DETERMINATION OF THE ANTINEOPLASTIC AGENT METHOTREXATE IN BODY-FLUIDS BY HIGH-PERFORMANCE LIQUID-CHROMATOGRAPHY WITH ELECTROCHEMICAL DETECTION
Some preliminary data concerning the anodic behavior of the antineplastic agent methotrexate [59-05-2] are reported, and a HPLC method with electrochem. detection for its detn. in human serum and urine is described. The method uses a reversed-phase C15 column and isocratic elution with amperometric detection on glassy carbon at +0.95 V vs. Ag/AgCl ref. electrode. Sample treatment consists of a simple deproteinization step (serum) or an extn. procedure on Sep-Pak C15 cartridges (serum) or Amberlite column (urine). The detection limit for methotrexate in serum is 2.2 nmol/L. The method should prove useful for the evaluation of the drug pharmacokinetics in patients undergoing anticancer therap
Flow injection with anodic polarographic detection for the determination of allopurinol in pharmaceutical formulations.
Data are reported concerning the anodic behavior of the antihyperuricemia agent allopurinol at a Hg electrode. A method based on flow injection with anodic polarog. detection (FI-APD) was developed for the drug assay in tablets. Polarog. detection is based on the formation of a sparingly sol. Hg compd. A high selectivity, a continuously renewable electrode surface, and no need for deoxygenation of the carrier stream, are the advantages of this approach. The FI-APD method provided an accurate and sensitive drug detn. with a detection limit of 1.8 M and a relative std. deviation of 3.1% at the 28 M level. The sample throughput was 90 samples h-7.
