    Intestinal gene expression in pigs: effects of reduced feed intake during weaning and potential impact of dietary components

    The weaning transition is characterised by morphological, histological and microbial changes, often leading to weaning-associated disorders. These intestinal changes can partly be ascribed to the lack of luminal nutrition arising from the reduced feed intake common in pigs after weaning. It is increasingly becoming clear that changes in the supply with enteral nutrients may have major impacts on intestinal gene expression. Furthermore, the major dietary constituents, i.e. carbohydrates, fatty acids and amino acids, participate in the regulation of intestinal gene expression. However, nutrients may also escape digestion by mammalian enzymes in the upper gastrointestinal tract. These nutrients can be used by the microflora, resulting in the production of bacterial metabolites, for example, SCFA, which may affect intestinal gene expression indirectly. The present review provides an insight on possible effects of reduced feed intake on intestinal gene expression, as it may occur post-weaning. Detailed knowledge on effects of reduced feed intake on intestinal gene expression may help to understand weaning-associated intestinal dysfunctions and diseases. Examples are given of intestinal genes which may be altered in their expression due to supply with specific nutrients. In that way, gene expression could be modulated by dietary means, thereby acting as a potential therapeutic tool. This could be achieved, for example, by influencing genes coding for digestive or absorptive proteins, thus optimising digestive function and metabolism, but also with regard to immune response, or by influencing proliferative processes, thereby enhancing mucosal repair. This would be of special interest when designing a diet to overcome weaning-associated problem

    Ca plays an essential role in bone development; however, little is known about its effect on intestinal gene expression in juvenile animals. In the present study, thirty-two weaned pigs (9·5 (sem 0·11) kg) were assigned to four diets that differed in Ca concentration (adequate v. high) and cereal composition (wheat–barley v. maize) to assess the jejunal and colonic gene expression of nutrient transporters, tight junction proteins, cytokines and pathogen-associated molecular patterns, nutrient digestibility, Ca balance and serum acute-phase response. To estimate the impact of mucosal bacteria on colonic gene expression, Spearman's correlations between colonic gene expression and bacterial abundance were computed. Faecal Ca excretion indicated that more Ca was available along the intestinal tract of the pigs fed high Ca diets as compared to the pigs fed adequate Ca diets (P0·35; P< 0·05) indicated an association between operational taxonomic units assigned to the phyla Bacteroidetes, Firmicutes and Proteobacteria and bacterial metabolites and mucosal gene expression in the colon. The present results indicate that high Ca diets have the potential to modify the jejunal and colonic mucosal gene expression response which, in turn, interacts with the composition of the basal diet and mucosa-associated bacteria in weaned pigs

    Differences in intestinal size, structure, and function contributing to feed efficiency in broiler chickens reared at geographically distant locations

It is also not clear if RFI-related differences in intestinal function are similar in chickens raised in different environments. The objective was to investigate differences in nutrient retention, visceral organ size, intestinal morphology, jejunal permeability and expression of genes related to barrier function, and innate immune response in chickens of diverging RFI raised at 2 locations (L1: Austria; L2: UK). The experimental protocol was similar, and the same dietary formulation was fed at the 2 locations. Individual BW and feed intake (FI) of chickens (Cobb 500FF) were recorded from d 7 of life. At 5 wk of life, chickens (L1, n = 157; L2 = 192) were ranked according to their RFI, and low, medium, and high RFI chickens were selected (n = 9/RFI group, sex, and location). RFI values were similar between locations within the same RFI group and increased by 446 and 464 g from low to high RFI in females and males, respectively. Location, but not RFI rank, affected growth, nutrient retention, size of the intestine, and jejunal disaccharidase activity. Chickens from L2 had lower total body weight gain and mucosal enzyme activity but higher nutrient retention and longer intestines than chickens at L1. Parameters determined only at L1 showed increased crypt depth in the duodenum and jejunum and enhanced paracellular permeability in low vs. high RFI females. Jejunal expression of IL1B was lower in low vs. high RFI females at L2, whereas that of TLR4 at L1 and MCT1 at both locations was higher in low vs. high RFI males. Correlation analysis between intestinal parameters and feed efficiency metrics indicated that feed conversion ratio was more correlated to intestinal size and function than was RFI. In conclusion, the rearing environment greatly affected intestinal size and function, thereby contributing to the variation in chicken RFI observed across locations

    The objective of this study was to examine the influences of different levels of grain in the ration on clinical parameters and histological variables of the rumen epithelium. Seventeen 4-months old goats were allocated to three feeding groups (hay, 30 % grain, 60 % grain). After an adaptation time of three weeks the goats were fed four weeks the experimental ration, weekly adjusted to the body weight. Pulse, respiration, rumen activity and fecal consistency were recorded at the beginning and the end of the adaptation and experimental period, respectively. In the first and fifth week rumen liquid was collected via tube. At the end of the experiment goats were euthanized and rumen liquid was collected for pH measuring, and rumen epithelium for histological examination. Feeding grain in 4 months old goats had influence on clinical parameters (within physiological ranges) and fecal score. A differentiation of the rumen epithelium was recorded with thickness of the stratum corneum. Feeding more grain was associated with lowered rumen pH. In conclusion, although clinical and histological changes were detected in goats in response to feeding different amounts of grain, these changes did not show consequences for clinical health except for fecal consistency

    Role of physically effective fiber and estimation of dietary fiber adequacy in high-producing dairy cattle

    Highly fermentable diets require the inclusion of adequate amounts of fiber to reduce the risk of subacute rumen acidosis (SARA). To assess the adequacy of dietary fiber in dairy cattle, the concept of physically effective neutral detergent fiber (peNDF) has received increasing attention because it amalgamates information on both chemical fiber content and particle size (PS) of the feedstuffs. The nutritional effects of dietary PS and peNDF are complex and involve feed intake behavior (absolute intake and sorting behavior), ruminal mat formation, rumination and salivation, and ruminal motility. Other effects include fermentation characteristics, digesta passage, and nutrient intake and absorption. Moreover, peNDF requirements depend on the fermentability of the starch source (i.e., starch type and endosperm structure). To date, the incomplete understanding of these complex interactions has prevented the establishment of peNDF as a routine method to determine dietary fiber adequacy so far. Therefore, this review is intended to analyze the quantitative effects of and interactions among forage PS, peNDF, and diet fermentability with regard to rumen metabolism and prevention of SARA, and aims to give an overview of the latest achievements in the estimation of dietary fiber adequacy in high-producing dairy cattle. Recently developed models that synthesize the effects of both peNDF and fermentable starch on rumen metabolism appear to provide an appropriate basis for estimation of dietary fiber adequacy in high-producing dairy cows. Data suggest that a period lasting more than 5 to 6h/d during which ruminal pH is <5.8 should be avoided to minimize health disturbances due to SARA. The knowledge generated from these modeling approaches recommends that average amounts of 31.2% peNDF inclusive particles >1.18mm (i.e., peNDF(>1.18)) or 18.5% peNDF inclusive particles >8mm (i.e., peNDF(>8)) in the diet (DM basis) are required. However, inclusion of a concentration of peNDF(>8) in the diet beyond 14.9% of diet DM may lower DM intake level. As such, more research is warranted to develop efficient feeding strategies that encourage inclusion of energy-dense diets without the need to increase their content in peNDF above the threshold that leads to lower DM intake. The latter would require strategies that modulate the fermentability characteristics of the diet and promote absorption and metabolic capacity of ruminal epithelia of dairy cows

    Feeding barley grain-rich diets altered electrophysiological properties and permeability of the ruminal wall in a goat model

    High-producing ruminants are commonly fed large amounts of concentrate to meet their high energy demands for rapid growth or high milk production. However, this feeding strategy can severely impair rumen functioning, leading to subacute ruminal acidosis. Subacute ruminal acidosis might have consequences for electrophysiological properties by changing the net ion transfer and permeability of ruminal epithelia, which may increase the uptake of toxic compounds generated in the rumen into the systemic circulation. The objective of the present study was to investigate the effects of excessive barley feeding on the electrophysiological and barrier functions of the ruminal epithelium and serum inflammation and ketogenesis markers after a long-term feeding challenge, using growing goats as a ruminant model. A feeding trial was carried out with growing goats allocated to 1 of the 3 groups (n=5-6 animals/group), with diets consisting exclusively of hay (control diet) or hay with 30 or 60% barley grain. Samples of the ventral ruminal epithelium were taken after euthanasia and instantly subjected to Ussing chamber experiments, where electrophysiological properties of the epithelium were measured in parallel with the permeability of marker molecules of different sizes [fluorescein 5(6)-isothiocyanate and horseradish peroxidase] from luminal to apical side. Additionally, ruminal fluid and blood samples were taken at the beginning of the experiment as well as shortly before euthanasia. Ruminal fluid samples were analyzed for volatile fatty acids and pH, whereas blood samples were analyzed for lipopolysaccharide, serum amyloid A, and β-hydroxybutyrate. Electrophysiological data indicated that barley feeding increased the epithelial short-circuit current compared with the control. Tissue conductance also increased with dietary barley inclusion. As shown with both marker molecules, permeability of ruminal epithelia increased with barley inclusion in the diet. Despite a lowered ruminal pH associated with increased volatile fatty acids (such as propionate and butyrate) concentrations as well as altered epithelial properties in response to high-grain feeding, no signs of inflammation became apparent, as blood serum amyloid A concentrations remained unaffected by diet. However, greater amounts of grain in the diet were associated with a quadratic increase in lipopolysaccharide concentration in the serum. Also, increasing the amounts of barley grain in the diet resulted in a tendency to quadratically augment serum concentrations of β-hydroxybutyrate and, hence, the alimentary ketogenesis. Further studies are needed to clarify the role of barley inclusion in the development of subacute ruminal acidosis in relation to ruminal epithelial damage and the translocation of toxic compounds in vivo.     Feed Restriction Reveals Distinct Serum Metabolome Profiles in Chickens Divergent in Feed Efficiency Traits

This study investigated the effect of ad libitum versus restrictive feeding (85% of ad libitum) on the serum metabolome and white blood cell composition in chickens of diverging residual feed intake (RFI; metric for feed efficiency). Blood samples were collected between days 33 and 37 post-hatch. While serum glucose was similar, serum uric acid and cholesterol were indicative of the nutritional status and chicken’s RFI, respectively. Feed restriction and RFI rank caused distinct serum metabolome profiles, whereby restrictive feeding also increased the blood lymphocyte proportion. Most importantly, 10 amino acids were associated with RFI rank in birds, whereas restrictive feeding affected almost all detected lysophosphatidylcholines, with 3 being higher and 6 being lower in restrictively compared to ad libitum fed chickens. As indicated by relevance networking, isoleucine, lysine, valine, histidine, and ornithine were the most discriminant for high RFI, whereas 3 biogenic amines (carnosine, putrescine, and spermidine) and 3 diacyl-glycerophospholipids (38:4, 38:5, and 40:5) positively correlated with feed intake and body weight gain, respectively. Only for taurine, feed intake mostly explained the RFI-associated variation, whereas for most metabolites, other host physiological factors played a greater role for the RFI-associated differences, and was potentially related to insulin-signaling, phospholipase A2, and arachidonic acid metabolism. Alterations in the hepatic synthesis of long-chain fatty acids and the need for precursors for gluconeogenesis due to varying energy demand may explain the marked differences in serum metabolite profiles in ad libitum and restrictively fed birds.This project (ECO-FCE) has received funding from the European Union’s Seventh Framework Programme for research, technological development, and demonstration under grant agreement no. 311794

    Effects of bovine colostrum on performance, survival, and immunoglobulin status of suckling piglets during the first days of life

    Supplementation of bovine colostrum (BC) has shown to improve growth performance, intestinal development, and immune response in early-weaned pigs. Little is known about whether BC may have similar effects in neonatal piglets. In the present study, the effect of BC supplementation on mortality, growth performance, and blood parameters (plasma proteins and white blood count) of suckling piglets in the first 10 days of life was investigated under practical conditions with special emphasis on low birth weight piglets. In total, 258 newborn piglets from 30 multiparous sows in a commercial breeding unit were randomly assigned to two different treatment groups. Piglets received either 1 ml of BC orally on days 1-3 of life (group BC, n = 128) or 1 ml of saline (0.9%) (control (CON) group; n = 130). Body weight was measured on days 1, 4, and 10 of life. Blood was collected on days 1 and 4 from 60 piglets per group. No differences in mortality, body weight, and average daily weight gain were observed between treatment groups in days 1-10. However, compared to CON, particularly in low birth weight piglets the administration of BC supported (P < 0.01) their survival. Group BC exhibited lower plasma total protein (P = 0.03) and beta-globulin (P = 0.02) concentrations compared to group CON. In conclusion, BC improved low and normal birth weight piglets' survival during their first 10 days of life. Further research is needed to clarify whether the survival rate is related to earlier gut closure indicated by lower plasma protein levels, which might be beneficial due to a lower uptake of potential antigenic substances
