    Utility of clinical metagenomics in diagnosing malignancies in a cohort of patients with Epstein-Barr virus positivity

    BackgroundsDifferentiation between benign and malignant diseases in EBV-positive patients poses a significant challenge due to the lack of efficient diagnostic tools. Metagenomic Next-Generation Sequencing (mNGS) is commonly used to identify pathogens of patients with fevers of unknown-origin (FUO). Recent studies have extended the application of Next-Generation Sequencing (NGS) in identifying tumors in body fluids and cerebrospinal fluids. In light of these, we conducted this study to develop and apply metagenomic methods to validate their role in identifying EBV-associated malignant disease.MethodsWe enrolled 29 patients with positive EBV results in the cohort of FUO in the Department of Infectious Diseases of Huashan Hospital affiliated with Fudan University from 2018 to 2019. Upon enrollment, these patients were grouped for benign diseases, CAEBV, and malignant diseases according to their final diagnosis, and CNV analysis was retrospectively performed in 2022 using samples from 2018 to 2019.ResultsAmong the 29 patients. 16 of them were diagnosed with benign diseases, 3 patients were diagnosed with CAEBV and 10 patients were with malignant diseases. 29 blood samples from 29 patients were tested for mNGS. Among all 10 patients with malignant diagnosis, CNV analysis suggested neoplasms in 9 patients. Of all 19 patients with benign or CAEBV diagnosis, 2 patients showed abnormal CNV results. The sensitivity and specificity of CNV analysis for the identification for tumors were 90% and 89.5%, separately.ConclusionsThe application of mNGS could assist in the identification of microbial infection and malignancies in EBV-related diseases. Our results demonstrate that CNV detection through mNGS is faster compared to conventional oncology tests. Moreover, the convenient collection of peripheral blood samples adds to the advantages of this approach

    Effects of Users’ Familiarity With the Objects Depicted in Icons on the Cognitive Performance of Icon Identification

    This study investigated the effects of users’ familiarity with the objects depicted in icons on the cognitive performance of icon identification. First, without knowing the specific semantic information of icons, 20 participants were required to search for target icons among visually similar distractors for 3-hour-long training sessions across 1 week, during which their familiarity with different icons was manipulated by differential exposure frequencies. Half of the icons were presented 10 times more often than the other half. Subsequently, participants’ abilities to recall corresponding semantic information when cued with associated target icons were tested after they had learned all the icons. The results showed that, in both the visual search task and the semantic information recall task, participants performed significantly better when the icons were more familiar. Importantly, the effects of icon complexity in the visual search task diminished as participants became familiar with the icons, and the beneficial effects of familiarity in the semantic information recall task were larger when the icons were complex. These findings have practical implications for icon design. When creating new icons for time critical user interfaces, icons should be kept as simple as possible and employ familiar, commonly used, graphics

    Do corporate site visits affect the informational role of independent directors?

    This study examines the influence of corporate site visits on information acquisition and dissemination by independent directors (IDs). Employing two unique sources of data from Chinese listed firms based on the mandatory disclosure of IDs’ (i) site visit activities and (ii) opinions about corporate decisions, we find that the acquisition of firm-specific information and the dissemination of such information to the market is greater among IDs who conduct visits than their counterparts without such visits. Moreover, this effect is more pronounced in firms with complex information environments, lower proprietary costs, and among IDs with greater internal information asymmetry, superior external information, and stronger incentives and abilities to perform their duties. Furthermore, the firm-specific information disseminated by IDs leads to stronger market reactions and improves stock price efficiency. Our study shows that site visits contribute to the informational role of IDs, and our evidence offers important corporate governance and regulatory implications

    Quantitative Analysis of Industrial Solid Waste Based on Terahertz Spectroscopy

    Industrial solid waste refers to the solid waste that is produced in industrial production activities. Without correct treatment and let-off, industrial solid waste may cause environmental pollution due to a variety of pollutants and toxic substances that are contained in it. Conventional detection methods for identifying harmful substances are high performance liquid chromatography (HPLC) and gas chromatography-mass spectrometry (GC-MS), which are complicated, time-consuming, and highly demanding for the testing environment. Here, we propose a method for the quantitative analysis of harmful components in industrial solid waste by using terahertz (THz) spectroscopy combined with chemometrics. Pyrazinamide, benazepril, cefprozil, and bisphenol A are four usual hazardous components in industrial solid waste. By comparing with the Raman method, the THz method shows a much higher accuracy for their concentration analysis (90.3–99.8% vs. 11.7–86.9%). In addition, the quantitative analysis of mixtures was conducted, and the resulting prediction accuracy rate was above 95%. This work has high application value for the rapid, accurate, and low-cost detection of industrial solid waste

    Research on the evaluation of green suppliers of high energy-consuming enterprises--based on rough number-grey correlation TOPSIS method

    With the Goal-Establishment of “carbon compliance” and “carbon neutrality”, enterprises should upgrade “green development” to a strategic position and implement it through all aspects of business operations. Green supplier selection is the initial phase of supply chain management, therefore a green supplier evaluation system is needed to achieve green development. Based on a literature analysis, We selected 45 metrics as candidates for evaluating suppliers. Then through expert interviews, some indicators were revised and supplemented, and finally a green supplier evaluation index system for high energy-consuming enterprises was constructed. A unique aspect of this paper is the introduction of rough number theory into the supplier evaluation process to improve the indicator assignment and the grey correlation TOPSIS method, which optimizes the processing of uncertain semantic information in the evaluation process. The rough number-grey correlation TOPSIS supplier evaluation model developed in this paper has been verified to be applicable and stable in case studies and successfully implemented

    Identification and Expression Pattern of <i>cyp26b1</i> Gene in Gonad of the Chinese Tongue Sole (<i>Cynoglossus semilaevis</i>)

    As an RA-metabolizing enzyme, cyp26b1 has a substantial impact on RA-signaling pathways. The cyp26b1 gene from the Chinese tongue sole was cloned and identified in this investigation. The cyp26b1 ORF was 1536 bp in length and encoded a 512 amino acid protein. A quantitative real-time PCR (qPCR) indicated that the cyp26b1 expression is no significant sexual dimorphism in the gonads at the 80 days post-hatching (dph) stages. After 4 months post-hatching (mph), the expression of cyp26b1 showed sexual dimorphism and lower level of expression in the ovaries than in the testes. An in situ hybridization demonstrated that cyp26b1 mRNA was primarily located in the testis. Interestingly, the cyp26b1 mRNA probe was also detected in the ovaries. These results suggested that cyp26b1 participates in the sex-differentiation and gonadal development of the Chinese tongue sole

    Effect of histone deacetylase inhibitor romidepsin on the in vitro growth of foetal fibroblast cells and early development of porcine-cloned embryos

    This study was designed to explore the effects of histone deacetylase inhibitor romidepsin (FK228) treatment on the morphology, proliferation and karyotype of porcine foetal fibroblast cells and early developmental competence of somatic cell nuclear transfer (SCNT) embryos. We found that the treatment of foetal fibroblast cells with 0.1μM FK228 for 24 h did not alter normal morphology, proliferation rate and chromosome number of donor cells, while other treatments with different does and durations altered the above characteristics of donor cells seriously. Simultaneously, fusion rate, blastocyst rate and total cell number per SCNT blastocysts from different donor cell treatment groups were similar to the control group. We further found that the treatment of SCNT embryos with low-dose FK228 did not affect their developmental efficiency, but treatment with high dose dramatically caused blastocyst formation failure. In addition, 0.1μM FK228 treatment for 36 h significantly elevated total cell number per SCNT blastocysts. Finally, combined treatments of both donor cells and embryos significantly improved the cleavage rate of cloned embryos, but did not affect the blastocyst rate and total cell number. Taken together, histone deacetylase inhibitor FK228 with optimal dose and exposure duration can enhance early developmental efficiency of porcine SCNT embryos and blastocyst quality

    Autophagy Activation by Rapamycin Before Hypoxia-Reoxygenation Reduces Endoplasmic Reticulum Stress in Alveolar Epithelial Cells

    Background: To determine potential effects of autophagy activation on hypoxia-reoxygenation (H/R) induced damage of a rat alveolar epithelial cell line. Methods: CCL149 cells were subjected to autophagy agonist (rapamycin, Rap), autophagy inhibitor (3-methyladenine, 3-MA) or PBS for 1 h before H/R treatment for 2 h, 4 h and 6 h. The optimal concentration of Rap (150 nM, 200 nM and 250 nM) or 3-MA (5 mM, 10 mM and 15 mM) was obtained from MTT assay. Autophagy was determined by fluorescence microscopy of eRFP-LC3 positive cells, transmission electron microscopy of autophagosome, western blot of LC3, AMPK, Beclin-1, HDAC6 and p62 proteins. Endoplasmatic reticulum stress was indicated by detecting expressions of BIP, XBP-1 and CHOP via western blot. Results: Rap at concentration of 250 nM before H/R increased the autophagy formation with more eRFP-LC3 positive cells and higher expressions of LC3-II, Beclin-1, HDAC6 and p62, but lower expressions of BIP, XBP-1 and CHOP in H/R treated CCL149. This effect seemed to be still obvious after H/R exposure for 6 h. The contrary results were obtained by treatment with 5 mM 3-MA. Conclusion: Rap might be a promising agent before mechanical ventilation or reperfusion to prevent re-damage in hypoxia related lung diseases
