    Acetylome analyses provide novel insights into the effects of chronic intermittent hypoxia on hippocampus-dependent cognitive impairment

    IntroductionChronic intermittent hypoxia (CIH) can negatively affect hippocampal function through various molecular mechanisms. Protein acetylation, a frequently occurring modification, plays crucial roles in synaptic plasticity and cognitive processes. However, the global protein acetylation induced by CIH in the hippocampus and its specific effects on hippocampal function and behavior remain poorly understood.MethodsTo address this gap, we conducted a study using liquid chromatography-tandem mass spectrometry to analyze the lysine acetylome and proteome of the hippocampus in healthy adult mice exposed to intermittent hypoxia for 4 weeks (as a CIH model) compared to normoxic mice (as a control).ResultsWe identified and quantified a total of 2,184 lysine acetylation sites in 1,007 proteins. Analysis of these acetylated proteins revealed disturbances primarily in oxidative phosphorylation, the tricarboxylic acid (TCA) cycle, and glycolysis, all of which are localized exclusively to mitochondria. Additionally, we observed significant changes in the abundance of 21 proteins, some of which are known to be associated with cognitive impairments.DiscussionThis study helps to elucidate the molecular mechanisms underlying CIH-induced changes in protein acetylation in the hippocampus. By providing valuable insights into the pathophysiological processes associated with CIH and their impacts on hippocampal function, our findings contribute to a better understanding of the consequences of CIH-induced changes in protein acetylation in the hippocampus and the potential role of CIH in cognitive impairment

    Protective role of curcumin in disease progression from non-alcoholic fatty liver disease to hepatocellular carcinoma: a meta-analysis

    Background: Pathological progression from non-alcoholic fatty liver disease (NAFLD) to liver fibrosis (LF) to hepatocellular carcinoma (HCC) is a common dynamic state in many patients. Curcumin, a dietary supplement derived from the turmeric family, is expected to specifically inhibit the development of this progression. However, there is a lack of convincing evidence.Methods: The studies published until June 2023 were searched in PubMed, Web of Science, Embase, and the Cochrane Library databases. The SYstematic Review Center for Laboratory animal Experimentation (SYRCLE) approach was used to evaluate the certainty of evidence. StataSE (version 15.1) and Origin 2021 software programs were used to analyze the critical indicators.Results: Fifty-two studies involving 792 animals were included, and three disease models were reported. Curcumin demonstrates a significant improvement in key indicators across the stages of NAFLD, liver fibrosis, and HCC. We conducted a detailed analysis of common inflammatory markers IL-1β, IL-6, and TNF-α, which traverse the entire disease process. The research results reveal that curcumin effectively hinders disease progression at each stage by suppressing inflammation. Curcumin exerted hepatoprotective effects in the dose range from 100 to 400 mg/kg and treatment duration from 4 to 10 weeks. The mechanistic analysis reveals that curcumin primarily exerts its hepatoprotective effects by modulating multiple signaling pathways, including TLR4/NF-κB, Keap1/Nrf2, Bax/Bcl-2/Caspase 3, and TGF-β/Smad3.Conclusion: In summary, curcumin has shown promising therapeutic effects during the overall progression of NAFLD–LF–HCC. It inhibited the pathological progression by synergistic mechanisms related to multiple pathways, including anti-inflammatory, antioxidant, and apoptosis regulation

    Folate-deficiency induced acyl-CoA synthetase short-chain family member 2 increases lysine crotonylome involved in neural tube defects

    Maternal folate deficiency increases the risk of neural tube defects (NTDs), but the mechanism remains unclear. Here, we established a mouse model of NTDs via low folate diets combined with MTX-induced conditions. We found that a significant increase in butyrate acid was observed in mouse NTDs brains. In addition, aberrant key crotonyl-CoA-producing enzymes acyl-CoA synthetase short-chain family member 2 (ACSS2) levels and lysine crotonylation (Kcr) were elevated high in corresponding low folate content maternal serum samples from mouse NTD model. Next, proteomic analysis revealed that folate deficiency led to global proteomic modulation, especially in key crotonyl-CoA-producing enzymes, and dramatic ultrastructural changes in mouse embryonic stem cells (mESCs). Furthermore, we determined that folate deficiency induced ACSS2 and Kcr in mESCs. Surprisingly, folic acid supplementation restored level of ACSS2 and Kcr. We also investigated overall protein post-translational Kcr under folate deficiency, revealing the key regulation of Kcr in glycolysis/gluconeogenesis, and the citric acid cycle. Our findings suggest folate deficiency leads to the occurrence of NTDs by altering ACSS2. Protein crotonylation may be the molecular basis for NTDs remodeling by folate deficiency

    Effects of Community Perceptions and Institutional Capacity on Smallholder Farmers’ Responses to Water Scarcity: Evidence from Arid Northwestern China

    Community contextual factors including community perceptions and institutional capacity are among the key determinants in community-based water resource management. The Institutional Analysis and Development (IAD) framework proposed by Ostrom is commonly employed to examine the outcome of common-pool resource management including water resources. However, community perceptions typically examined in behavioral economics and comparative community analysis literature are rarely incorporated in institutional analysis studies. This study draws on the IAD framework to investigate smallholder farmer communities’ responses to water scarcity in arid northwestern China. Adopting alternating multiple regression and multivariate regression models, this study conducts an empirical analysis using farmer survey data. The results show that the perceptions of water scarcity promote community actions in coping with water shortage. The perception of production risks encourages overall community responses, as well as farming- and irrigation-related responses. Communities with a stronger institutional enforcement are more responsive in taking farming-, irrigation-, and infrastructure-related actions, as well as having better overall responses. The analysis also shows that community interactional capacities and socio-economic factors may influence community actions to mitigate and adapt to adverse effects of local water scarcity. Our findings provide insights for understanding social and institutional aspects of rural farming communities toward sustainable response decisions to overcome water scarcity challenges

    Hormonal Regulatory Patterns of LaKNOXs and LaBEL1 Transcription Factors Reveal Their Potential Role in Stem Bulblet Formation in LA Hybrid Lily

    In lily reproduction, the mechanism of formation of bulbs has been a hot topic. However, studies on stem bulblet formation are limited. Stem bulblets, formed in the leaf axils of under- and above-ground stems, provide lilies with a strong capacity for self-propagation. First, we showed that above-ground stem bulblets can be induced by spraying 100 mg/L 6-BA on the LA hybrid lily ‘Aladdin’, with reduced endogenous IAA and GA4 and a higher relative content of cytokinins. Then, expression patterns of three potential genes (two KNOTTED1-like homeobox (KNOX) and one partial BEL1-like homeobox (BELL)), during stem bulblet formation from our previous study, were determined by RT-qPCR, presenting a down-up trend in KNOXs and a rising tendency in BELL. The partial BELL gene was cloned by RACE from L. ‘Aladdin’ and denoted LaBEL1. Physical interactions of LaKNOX1-LaBEL1 and LaKNOX1-LaKNOX2 were confirmed by yeast two-hybrid and bimolecular fluorescence complementation assays. Furthermore, hormonal regulatory patterns of single LaKNOX1, LaKNOX2, LaBEL1, and their heterodimers, were revealed in transgenic Arabidopsis, suggesting that the massive mRNA accumulations of LaKNOX1, LaKNOX2 and LaBEL1 genes during stem bulblet formation could cause the dramatic relative increase of cytokinins and the decline of GAs and IAA. Taken together, a putative model was proposed that LaKNOX1 interacts with LaKNOX2 and LaBEL1 to regulate multiple phytohormones simultaneously for an appropriate hormonal homeostasis, which suggests their potential role in stem bulblet formation in L. ‘Aladdin’

    Is East Asia decoupled from the US?

    Since the Asian Financial Crisis, there have been a lot of restructurings and reforms implemented in the East Asian countries. Thanks to all those changes, East Asia is now more dynamic and resilient than ever. As a result, there were many arguments that East Asia would start to decouple from the United States as the countries in the region become more and more dependent on the emerging markets of China and Korea, and as demand became more robust, made possible by the economic boom in the region. The issue of East Asia’s decoupling from the US arose once again in the recent financial crisis, from which East Asian countries have recovered quickly as compared with a slowing US economy.This project aims to look through the increasingly popular hypothesis that East Asian economies are "decoupling" from the United States. We will analyse both supportive and opposing viewpoints revolving around this hypothesis qualitatively. In addition, an empirical econometric model which depicts the relationship between East Asian exports and the US economy will be exploited to determine whether the “decoupling” process is under way or not.Bachelor of Art

    Integrative evidence construction for resveratrol treatment of nonalcoholic fatty liver disease: preclinical and clinical meta-analyses

    Background: Resveratrol, a polyphenol found in various plants, is known for its diverse bioactivities and has been explored in relation to nonalcoholic fatty liver disease (NAFLD). However, no high-quality evidence exists regarding its efficacy.Objective: a meta-analysis was conducted to evaluate the potential efficacy of resveratrol in treating nonalcoholic fatty liver disease by analyzing both preclinical studies and clinical trials.Method: PubMed, Embase and Web of Science were searched for the included literature with the criteria for screening. Quantitative synthesis and meta-analyses were performed by STATA 16.0.Results: Twenty-seven studies were included, and the results indicated that resveratrol effectively improved liver function, reduced fatty liver indicators, and affected other indices in preclinical studies. The effective dosage ranged from 50 mg/kg-200 mg/kg, administered over a period of 4–8 weeks. While there were inconsistencies between clinical trials and preclinical research, both study types revealed that resveratrol significantly reduced tumor necrosis factor-α levels, further supporting its protective effect against nonalcoholic fatty liver disease. Additionally, resveratrol alleviated nonalcoholic fatty liver disease primarily via AMPK/Sirt1 and anti-inflammatory signaling pathways.Conclusion: Current meta-analysis could not consistently verify the efficacy of resveratrol in treating nonalcoholic fatty liver disease, but demonstrated the liver-protective effects on nonalcoholic fatty liver disease.     Cancelable biometric schemes for Euclidean metric and Cosine metric

    Abstract The handy biometric data is a double-edged sword, paving the way of the prosperity of biometric authentication systems but bringing the personal privacy concern. To alleviate the concern, various biometric template protection schemes are proposed to protect the biometric template from information leakage. The preponderance of existing proposals is based on Hamming metric, which ignores the fact that predominantly deployed biometric recognition systems (e.g. face, voice, gait) generate real-valued templates, more applicable to Euclidean metric and Cosine metric. Moreover, since the emergence of similarity-based attacks, those schemes are not secure under a stolen-token setting. In this paper, we propose a succinct biometric template protection scheme to address such a challenge. The proposed scheme is designed for Euclidean metric and Cosine metric instead of Hamming distance. Mainly, the succinct biometric template protection scheme consists of distance-preserving, one-way, and obfuscation modules. To be specific, we adopt location sensitive hash function to realize the distance-preserving and one-way properties simultaneously and use the modulo operation to implement many-to-one mapping. We also thoroughly analyze the proposed scheme in three aspects: irreversibility, unlinkability and revocability. Moreover, comprehensive experiments are conducted on publicly known face databases. All the results show the effectiveness of the proposed scheme

    Grassland Transfer and Its Income Effect: Evidence from Pastoral Areas of the Qinghai–Tibet Plateau

    Under the condition of different endowment factors of herders and imperfect market mechanisms, grassland transfer can promote the redistribution and rational utilization of grassland resources, which has a crucial impact on herders’ livelihood. This study fully examined how the grassland rental market improves herders’ income and to what extent, using unbalanced panel data with 560 herder households in the Qinghai–Tibet Plateau pastoral areas. A fixed effect model was used as the baseline model. The instrumental variable approach and propensity score matching method were utilized to address the endogeneity problem and sample selection bias. Finally, the mediating effect model was used to analyze the path mechanism of grassland transfer in/out on herder income. The results showed that the total household income increased significantly after participating in grassland transfer. Grassland rent-in increased livestock income, and grassland rent-out increased non-livestock income. After correcting for the selection bias, the income effect of grassland transfer became larger. The grassland rent-in increased the livestock income by promoting production investment. Grassland rent-out increased the non-livestock income by promoting non-pastoral employment
