    Identification of complex biological network classes using extended correlation analysis

    Modeling and analysis of complex biological networks necessitates suitable handling of data on a parallel scale. Using the IkB-NF-kB pathway model and a basis of sensitivity analysis, analytic methods are presented, extending correlation from the network kinetic reaction rates to that of the rate reactions. Alignment of correlated processed components, vastly outperforming correlation of the data source, advanced sets of biological classes possessing similar network activities. Additional construction generated a naturally structured, cardinally based system for component-specific investigation. The computationally driven procedures are described, with results demonstrating viability as mechanisms useful for fundamental oscillatory network activity investigation

    This theoretical thesis is based on the work of French philosopher François Jullien. The thesis considers issues and challenges in existing leadership scholarship as an outcome of the Western cultural lens. Jullien’s work investigates Western and Chinese thinking traditions and recognises that the emergence of a cultural scholarship is heavily influenced by the ways the sensory world is categorised. The categorisation of reality on the basis of ‘being’ influences aspects of the sensory world a scholar is attentive to and created conditions for the emergence of Western scholarship. The Chinese ideographical language categorised the world on the basis of motion and produced a scholarship that is attentive to silent motions in the sensory world and not identifiable “being” and studies the propensity of things and not identity. By taking a Chinese perspective to reinvestigate Western thinking and vice versa, Jullien’s work makes a contribution by uncovering how separate cultural traditions contribute to each other by revealing insights that are unavailable from only one cultural scholarship (Jullien, 2014, 2015). Jullien calls the knowledge that emerges from between cultural thoughts unthought. This thesis aims to address the question of How can François Jullien’s work contribute to contemporary leadership studies? Following Jullien’s approach, I investigate leadership through a Chinese lens provided by Jullien’s work and uncover unthought in existing leadership scholarship by revealing insights about leadership from a Chinese perspective. This insight adds to leadership knowledge and provides alternative ways of approaching leadership through silent tendencies behind the emergence of identifiable aspects of leadership

    Are Consumers Willing to Pay More for Biodegradable Containers Than for Plastic Ones? Evidence from Hypothetical Conjoint Analysis and Nonhypothetical Experimental Auctions

    This study used and compared hypothetical conjoint analysis and nonhypothetical experimental auctions to elicit floral customers’ willingness to pay for biodegradable plant containers. The results of the study show that participants were willing to pay a price premium for biodegradable containers, but the premium is not the same for different types of containers.     Oxidative Stress Accumulates in Adipose Tissue during Aging and Inhibits Adipogenesis

    Aging constitutes a major independent risk factor for the development of type 2 diabetes and is accompanied by insulin resistance and adipose tissue dysfunction. One of the most important factors implicitly linked to aging and age-related chronic diseases is the accumulation of oxidative stress. However, the effect of increased oxidative stress on adipose tissue biology remains elusive. In this study, we demonstrate that aging in mice results in a loss of fat mass and the accumulation of oxidative stress in adipose tissue. In vitro, increased oxidative stress through glutathione depletion inhibits preadipocyte differentiation. This inhibition of adipogenesis is at least in part the result of reduced cell proliferation and an inhibition of G1→S-phase transition during the initial mitotic clonal expansion of the adipocyte differentiation process. While phosphorylation of the retinoblastoma protein (Rb) by cyclin/cdk complexes remains unaffected, oxidative stress decreases the expression of S-phase genes downstream of Rb. This silencing of S phase gene expression by increased oxidative stress is mediated through a transcriptional mechanism involving the inhibition of E2F recruitment and transactivation of its target promoters. Collectively, these data demonstrate a previously unrecognized role of oxidative stress in the regulation of adipogenesis which may contribute to age-associated adipose tissue dysfunction

    Osteopontin Mediates Obesity-Induced Adipose Tissue Macrophage Infiltration and Insulin Resistance in Mice

    Obesity is associated with a state of chronic, low-grade inflammation characterized by abnormal cytokine production and macrophage infiltration into adipose tissue, which may contribute to the development of insulin resistance. During immune responses, tissue infiltration by macrophages is dependent on the expression of osteopontin, an extracellular matrix protein and proinflammatory cytokine that promotes monocyte chemotaxis and cell motility. In the present study, we used a murine model of diet-induced obesity to examine the role of osteopontin in the accumulation of adipose tissue macrophages and the development of insulin resistance during obesity. Mice exposed to a high-fat diet exhibited increased plasma osteopontin levels, with elevated expression in macrophages recruited into adipose tissue. Obese mice lacking osteopontin displayed improved insulin sensitivity in the absence of an effect on diet-induced obesity, body composition, or energy expenditure. These mice further demonstrated decreased macrophage infiltration into adipose tissue, which may reflect both impaired macrophage motility and attenuated monocyte recruitment by stromal vascular cells. Finally, obese osteopontin-deficient mice exhibited decreased markers of inflammation, both in adipose tissue and systemically. Taken together, these results suggest that osteopontin may play a key role in linking obesity to the development of insulin resistance by promoting inflammation and the accumulation of macrophages in adipose tissue

    In-depth mesocrystal formation analysis of microwave-assisted synthesis of LiMnPO4nanostructures in organic solution

    In the present work, we report on the preparation of LiMnPO4 (lithiophilite) nanorods and mesocrystals composed of self-assembled rod subunits employing microwave-assisted precipitation with processing times on the time scale of minutes. Starting from metal salt precursors and H3PO4 as phosphate source, single-phase LiMnPO4 powders with grain sizes of approx. 35 and 65 nm with varying morphologies were obtained by tailoring the synthesis conditions using rac-1-phenylethanol as solvent. The mesocrystal formation, microstructure and phase composition were determined by electron microscopy, nitrogen physisorption, X-ray diffraction (including Rietveld refinement), dynamic light scattering, X-ray absorption and X-ray photoelectron spectroscopy, and other techniques. In addition, we investigated the formed organic matter by gas chromatography coupled with mass spectrometry in order to gain a deeper understanding of the dissolution\u2013precipitation process. Also, we demonstrate that the obtained LiMnPO4 nanocrystals can be redispersed in polar solvents such as ethanol and dimethylformamide and are suitable as building blocks for the fabrication of nanofibers via electrospinning

    Complete Strain Mapping of Nanosheets of Tantalum Disulfide

    Quasi-two-dimensional (quasi-2D) materials hold promise for future electronics because of their unique band structures that result in electronic and mechanical properties sensitive to crystal strains in all three dimensions. Quantifying crystal strain is a prerequisite to correlating it with the performance of the device, and calls for high resolution but spatially resolved rapid characterization methods. Here we show that using fly-scan nano X-ray diffraction we can accomplish a tensile strain sensitivity below 0.001% with a spatial resolution of better than 80 nm over a spatial extent of 100 μ\mum on quasi 2D flakes of 1T-TaS2. Coherent diffraction patterns were collected from a \sim 100 nm thick sheet of 1T-TaS2 by scanning 12keV focused X-ray beam across and rotating the sample. We demonstrate that the strain distribution around micron and sub-micron sized 'bubbles' that are present in the sample may be reconstructed from these images. The experiments use state of the art synchrotron instrumentation, and will allow rapid and non-intrusive strain mapping of thin film samples and electronic devices based on quasi 2D materials
