    Radical-Enhanced Chinese Character Embedding

    We present a method to leverage radical for learning Chinese character embedding. Radical is a semantic and phonetic component of Chinese character. It plays an important role as characters with the same radical usually have similar semantic meaning and grammatical usage. However, existing Chinese processing algorithms typically regard word or character as the basic unit but ignore the crucial radical information. In this paper, we fill this gap by leveraging radical for learning continuous representation of Chinese character. We develop a dedicated neural architecture to effectively learn character embedding and apply it on Chinese character similarity judgement and Chinese word segmentation. Experiment results show that our radical-enhanced method outperforms existing embedding learning algorithms on both tasks.Comment: 8 pages, 4 figure

    A Parallel FP-Growth Mining Algorithm with Load Balancing Constraints for Traffic Crash Data

    Traffic safety is an important part of the roadway in sustainable development. Freeway traffic crashes typically cause serious casualties and property losses, being a serious threat to public safety. Figuring out the potential correlation between various risk factors and revealing their coupling mechanisms are of effective ways to explore and identity freeway crash causes. However, the existing association rule mining algorithms still have some limitations in both efficiency and accuracy. Based on this consideration, using the freeway traffic crash data obtained from WDOT (Washington Department of Transportation), this research constructed a multi-dimensional multilevel system for traffic crash analysis. Considering the load balancing, the FP-Growth (Frequent Pattern- Growth) algorithm was optimized parallelly based on Hadoop platform, to achieve an efficient and accurate association rule mining calculation for massive amounts of traffic crash data; then, according to the results of the coupling mechanism among the crash precursors, the causes of freeway traffic crashes were identified and revealed. The results show that the parallel FPgrowth algorithm with load balancing constraints has a better operating speed than both the conventional FP-growth algorithm and parallel FP-growth algorithm towards processing big data. This improved algorithm makes full use of Hadoop cluster resources and is more suitable for large traffic crash data sets mining while retaining the original advantages of conventional association rule mining algorithm. In addition, the mining association rules model with the improvement of multi-dimensional interaction proposed in this research can catch the occurrence mechanism of freeway traffic crash with serious consequences (lower support degree probably) accurately and efficiently

    Optimization Model of Taxi Fleet Size Based on GPS Tracking Data

    reasonable taxi fleet size has a significant impact on the satisfaction of urban traffic demand, the alleviation of urban traffic congestion, and the stability of taxi business groups. Most existing studies measure the overall scale by using macro indices, and few studies are from the micro level. To meet the transportation demand for taxis, mitigating the mismatch between taxi supply and demand, this research proposes an urban taxi fleet size calculating model based on GPS tracking data. Firstly, on the basis of road network segmentation, the probability model of a passenger taxi-taking a road section as a unit is built to evaluate the difficulty of taxi-taking on a road section. Furthermore, a user queuing model is built for the &ldquo difficult to take a taxi&rdquo road section in the peak period, and the service mileage required by potential taxi users is calculated. After that, a transportation capacity measurement model is built to estimate the number of taxis required in different time periods, Finally, the income constraint model is used to explain the impact of different vehicle fleet sizes on the income of taxi groups, so as to provide a reference for the determination of the final fleet size. The model is applied to data from Xi&rsquo an. The calculation results are based on data from May 2014, and show that the scale of taxi demand is about 654&ndash 2237, and after considering the impact of different fleet size increases on income, when the income variation index is limited to 0.10, i.e., the decrease of drivers&rsquo income will not exceed 10%, an increase of 1286 taxis will be able to meet 66% of the unmet demand in the peak period. The conclusion indicates that the model can effectively calculate the required fleet size and formulate the constraint solutions.     Promotive role of USP29-mediated deubiquitination in malignant proliferation of colorectal cancer cells via the KIAA1429/SOX8 axis

    Colorectal cancer (CRC) is regarded as one of the most prevalent neoplasms worldwide, and ubiquitination and N6-methyladenosine (m6A) modification regulate the outgrowth of multiple cancers. This study attempted to explore the effect of ubiquitin-specific peptidases 29 (USP29) on the malignant proliferation of CRC cells via stabilizing Vir-like m6A methyltransferase associated (VIRMA/KIAA1429). First, upregulations of USP29, KIAA1429, and SRY-box transcription factor 8 (SOX8) were found in CRC tissues and cells through real-time quantitative polymerase chain reaction and Western blotting. After transfection of si-USP29, the proliferation of CRC cells was evaluated by the cell counting kit-8, colony formation and 5-ethynyl-2’-deoxyuridine assays, and we observed that depletion of USP29 inhibited the proliferation of CRC cells. Co-immunoprecipitation confirmed the binding of USP29 to KIAA1429. Mechanically, USP29 mediated deubiquitination to stabilize the protein levels of KIAA1429, and KIAA1429 promoted the stability of SOX8 mRNA through m6A modification. Moreover, overexpression of KIAA1429 or SOX8 reversed the inhibitory effects of USP29 depletion on CRC cell proliferation. Finally, the xenograft tumor model revealed the promotive role of USP29 in the proliferation of CRC cells in vivo. Altogether, USP29 facilitates the malignant proliferation of CRC cells via upregulating the KIAA1429/SOX8 axis

    A Mismatch-Tolerant Reverse Transcription Loop-Mediated Isothermal Amplification Method and Its Application on Simultaneous Detection of All Four Serotype of Dengue Viruses

    Loop-mediated isothermal amplification (LAMP) has been widely used in the detection of pathogens causing infectious diseases. However, mismatches between primers (especially in the 3′-end) and templates significantly reduced the amplification efficiency of LAMP, and limited its application to genetically diverse viruses. Here, we reported a novel mismatch-tolerant LAMP assay and its application in the detection of dengue viruses (DENV). The novel method features the addition of as little as 0.15 U of high-fidelity DNA polymerase to the standard 25 μl LAMP reaction mixture. This amount was sufficient to remove the mismatched bases at the 3′-end of primers, thereby resulting in excellent tolerance for various mismatches occurring at the 3′-end of the LAMP primers during amplification. This novel LAMP assay has a markedly improved amplification efficiency especially for the mutants forming mismatches with internal primers (FIP/BIP) and loop primers (FLP/BLP). The reaction time of the novel method was about 5.6–22.6 min faster than the conventional LAMP method regardless of the presence or absence of mismatches between primers and templates. Using the novel method, we improved a previously established pan-serotype assay for DENV, and demonstrated greater sensitivity for detection of four DENV serotypes than the previous one. The limit of detection (LOD) of the novel assay was 74, 252, 78, and 35 virus RNA copies per reaction for DENV-1, DENV-2, DENV-3, and DENV-4, respectively. Among 153 clinical samples from patients with suspected DENV infection, the novel assay detected 94.8% samples being DENV positive, higher than that detected by the commercial NS1 antigen assay (92.2%), laboratory-based RT-PCR method (78.4%), and the conventional RT-LAMP assay (86.9%). Furthermore, the novel RT-LAMP assay has been developed into a visual determination method by adding colorimetric dyes. Because of its simplicity, all LAMP-based diagnostic assays may be easily updated to the newly improved version. The novel mismatch-tolerant LAMP method represents a simple, sensitive and promising approach for molecular diagnosis of highly variable viruses, and it is especially suited for application in resource-limited settings
