    Rosavin exerts an antitumor role and inactivates the MAPK/ERK pathway in small-cell lung carcinoma in vitro

    This study attempts to explore the function and mechanism of action of rosavin in small-cell lung cancer (SCLC) in vitro. The viability and clone formation of SCLC cells were assessed using cell counting kit-8 and colony formation assays, respectively. Apoptosis and cell cycle were detected using flow cytometry and cell cycle analysis, respectively. Wound healing and transwell assays were performed to evaluate the migration and invasion of SCLC cells. Besides, protein levels of p-ERK, ERK, p-MEK and MEK were determined using western blot analysis. Rosavin repressed the viability and clone formation of SCLC cells, and promoted apoptosis and G0/G1 arrest of SCLC cells. At the same time, rosavin suppressed migration and invasion of SCLC cells. Moreover, protein levels of p-ERK/ERK and p-MEK/MEK were decreased after rosavin addition in SCLC cells. Rosavin impaired malignant behaviors of SCLC cells, which may be associated with inhibition of the MAPK/ERK pathway in vitro

    AUDIT: Audio Editing by Following Instructions with Latent Diffusion Models

    Audio editing is applicable for various purposes, such as adding background sound effects, replacing a musical instrument, and repairing damaged audio. Recently, some diffusion-based methods achieved zero-shot audio editing by using a diffusion and denoising process conditioned on the text description of the output audio. However, these methods still have some problems: 1) they have not been trained on editing tasks and cannot ensure good editing effects; 2) they can erroneously modify audio segments that do not require editing; 3) they need a complete description of the output audio, which is not always available or necessary in practical scenarios. In this work, we propose AUDIT, an instruction-guided audio editing model based on latent diffusion models. Specifically, AUDIT has three main design features: 1) we construct triplet training data (instruction, input audio, output audio) for different audio editing tasks and train a diffusion model using instruction and input (to be edited) audio as conditions and generating output (edited) audio; 2) it can automatically learn to only modify segments that need to be edited by comparing the difference between the input and output audio; 3) it only needs edit instructions instead of full target audio descriptions as text input. AUDIT achieves state-of-the-art results in both objective and subjective metrics for several audio editing tasks (e.g., adding, dropping, replacement, inpainting, super-resolution).     Porcine FcγRIIb Mediates Enhancement of Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) Infection

    Antibody-dependent enhancement (ADE) of virus infection caused by the uptake of virus-antibody complexes by FcγRs is a significant obstacle to the development of effective vaccines to control certain human and animal viral diseases. The activation FcγRs, including FcγRI and FcγRIIa have been shown to mediate ADE infection of virus. In the present paper, we showed that pocine FcγRIIb, an inhibitory FcγR, mediates ADE of PRRSV infection. Stable Marc-145 cell lines expressing poFcγRIIb (Marc-poFcγRII) were established. The relative yield of progeny virus was significantly increased in the presence of sub-neutralization anti-PRRSV antibody. The Fab fragment and normal porcine sera had no effect. Anti-poFcγRII antibody inhibited the enhancement of infection when cells were infected in the presence of anti-PRRSV antibody, but not when cells were infected in the absence of antibody. These results indicate that enhancement of infection in these cells by anti-PRRSV virus antibody is FcγRII-mediated. Identification of the inhibitory FcγR mediating ADE infection should expand our understanding of the mechanisms of pathogenesis for a broad range of infectious diseases and may open many approaches for improvements to the treatment and prevention of such diseases

    Sciences for The 2.5-meter Wide Field Survey Telescope (WFST)

    The Wide Field Survey Telescope (WFST) is a dedicated photometric survey facility under construction jointly by the University of Science and Technology of China and Purple Mountain Observatory. It is equipped with a primary mirror of 2.5m in diameter, an active optical system, and a mosaic CCD camera of 0.73 Gpix on the main focus plane to achieve high-quality imaging over a field of view of 6.5 square degrees. The installation of WFST in the Lenghu observing site is planned to happen in the summer of 2023, and the operation is scheduled to commence within three months afterward. WFST will scan the northern sky in four optical bands (u, g, r, and i) at cadences from hourly/daily to semi-weekly in the deep high-cadence survey (DHS) and the wide field survey (WFS) programs, respectively. WFS reaches a depth of 22.27, 23.32, 22.84, and 22.31 in AB magnitudes in a nominal 30-second exposure in the four bands during a photometric night, respectively, enabling us to search tremendous amount of transients in the low-z universe and systematically investigate the variability of Galactic and extragalactic objects. Intranight 90s exposures as deep as 23 and 24 mag in u and g bands via DHS provide a unique opportunity to facilitate explorations of energetic transients in demand for high sensitivity, including the electromagnetic counterparts of gravitational-wave events detected by the second/third-generation GW detectors, supernovae within a few hours of their explosions, tidal disruption events and luminous fast optical transients even beyond a redshift of 1. Meanwhile, the final 6-year co-added images, anticipated to reach g about 25.5 mag in WFS or even deeper by 1.5 mag in DHS, will be of significant value to general Galactic and extragalactic sciences. The highly uniform legacy surveys of WFST will also serve as an indispensable complement to those of LSST which monitors the southern sky.Comment: 46 pages, submitted to SCMP

    Electric Power Steering System Control Strategy Based on Robust H∞ Control for Electric Forklift

    The dynamic model of the Electric Power Steering (EPS) system for electric forklifts and the two-degree-of-freedom (2DOF) forklift models has been established. Based on the control target of electric forklift EPS, a generalized EPS control model with model perturbation and interference is established. The standard H∞control model of the EPS system is transformed, and the EPS generalized state equation of electric forklift is derived. The principle of robust control with genetic optimization is expounded, aiming at the parameters optimization of the weighted function of H∞control model, the constraint function of the genetic algorithm (GA) is constructed, and the robust controller of genetic optimization is derived. Taking the electric forklift TFC20 as the research object, according to the actual data on the forklift, the system model is established in MATLAB, and the effects of PID control, robust control, and genetic robust control are compared, the results show that the power steering characteristic of the robust control is better than the power steering characteristic of the PID control, and the stability of the forklift under the control of robust control is better. Also, the power steering characteristic of the H∞ controller with GA optimization is better than that of an unoptimized one, and its robustness is better, under external pavement interference, the following ability is stronger for the ideal current and the steering is more stable

    Immersive experiences in museums for elderly with cognitive disorders: a user-centered design approach

    Abstract In the context of global aging, to explore the design needs of elderly with dementia in museum environments, to establish a user cognitive psychological model based on immersion theory, and to enhance the satisfaction of cognitively impaired dementia with the museum service experience. Using literature research, surveys, questionnaires, interviews, and focus groups, we analyze the experience design from the psychological demands of elderly with dementia, build a method of mining user needs by combining the KANO model with the analytic hierarchy process (AHP) method, and establish a model for evaluating the excellence of the experience of the museum environment. The conclusion shows that displaying museum virtual scenes or old objects can effectively increase the subjective well-being of people suffering from various health conditions. The method can accurately tap the attributes of the needs of elderly with dementia, break through the drawbacks of the traditional museum experience design which is dominated by the designer's subjective consciousness, and allow the audience to better experience the museum immersive experience, which provides a new idea and method for the effectiveness of cognitive interventions for elderly with cognitive disorders

    Role of microRNA-92a in metastasis of osteosarcoma cells in vivo and in vitro by inhibiting expression of TCF21 with the transmission of bone marrow derived mesenchymal stem cells

    Abstract Background This study aims to investigate the role of microRNA-92a (miR-92a) in metastasis of osteosarcoma (OS) cells in vivo and in vitro by regulatingTCF21 with the transmission of bone marrow derived mesenchymal stem cells (BMSCs). Methods BMSCs were isolated, purified and cultured from healthy adult bone marrow tissues. The successfully identified BMSCs were co-cultured with OS cells, and the effects of BMSCs on the growth metastasis of OS cells in vitro and in vivo were determined. qRT-PCR and western blot analysis was used to detect the expression of miR-92a and TCF21 in OS cells and OS cells co-cultured with BMSCs. Proliferation, invasion and migration of OS cells transfected with miR-92a mimics and miR-92a inhibitors was determined, and the tumorigenesis and metastasis of OS cells in nude mice were observed. Expression of miR-92a and TCF21 mRNA in tissue specimens as well as the relationship between the expression of miR-92a and the clinicopathological features of OS was analyzed. Results BMSCs promoted proliferation, invasion and migration of OS cells in vitro together with promoted the growth and metastasis of OS cells in vivo. Besides, high expression of miR-92a was found in OS cells co-cultured with BMSCs. Meanwhile, overexpression of miR-92a promoted proliferation, invasion and migration of OS cells in vitro as well as promoted growth and metastasis of OS cells in vivo. The expression of miR-92a increased significantly, and the expression of TCF21 mRNA and protein decreased significantly in OS tissues. Expression of miR-92a was related to Ennecking staging and distant metastasis in OS patients. Conclusion Collectively, this study demonstrates that the expression of miR-92a is high in OS and BMSCs transfers miR-92a to inhibit TCF21 and promotes growth and metastasis of OS in vitro and in vivo

    Comparative analysis of low-energy buildings to help achieve carbon neutrality technology

    The development of low-energy buildings is an important initiative to achieve carbon peaking by 2030 and carbon neutrality by 2060. According to the data of the relevant papers, if all the northern urban and rural buildings in China adopt passive ultra low energy building technology, it can save about 350 million tons of coal for heating and reduce about 900 million tons of carbon dioxide emissions each year. It’s of great significance to achieve the goals of “peak carbon dioxide emissions” and “carbon neutrality”. Starting from four key technologies for low-energy buildings, explanation and analysis the energy-saving methods for low-energy buildings, It also presents the challenges and suggestions for the development of low-energy buildings in China
