Heisenberg-limited spin squeezing in coupled spin systems
Spin squeezing plays a crucial role in quantum metrology and quantum
information science. Its generation is the prerequisite for further
applications but still faces an enormous challenge since the existing physical
systems rarely contain the required squeezing interactions. Here we propose a
universal scheme to generate spin squeezing in coupled spin models with
collective spin-spin interactions, which commonly exist in various systems. Our
scheme can transform the coupled spin interactions into squeezing interactions,
and reach the extreme squeezing with Heisenberg-limited measurement precision
scaling as for particles. Only constant and continuous driving fields
are required, which is accessible to a series of current realistic experiments.
This work greatly enriches the variety of systems that can generate the
Heisenberg-limited spin squeezing, with broad applications in quantum precision
measurement
Climate change and its influence on the Karst groundwater recharge in the Jinci Spring Region, Northern China
Due to climate change and human activities over the last fifty years, the spring flow
volume of karst groundwater has sharply diminished in China. Climate change is one of the critical
factors that initiates a series of karst hydrogeologic and water ecological environmental problems,
because the precipitation shows a decreasing trend while the temperature shows an increasing trend.
The Jinci Spring is one of the largest, most famous springs in northern China. This study employed
data from the Taiyuan Meteorological Station and ten precipitation stations in and around the Jinci
Spring region as well as the runoff data gathered from two hydrological monitoring stations during
1960-2012. The sliding average method and the Mann-Kendall test were used to analyze the variation
tendency of precipitation, temperature, and land evaporation in this area. Finally, the following
were calculated: the varying pattern of the karst groundwater recharge amount and the response
of the recharge amount to precipitation, land evaporation, and river runoff by quantitative analysis.
The results indicated that the precipitation and land evaporation amount decreased at first and then
subsequently increased. Likewise, the variation trend of the karst groundwater recharge amount
in the spring region was roughly consistent with the precipitation variation pattern. In contrast,
the temperature displayed an increasing trend. The climate change resulted in a reduction of the
karst groundwater recharge amount, and it had the greatest influence in the 1990s, which caused the
karst groundwater recharge amount to decrease 26.75 mm as compared to that of the 1960s (about
39.68% lower than that of the 1960s). The Jinci Spring had zero flow during this period. The reduction
in precipitation was one of main factors that caused the cutoff of the Jinci Spring.IS
Identification, physical mechanisms and impacts of drought–flood abrupt alternation: a review
Climate change has led to an increase in the frequency of extreme events, such as droughts and floods. This study aims to review the literature on the newly proposed phenomenon known as drought-flood abrupt alternation (DFAA). A comprehensive summary is provided to round up the numerous approaches employed to identify DFAA events, as well as its mechanisms and impacts. To provide a reference for responding and managing the emerging intensity and frequency of DFAA events, we conclude the paper by listing the insufficiency of current research and suggesting possible future research directions. As for the impact of DFAA, besides the loss of life and property which can be caused by any natural disaster, a DFAA event severely threatens food security by making a lasting and profound impact on the land productivity through the alteration of the combining conditions of water, soil, and temperature. As for the future research directions, existing indexes developed for DFAA identification should be improved by downscaling the temporal and spatial scale, with interactions of neighboring drought and flood events taken into consideration. What’s more, to better protect human society from the losses caused by DFAA, researches on accurate DFAA prediction are encouraged
Immunological Responses to Transgene-Modified Neural Stem Cells After Transplantation
Neural stem cell (NSC) therapy is a promising therapeutic strategy for stroke. Researchers have frequently carried out genetic modification or gene editing of stem cells to improve survival or therapeutic function. However, NSC transplantation carries the risk of immune rejection, and genetic modification or gene-editing might further increase this risk. For instance, recent studies have reported on manipulating the stem cell genome and transplantation via the insertion of an exogenous gene derived from magnetotactic bacteria. However, whether transgene-modified stem cells are capable of inducing immunological reactions has not been explored. Although NSCs rarely express the major histocompatibility complex (MHC), they can still cause some immunological issues. To investigate whether transgene-modified NSCs aggravate immunological responses, we detected the changes in peripheral immune organs and intracerebral astrocytes, glial cells, and MHC-I and MHC-II molecules after the injection of GFP-labeled or mms6-GFP-labeled NSCs in a rat model. Xenogeneic human embryonic kidney (HEK-293T) cells were grafted as a positive control group. Our results indicated that xenogeneic cell transplantation resulted in a strong peripheral splenic response, increased astrocytes, enhanced microglial responses, and upregulation of MHC-I and MHC-II expression on the third day of transplantation. But they decreased obviously except Iba-1 positive cells and MHC-II expression. When injection of both mms6-GFP-labeled NSCs and GFP-labeled NSCs also induced similar responses as HEK-293T cells on the third days, but MHC-I and MHC-II expression decreased 3 weeks after transplantation. In addition, mms6 transgene-modified NSCs did not produce peripheral splenic response responses as well as astrocytes, microglial cells, MHC-I and MHC-II positive cells responses when compared with non-modified NSCs. The present study provides preliminary evidence that transgenic modification does not aggravate immunological responses in NSC transplantation
Metastatic patterns and prognosis of patients with primary malignant cardiac tumor
BackgroundDistant metastases are independent negative prognostic factors for patients with primary malignant cardiac tumors (PMCT). This study aims to further investigate metastatic patterns and their prognostic effects in patients with PMCT.Materials and methodsThis multicenter retrospective study included 218 patients with PMCT diagnosed between 2010 and 2017 from Surveillance, Epidemiology, and End Results (SEER) database. Logistic regression was utilized to identify metastatic risk factors. A Chi-square test was performed to assess the metastatic rate. Kaplan–Meier methods and Cox regression analysis were used to analyze the prognostic effects of metastatic patterns.ResultsSarcoma (p = 0.002) and tumor size¿4 cm (p = 0.006) were independent risk factors of distant metastases in patients with PMCT. Single lung metastasis (about 34%) was the most common of all metastatic patterns, and lung metastases occurred more frequently (17.9%) than bone, liver, and brain. Brain metastases had worst overall survival (OS) and cancer-specific survival (CSS) among other metastases, like lung, bone, liver, and brain (OS: HR = 3.20, 95% CI: 1.02–10.00, p = 0.046; CSS: HR = 3.53, 95% CI: 1.09–11.47, p = 0.036).ConclusionPatients with PMCT who had sarcoma or a tumor larger than 4 cm had a higher risk of distant metastases. Lung was the most common metastatic site, and brain metastases had worst survival among others, such as lung, bone, liver, and brain. The results of this study provide insight for early detection, diagnosis, and treatment of distant metastases associated with PMCT
TP53-related signature for predicting prognosis and tumor microenvironment characteristics in bladder cancer: A multi-omics study
Background: The tumor suppressor gene TP53 is frequently mutated or inactivated in bladder cancer (BLCA), which is implicated in the pathogenesis of tumor. However, the cellular mechanisms of TP53 mutations are complicated, yet well-defined, but their clinical prognostic value in the management of BLCA remains controversial. This study aimed to explore the role of TP53 mutation in regulating the tumor microenvironment (TME), elucidate the effects of TP53 activity on BLCA prognosis and immunotherapy response.Methods: A TP53-related signature based on TP53-induced and TP53-repressed genes was used to construct a TP53 activity-related score and classifier. The abundance of different immune cell types was determined using CIBERSORT to estimate immune cell infiltration. Moreover, the heterogeneity of the tumor immune microenvironment between the high and low TP53 score groups was further evaluated using single-cell mass cytometry (CyTOF) and imaging mass cytometry (IMC). Moreover, pathway enrichment analysis was performed to explore the differential biological functions between tumor epithelial cells with high and low TP53 activity scores. Finally, the receptor–ligand interactions between immune cells and tumor epithelial cells harboring distinct TP53 activity were analyzed by single-cell RNA-sequencing.Results: The TP53 activity-related gene signature differentiated well between TP53 functional retention and inactivation in BLCA. BLCA patients with low TP53 scores had worse survival prognosis, more TP53 mutations, higher grade, and stronger lymph node metastasis than those with high TP53 scores. Additionally, CyTOF and IMC analyses revealed that BLCA patients with low TP53 scores exhibited a potent immunosuppressive TME. Consistently, single-cell sequencing results showed that tumor epithelial cells with low TP53 scores were significantly associated with high cell proliferation and stemness abilities and strongly interacted with immunosuppressive receptor–ligand pairs.Conclusion: BLCA patients with low TP53 scores have a worse prognosis and a more immunosuppressive TME. This TP53 activity-related signature can serve as a potential prognostic signature for predicting the immune response, which may facilitate the development of new strategies for immunotherapy in BLCA
Internet-Based HIV Self-Testing Among Men Who Have Sex With Men Through Pre-exposure Prophylaxis: 3-Month Prospective Cohort Analysis From China.
BACKGROUND: Routine HIV testing accompanied with pre-exposure prophylaxis (PrEP) requires innovative support in a real-world setting. OBJECTIVE: This study aimed to determine the usage of HIV self-testing (HIVST) kits and their secondary distribution to partners among men who have sex with men (MSM) in China, who use PrEP, in an observational study between 2018 and 2019. METHODS: In 4 major cities in China, we prospectively followed-up MSM from the China Real-world oral PrEP demonstration study, which provides daily or on-demand PrEP for 12 months, to assess the usage and secondary distribution of HIVST on quarterly follow-ups. Half of the PrEP users were randomized to receive 2 HIVSTs per month in addition to quarterly facility-based HIV testing. We evaluated the feasibility of providing HIVST to PrEP users. RESULTS: We recruited 939 MSM and randomized 471 to receive HIVST, among whom 235 (49.9%) were daily and 236 (50.1%) were on-demand PrEP users. At baseline, the median age was 29 years, 390 (82.0%) men had at least college-level education, and 119 (25.3%) had never undergone facility-based HIV testing before. Three months after PrEP initiation, 341 (74.5%) men had used the HIVST provided to them and found it very easy to use. Among them, 180 of 341 (52.8%) men had distributed the HIVST kits it to other MSM, and 132 (51.6%) among the 256 men who returned HIVST results reported that used it with their sexual partners at the onset of intercourse. Participants on daily PrEP were more likely to use HIVST (adjusted hazard ratio=1.3, 95% CI 1.0-1.6) and distribute HIVST kits (adjusted hazard ratio=1.3, 95% CI 1.1-1.7) than those using on-demand PrEP. CONCLUSIONS: MSM who used PrEP had a high rate of usage and secondary distribution of HIVST kits, especially among those on daily PrEP, which suggested high feasibility and necessity for HIVST after PrEP initiation. Assuming that fourth-generation HIVST kits are available, HIVST may be able to replace facility-based HIV testing to a certain extent.
