Suppressive activity of tiotropium bromide on matrix metalloproteinase production from lung fibroblasts in vitro

There is also increasing evidence that metalloproteinases (MMPs) may contribute to the pathogenesis of COPD, but the influence of agents that used for the treatment of COPD is not well understood.Objective: We evaluated whether tiotropium bromide hydrate (TBH), a M3 muscarinic receptor antagonist, could inhibit MMP production from lung fibroblasts (LFs) in response to tumor necrosis factor (TNF)-α stimulation.Methods: LFs were established from normal lung tissues taken from patients with lung tumors. LFs (5 × 105 cells/ml) were stimulated with TNF-α in the presence of various concentrations of TBH. After 24 h, culture supernatants were obtained and assayed for the levels of MMPs and tissue inhibitor of metalloproteinases (TIMPs) by ELISA. The influence of TBH on mRNA expression of MMPs and TIMPs in 4 h-cultured cells was also examined by real-time RT-PCR. Furthermore, nuclear factor-κB (NF-κB) and activator protein-1 (AP-1) in LFs treated with TBH for 4 h was examined by ELISA.Results: TBH at more than 15 pg/ml inhibited the production of MMP-2 from LFs after TNF-α stimulation, whereas TIMP-1 and TIMP-2 production was scarcely affected by TBH through the suppression of both mRNA expression and transcription factor, NF-κB, activation in LFs induced by TNF-α stimulation.Conclusion: These results suggest that the attenuating effect of TBH on MMP-2 production from LFs induced by inflammatory stimulation may be additional beneficial therapeutic effects not directly relating to its bronchodilatory effects.Keywords: tiotropium, lung fibroblasts, matrix metalloproteinases, suppression, in vitr
Study of hadron interactions in a lead-emulsion target
Topological and kinematical characteristics of hadron interactions have been
studied using a lead-emulsion target exposed to 2, 4 and 10 GeV/c hadron beams.
A total length of 60 m tracks was followed using a high speed automated
emulsion scanning system. A total of 318 hadron interaction vertices and their
secondary charged particle tracks were reconstructed. Measurement results of
interaction lengths, charged particle multiplicity, emission angles and momenta
of secondary charged particles are compared with a Monte Carlo simulation and
appear to be consistent. Nuclear fragments emitted from interaction vertices
were also detected by a newly developed emulsion scanning system with
wide-angle acceptance. Their emission angle distributions are in good agreement
with the simulated distributions. Probabilities of an event being associated
with at least one fragment track are found to be greater than 50% for beam
momentum GeV/c and are well reproduced by the simulation. These
experimental results validate estimation of the background due to hadron
interactions in the sample of decay candidates in the OPERA oscillation experiment.Comment: 14 pages, 11 figure
Safety and Efficacy of the Surgical Management of Hemodialysis Patients with Gastric Cancer
This retrospective study evaluated the short- and long-term outcomes after surgical management for gastric cancer in hemodialysis patients compared to non-dialysis patients. Twelve hemodialysis patients were compared with a propensity score-matched cohort of 39 gastric cancer patients who had not undergone hemodialysis. Short- and long-term outcomes along with scores estimating physiological ability and surgical stress were evaluated in both groups. The incidence of postoperative morbidity according to the Clavien-Dindo classification was higher in the hemodialysis gastric cancer group than in the non-dialysis gastric cancer group. The 5-year overall survival rate in the non-dialysis group was 69.2% after surgical resection for gastric cancer and 22.2% in the hemodialysis group. Patients with preoperative risk scores≥0.48 had significantly poorer survival outcomes compared to those with preoperative risk scores<0.48 (5-year survival rate, 83.3% vs. 39.4%, respectively). Our analyses suggest that hemodialysis patients undergoing surgery for gastric cancer have a significantly poorer postoperative prognosis and an elevated risk of postoperative complications
Multicascade-linked synthetic wavelength digital holography using an optical-comb-referenced frequency synthesizer
Digital holography (DH) is a promising method for non-contact surface
topography because the reconstructed phase image can visualize the nanometer
unevenness in a sample. However, the axial range of this method is limited to
the range of the optical wavelength due to the phase wrapping ambiguity.
Although the use of two different wavelengths of light and the resulting
synthetic wavelength, i.e., synthetic wavelength DH, can expand the axial range
up to a few tens of microns, this method is still insufficient for practical
applications. In this article, a tunable external cavity laser diode
phase-locked to an optical frequency comb, namely, an optical-comb-referenced
frequency synthesizer, is effectively used for multiple synthetic wavelengths
within the range of 32 um to 1.20 m. A multiple cascade link of the phase
images among an optical wavelength (= 1.520 um) and 5 different synthetic
wavelengths (= 32.39 um, 99.98 um, 400.0 um, 1003 um, and 4021 um) enables the
shape measurement of a reflective millimeter-sized stepped surface with the
axial resolution of 34 nm. The axial dynamic range, defined as the ratio of the
maximum axial range (= 0.60 m) to the axial resolution (= 34 nm), achieves
1.7*10^8, which is much larger than that of previous synthetic wavelength DH.
Such a wide axial dynamic range capability will further expand the application
field of DH for large objects with meter dimensions.Comment: 19 pages, 7 figure
Rock magnetism of tiny exsolved magnetite in plagioclase from a Paleoarchean granitoid in the Pilbara craton
金沢大学理工研究域地球社会基盤学系Granitoids are widespread in Precambrian terranes as well as the Phanerozoic orogenic belts, but they have garnered little attention in paleomagnetic studies, because granitoids often contain abundant coarse-grained, magnetically unstable oxides. In this study, the first example of tiny, needle-shaped, exsolved oxides in plagioclase in a Paleoarchean granitoid is reported. The magnetic properties of single plagioclase crystals with the exsolved oxide inclusions have been studied to determine their paleomagnetic recording fidelity. Demagnetization experiments and hysteresis parameters indicate that the oxide inclusions are near stoichiometric magnetite and magnetically very stable. First-order reversal curve (FORC) diagrams reveal negligible magnetostatic interactions. Minimal interactions are also reflected by very efficient acquisition of anhysteretic remanent magnetization. Single plagioclase crystals exhibit strong magnetic remanence anisotropies, which require corrections to their paleodirectional and paleointensity data. Nonetheless, quantitative consideration of anisotropy tensors of the single plagioclase crystals indicates that the bias can be mitigated by properly averaging data from a few tens of single crystals. From the nonlinear thermoremanence acquisition of the plagioclase crystals, we estimate that the plagioclase crystals can reconstruct paleointensity up to 50 μT. Local metamorphic condition suggests that those magnetite may carry remanence of ∼3.2 to 3.3 Ga. We suggest that exsolved magnetite in granitoids is potentially a suitable target for the study of the early history of the geomagnetic field, and prompt detailed microscopic investigations as well as paleomagnetic tests to constrain the age of remanence. © 2014. Brain perfusion asymmetry in patients with oral somatic delusions
Oral cenesthopathy is a somatic delusion or hallucination involving the oral area and is categorized as a delusional disorder, somatic type. The pathophysiology of this intractable condition remains obscure. In this study, we clarified the pathophysiology of oral cenesthopathy by evaluating regional brain perfusion. We performed single photon emission computed tomography (SPECT) using (99m)Tc-ethylcysteinate dimer in 16 subjects (cenesthopathy:control = 8:8). The SPECT images were visually assessed qualitatively, and quantitative analyses were also performed using a three-dimensional stereotactic region-of-interest template. The visual assessment revealed a right > left perfusion asymmetry in broad areas of the brain among the patients. The quantitative analysis confirmed that the regional cerebral blood flow values on the right side were significantly larger than those on the left side for most areas of the brain in the patients. A comparison of the R/(R + L) ratios in both groups confirmed the significant brain perfusion asymmetry between the two sides in the callosomarginal, precentral, and temporal regions in the patients. Qualitative evaluation of the SPECT images revealed right > left brain perfusion asymmetry in broad regions of the brain. Moreover, the quantitative analyses confirmed the perfusion asymmetry between the two sides in the frontal and temporal areas. Those may provide the key for elucidation of the pathophysiology of oral cenesthopathy
Late-onset benefit in progressive advanced hepatocellular carcinoma with continued sorafenib therapy: a case report
<p>Abstract</p> <p>Introduction</p> <p>In the past, no effective systemic therapy has existed for patients with advanced hepatocellular carcinoma. Sorafenib, an oral multikinase inhibitor, has recently been shown to improve overall survival in patients with advanced hepatocellular carcinoma in two randomized, double-blinded, placebo-controlled trials. This drug has been approved as the first-line therapy for advanced hepatocellular carcinoma patients. We report an intriguing case of advanced hepatocellular carcinoma in which the patient achieved late- onset partial response by prolonged administration of sorafenib in spite of progressive disease.</p> <p>Case presentation</p> <p>A 54-year-old Japanese man was treated with sorafenib for multiple lung metastases after surgical resection for advanced hepatocellular carcinoma accompanied by vascular invasion of the left branch of the portal vein. Although the effective diagnosis was progressive disease, almost all sites began to reduce or disappear eight months after the diagnosis of progressive disease. A dramatic reduction in alpha-fetoprotein and des-gamma-carboxy prothrombin levels was observed. The patient finally achieved partial response and his status remains unchanged.</p> <p>Conclusions</p> <p>If tolerated, prolonged sorafenib treatment may be beneficial.</p
