. We finish the paper with experimental measurements of Gnutella and discussing reduction in overhead compared to direct sampling methods

    New 11-Methoxymethylgermacranolides from the Whole Plant of <i>Carpesium divaricatum</i>

    Eight new 11-methoxymethylgermacranolides (1–8) were isolated from the ethanol extract of the whole plant of Carpesium divaricatum. The planar structures and relative configurations of the new compounds were determined by detailed spectroscopic analysis. The absolute configuration of 1 was established by electronic circular dichroism (ECD) spectrum and X-ray crystallographic analysis, and the stereochemistry of the new compounds 2–8 were determined by similar ECD data with 1. The absolute configurations of 5 and 7 were further confirmed by using quantum chemical electronic circular dichroism (ECD) calculations. Compound 4 exhibited weak cytotoxicity against MCF-7 cells. Compound 8 could potently decrease PGE2 productions in LPS-induced RAW 264.7 cells

    A Service-Based Method for Multiple Sensor Streams Aggregation in Fog Computing

    A surge in sensor data volume has exposed the shortcomings of cloud computing, particularly the limitation of network transmission capability and centralized computing resources. The dynamic intervention among sensor streams also brings challenges for IoT applications to derive meaningful information from multiple sensor streams. To handle these issues, this paper proposes a service-based method with fog computing paradigm based on our previous service abstraction, which can capture meaningful events from multiple sensor streams. In our service abstraction, we utilize correlation analysis method to capture events as variations of correlation among sensor streams. Facing inconsistent frequency and shift of correlation, we propose a Dynamic Time Warping- (DTW-) based algorithm to obtain sensor streams’ lag-correlation. For adaptively aggregating related events from different services, we also propose an event routing algorithm to assist the composition of cascaded events through service collaboration. This paper reports the tryout use of our method in Chinese power grid for detecting abnormal situations of power quality. Through a series of experiments based on real sensor data in power grid, we verified that our method can reduce the network transmission and computing resource with high accuracy
