The stress-responsive kinase DYRK2 activates heat shock factor 1 promoting resistance to proteotoxic stress

To survive proteotoxic stress, cancer cells activate the proteotoxic-stress response pathway, which is controlled by the transcription factor heat shock factor 1 (HSF1). This pathway supports cancer initiation, cancer progression and chemoresistance and thus is an attractive therapeutic target. As developing inhibitors against transcriptional regulators, such as HSF1 is challenging, the identification and targeting of upstream regulators of HSF1 present a tractable alternative strategy. Here we demonstrate that in triple-negative breast cancer (TNBC) cells, the dual specificity tyrosine-regulated kinase 2 (DYRK2) phosphorylates HSF1, promoting its nuclear stability and transcriptional activity. DYRK2 depletion reduces HSF1 activity and sensitises TNBC cells to proteotoxic stress. Importantly, in tumours from TNBC patients, DYRK2 levels positively correlate with active HSF1 and associates with poor prognosis, suggesting that DYRK2 could be promoting TNBC. These findings identify DYRK2 as a key modulator of the HSF1 transcriptional programme and a potential therapeutic target

Clinical relevance and biology of circulating tumor cells

    Get PDF
Most breast cancer patients die due to metastases, and the early onset of this multistep process is usually missed by current tumor staging modalities. Therefore, ultrasensitive techniques have been developed to enable the enrichment, detection, isolation and characterization of disseminated tumor cells in bone marrow and circulating tumor cells in the peripheral blood of cancer patients. There is increasing evidence that the presence of these cells is associated with an unfavorable prognosis related to metastatic progression in the bone and other organs. This review focuses on investigations regarding the biology and clinical relevance of circulating tumor cells in breast cancer

USING THE INTERNET TO SUPPORT GREEN BUSINESS FOR RURAL DEVELOPMENT AND ENVIRONMENTAL PROTECTION

Rural development constitutes the aspect for overall development of rural areas in physi- cal, technological, economic, socio-cultural and institutional levels, aiming to finally improve the quality of life of rural people. A ‘green’ business can be defined as a business that makes an effort to protect the environment, to reduce adhere environmental impact, to minimise energy use and natural resources consumption, and also to have a positive impact on the environment, to the local society and to corporate profitability as well Within green policy aspect, business begins to discover envi- ronmental initiatives, paperless policies, renewable energy solutions, innovative recycling programs, reducing policies for their waste and energy-saver technologies. The aim of this paper is to describe a model website structure, as internet training constitutes an useful tool for business owners that are environmentally concerned aiming to adapt practices to improve their profit and also to protect the environment. The proposed model website provides extended information and motivation on green policies and tactics, simple guidelines and case studies of green busines

The circular economy: A broader perspective for rural areas

The circular economy refers to a development strategy that allows economic growth by optimising the use of natural resources, minimising environmental pressures, transforming supply chains and consumption patterns and redesigning production systems so that they are restorative or regenerative by intention and design. A transition to a more circular economy requires a systemic approach that considers the interconnections that exist within and between sectors, institutions and local actors. The aim of the paper is to reflect on the characteristics of a territorial model for a circular rural economy, which directs its actions through a local lens to promote closer interaction between actors, by optimising the use of resources, and between production processes and social and cultural matrices

Wine, health and well-being sustainability: a responsible web communication strategy

The abuse of alcoholic beverages (beer, spirits and wine), also included in Agenda 2030 as one of the limits to the sustainability of well-being, results in considerable social and economic repercussions due not only to the increase in health spending but above all because of Of the loss of productivity arising in recent years especially from the youth component of human capital. Although the health benefit of a moderate consumption of wine is widely studied, the damage caused by its abuse is not so well known, and because the communication strategies implemented by the companies tend to make full use of only the positive aspect. From this point of view, starting from the health effects of wine consumption (both in case of abuse and in moderation) the paper provides the design of a web communication strategy to promote for Young people a culture of drinking in a sustainable way

Agricultural and Environmental Informatics, Governance and Management: Emerging Research Applications is a state-of-the-art reference book which explores how rural policymakers and stakeholders can use information and communication technologies to sustainably manage agricultural and natural resources. The book explores how ICT can support public governance and rural decision-making, how supply chain and agricultural informatics tools and methods can improve agricultural management, and how ICT is especially useful for environmental, resource, and ecosystems management and for geospatial landscape planning. The book integrates cross-disciplinary knowledge about agricultural and environmental applications of informatics, connecting science and policy on a subject critical to the present and future quality of life in rural area
