Use of a novel FĂśrster resonance energy transfer method to identify locations of site-bound metal ions in the U2âU6 snRNA complex
U2 and U6 snRNAs pair to form a phylogenetically conserved complex at the catalytic core of the spliceosome. Interactions with divalent metal ions, particularly Mg(II), at specific sites are essential for its folding and catalytic activity. We used a novel FĂśrster resonance energy transfer (FRET) method between site-bound luminescent lanthanide ions and a covalently attached fluorescent dye, combined with supporting stoichiometric and mutational studies, to determine locations of site-bound Tb(III) within the human U2âU6 complex. At pH 7.2, we detected three metal-ion-binding sites in: (1) the consensus ACACAGA sequence, which forms the internal loop between helices I and III; (2) the four-way junction, which contains the conserved AGC triad; and (3) the internal loop of the U6 intra-molecular stem loop (ISL). Binding at each of these sites is supported by previous phosphorothioate substitution studies and, in the case of the ISL site, by NMR. Binding of Tb(III) at the four-way junction and the ISL sites was found to be pH-dependent, with no ion binding observed below pH 6 and 7, respectively. This pH dependence of metal ion binding suggests that the local environment may play a role in the binding of metal ions, which may impact on splicing activity

Adiponectin and Leptin Metabolic Biomarkers in Chinese Children and Adolescents
Objective. To evaluate leptin and adiponectin as biomarkers of metabolic syndrome (MS) risk factors even in nonobese children/adolescents. Methods. Serum leptin, adiponectin, leptin:adiponectin ratio, lipids, glucose, and insulin concentrations as well as body size parameters and pubertal development were evaluated in a large population of Chinese children/adolescents (n = 3505, 6â18 years, 1722 girls and 1783 boys). Results. Leptin concentration increased while adiponectin decreased with obesity, both were influenced by pubertal development. Central obesity had an additive effect on leptin levels (above obesity alone). Leptin/adiponectin increased 8.4-fold and 3.2-fold in overweight/obesity, and 15.8- and 4.5-fold with obesity plus MS, in early and late puberty, respectively. Even in normal weight children/adolescents, higher leptin and lower adiponectin concentrations associated with increased risk profile. Conversely, overweight/obese with lower leptin or higher adiponectin concentrations had a less compromised metabolic profile. Conclusion. Leptin, adiponectin, and leptin:adiponectin ratio are informative biomarkers for obesity, central obesity, MS, and abnormal metabolic profile even in normal weight children/adolescents
Evaluation of an Adjustable Epidemiologic Information System
BACKGROUND: In order to facilitate public health response and to achieve early control of infectious disease epidemics, an adjustable epidemiologic information system (AEIS) was established in the Taiwan public health network in February 2006. METHODOLOGY/PRINCIPAL FINDINGS: The performance of AEIS for the period 2006 through 2008 was evaluated based on a number of response times (RT) and the public health impact. After implementation of the system, the apparent overall shortened RT was mainly due to the shortening of personnel response time (PRT) and the time needed to draft a new questionnaire that incurred as personnel-system interface (PSI); PRT dropped from a fluctuating range of 9.8 âź28.8 days in the first four months to <10 days in the following months and remained low till 2008 (0.88Âą1.52 days). The PSIs for newly emerged infectious diseases were 2.6 and 3.4 person-hours for H5N1 in 2007 and chikungunya in 2008, respectively, a much improvement from 1142.5 person-hours for SARS in 2003. The duration of each rubella epidemic cluster was evaluated as public health impact and showed a shortening trend (pâ=â0.019) that concurred with the shortening of PRT from 64.8Âą47.3 to 25.2Âą38.2 hours per cluster (p<0.0001). CONCLUSIONS/SIGNIFICANCE: The first evaluation of the novel instrument AEIS that had been used to assist Taiwan's multi-level government for infectious diseases control demonstrated that it was well integrated into the existing public health infrastructure. It provided flexible tools and computer algorithms with friendly interface for timely data collection, integration, and analysis; as a result, it shortened RTs, filled in gaps of personnel lacking sufficient experiences, created a more efficient flow of response, and identified asymptomatic/mild cases early to minimize further spreading. With further development, AEIS is anticipated to be useful in the application of other acute public health events needing immediate orchestrated data collection and public health actions
SN 2008inâBridging the Gap between Normal and Faint Supernovae of Type IIP
We present optical photometric and low-resolution spectroscopic observations of the Type II plateau supernova (SN) 2008in, which occurred in the outskirts of the nearly face-on spiral galaxy M61. Photometric data in the X-ray, ultraviolet, and near-infrared bands have been used to characterize this event. The SN field was imaged with the ROTSE-IIIb optical telescope about seven days before the explosion. This allowed us to constrain the epoch of the shock breakout to JD = 2454825.6. The duration of the plateau phase, as derived from the photometric monitoring, was ~98 days. The spectra of SN 2008in show a striking resemblance to those of the archetypal low-luminosity IIP SNe 1997D and 1999br. A comparison of ejecta kinematics of SN 2008in with the hydrodynamical simulations of Type IIP SNe by Dessart et al. indicates that it is a less energetic event (~5 Ă 10^(50) erg). However, the light curve indicates that the production of radioactive ^(56)Ni is significantly higher than that in the low-luminosity SNe. Adopting an interstellar absorption along the SN direction of AV ~ 0.3 mag and a distance of 13.2 Mpc, we estimated a synthesized ^(56)Ni mass of ~0.015 M_â. Employing semi-analytical formulae derived by Litvinova and Nadezhin, we derived a pre-SN radius of ~126 R_â, an explosion energy of ~5.4 Ă 10^(50) erg, and a total ejected mass of ~16.7 M_â. The latter indicates that the zero-age main-sequence mass of the progenitor did not exceed 20 M_â. Considering the above properties of SN 2008in and its occurrence in a region of sub-solar metallicity ([O/H] ~ 8.44 dex), it is unlikely that fall-back of the ejecta onto a newly formed black hole occurred in SN 2008in. We therefore favor a low-energy explosion scenario of a relatively compact, moderate-mass progenitor star that generates a neutron star
Differences in Inflammatory Markers between Nulliparous Women Admitted to Hospitals in Preactive vs Active Labor
Objective To determine whether labor-associated inflammatory markers differ between low-risk, nulliparous women in preactive vs active labor at hospital admission and over time. Study Design Prospective comparative study of low-risk, nulliparous women with spontaneous labor onset at term (n = 118) sampled from 2 large Midwestern hospitals. Circulating concentrations of inflammatory markers were measured at admission and again 2 and 4 hours later: namely, neutrophil, and monocyte counts; and serum inflammatory cytokines (interleukin -1β, interleukin-6, tumor necrosis factor-ι, interleukin-10) and chemokines (interleukin-8). Biomarker concentrations and their patterns of change over time were compared between preactive (n = 63) and active (n = 55) labor admission groups using Mann-Whitney U tests. Results Concentrations of interleukin-6 and interleukin-10 in the active labor admission group were significantly higher than concentrations in the preactive labor admission group at all 3 time points. Neutrophil levels were significantly higher in the active group at 2 and 4 hours after admission. The rate of increase in neutrophils and interleukin-10 between admission and 2 hours later was faster in the active group (P \u3c .001 and P = .003, respectively). Conclusion Circulating concentrations of several inflammatory biomarkers are higher and their rate of change over time since admission is faster among low-risk, nulliparous women admitted to hospitals in active labor, as compared with those admitted in preactive labor. More research is needed to determine if progressive changes in inflammatory biomarkers might be a useful adjunct to improving the assessment of labor progression and determining the optimal timing of labor admission

The Effects of Perinatal Testosterone Exposure on the DNA Methylome of the Mouse Brain Are Late-Emerging
Background
The biological basis for sex differences in brain function and disease susceptibility is poorly understood. Examining the role of gonadal hormones in brain sexual differentiation may provide important information about sex differences in neural health and development. Permanent masculinization of brain structure, function, and disease is induced by testosterone prenatally in males, but the possible mediation of these effects by long-term changes in the epigenome is poorly understood. Methods
We investigated the organizational effects of testosterone on the DNA methylome and transcriptome in two sexually dimorphic forebrain regionsâthe bed nucleus of the stria terminalis/preoptic area and the striatum. To study the contribution of testosterone to both the establishment and persistence of sex differences in DNA methylation, we performed genome-wide surveys in male, female, and female mice given testosterone on the day of birth. Methylation was assessed during the perinatal window for testosterone\u27s organizational effects and in adulthood. Results
The short-term effect of testosterone exposure was relatively modest. However, in adult animals the number of genes whose methylation was altered had increased by 20-fold. Furthermore, we found that in adulthood, methylation at a substantial number of sexually dimorphic CpG sites was masculinized in response to neonatal testosterone exposure. Consistent with this, testosterone\u27s effect on gene expression in the striatum was more apparent in adulthood. Conclusion
Taken together, our data imply that the organizational effects of testosterone on the brain methylome and transcriptome are dramatic and late-emerging. Our findings offer important insights into the long-term molecular effects of early-life hormonal exposure
Measuring nickel masses in Type Ia supernovae using cobalt emission in nebular phase spectra
The light curves of Type Ia supernovae (SNe Ia) are powered by the
radioactive decay of Ni to Co at early times, and the decay of
Co to Fe from ~60 days after explosion. We examine the evolution
of the [Co III] 5892 A emission complex during the nebular phase for SNe Ia
with multiple nebular spectra and show that the line flux follows the square of
the mass of Co as a function of time. This result indicates both
efficient local energy deposition from positrons produced in Co decay,
and long-term stability of the ionization state of the nebula. We compile 77
nebular spectra of 25 SN Ia from the literature and present 17 new nebular
spectra of 7 SNe Ia, including SN2014J. From these we measure the flux in the
[Co III] 5892 A line and remove its well-behaved time dependence to infer the
initial mass of Ni () produced in the explosion. We then examine
Ni yields for different SN Ia ejected masses ( - calculated
using the relation between light curve width and ejected mass) and find the
Ni masses of SNe Ia fall into two regimes: for narrow light curves (low
stretch s~0.7-0.9), is clustered near ~ 0.4 and
shows a shallow increase as increases from ~1-1.4; at high
stretch, clusters at the Chandrasekhar mass (1.4) while
spans a broad range from 0.6-1.2. This could constitute
