    EST sequencing and gene expression profiling in Scutellaria baicalensis

    Scutellaria baicalensis is an important medicinal plant, but few genomic resources are available for this species, as well as for other non-model plants. One of the major new directions in genome research is to discover the full spectrum of genes transcribed from the whole genome. Here, we report extensive transcriptome data of the early growth stage of S. baicalensis. This transcriptome consensus sequence was constructed by de novo assembly of shotgun sequencing data, obtained using multiple next-generation DNA sequencing (NGS) platforms (Roche/454 GS_FLX+ and Illumina/Solexa HiSeq2000). We show that this new approach to obtain extensive mRNA is an efficient strategy for genome-wide transcriptome analysis. We obtained 1,226,938 and 161,417,646 reads using the GS_FLX and the Illumina/Solexa HiS-eq2000, respectively. De novo assembly of the high-quality GS_FLX and Illumina reads (95 % and 75 %) resulted in more than 82 Mb of mRNA consensus sequence, which we assembled into 51,188 contigs, with at least 500 bp per contig. Of these contigs, 39,581 contained known genes, as determined by BLASTX searches against non-redundant NCBI database. Of these, 20,498 different genes were expressed during the early growth stage of S. baicalensis. We have made the expressed sequences available on a public database. Our results demonstrate the utility of combining NGS technologies as a basis for the development of genomic tools in non-model, medicinal plant species. Knowledge of all described genes and quantitation of the expressed genes, including the transcription factors involved, will be useful in studies of the biology of S. baicalensis gene regulation

    Effect of media and gelling agents on shoot organogenesis of Liriope platyphylla

    Liriope platyphylla can be multiplied either by planting seeds or dividing its tuberous roots. In this study, a method for L. platyphylla plant shoot organogenesis from meristem explants was developed employing medium and gelling agents. For the selection of optimal medium conditions for shoot organogenesis, the effects of full- and half-strength B5, SH, and MS media were examined. Different concentrations of the gelling agents such as phytagar (6, 7, 8, and 9 g L-1) and gellan gum (2, 3, 4, and 5 g L-1) were examined for efficient shoot formation. The results revealed the superiority of half-strength MS basal medium in shoot organogenesis and growth of Liriope platyphylla. But the half-strength B5 media performed poorly. Compared to plant agar, gellan gum performed well in terms of shoot regeneration and shoot length. When gellan gum was used at 3 g L-1 the maximum number of shoots explant-1 (5.8) and longest shoot (45.8 mm) was observed but the lowest number of shoots explant-1 (3.2) and shortest shoot (21.4 mm) was registered with 5 g L-1. It is proposed from our study that half-strength MS media and gellan gun gelling agent at 3 g/l could be applied in shoot organogenesis and growth of L. platyphylla

    Role of COVID-19 risk perception in predicting the intention to participate in exercise and health behaviors among Korean men

    The coronavirus disease 2019 (COVID-19) pandemic has prompted the implementation of social distancing policies worldwide, limiting participation in exercise and substantially impacting health behaviors. In accordance with the theory of planned behavior (TPB), the present study aimed to develop a model for predicting the intent to participate in exercise and engage in health behaviors among Korean men using the perception of COVID-19 risk as an exogenous variable. We analyzed data obtained from 374 Korean men who had completed a 32-item, online questionnaire. Structural equation modeling was performed to evaluate the effect of attitudes, subjective norms, and perceived behavioral control (PBC) on the intention to participate in exercise and health behaviors using COVID-19 risk perception as an antecedent variable. COVID-19 risk perception exerted significant negative effects on the attitude toward exercise participation (β = −0.857, p < 0.001), subjective norms associated with exercise participation (β = −0.862, p < 0.001), and PBC related to exercise (β = −0.738, p < 0.001). In addition, both attitude (β = 0.213, p < 0.001) and subjective norms (β = 0.168, p = 0.001) exerted significant effects on the intention to participate in exercise. PBC also exerted significant effects on the intention to participate in exercise (β = 0.580, p < 0.001) and health behaviors (β = 0.461, p < 0.001). Lastly, the intention to participate in exercise exerted a significant effect on health behaviors (β = 0.400, p < 0.001). The data indicated that, among TPB variables, PBC exerted the greatest influence on the intention to participate in exercise and had a significant effect on engagement in health behaviors. The current findings support TPB as an important theoretical model for predicting the intention to participate in exercise and patterns of health behavior among Korean men during the COVID-19 pandemic. Our study also highlights the importance of addressing PBC when designing interventions to promote exercise participation and health behaviors among Korean men

    E-p-methoxycinnamic acid production in hairy root cultures of Scrophularia buergeriana Miquel

    E-p-methoxycinnamic acid (MCA) is one of the main active constituents of Scrophularia buergeriana Miquel and has hepatoprotective, anti-amnestic, and neuroprotective activities. For studying in vitro production of MCA, we established a hairy root culture of S. buergeriana by infecting leaf explants with Agrobacterium rhizogenes R1000, and tested the growth and MCA production of these cultures using different strengths of media and concentrations of auxins. Hairy roots grown in half-strength MS medium showed the highest levels of growth (10.3 g/l) as well as MCA production (0.83 mg/100 g dry weight). Hairy root culture with the treatment of 0.5 mg/l IBA produced the highest amount of dry weight (11.8 g/l) and MCA (1.26 mg/100 g dry weight) production. These results demonstrate that the hairy root culture of S. buergeriana is a valuable alternative approach for the production of MCA.

    Analysis of volatile compounds from three species of Atractylodes by Gas Chromatography-Mass Spectrometry

    A total of 99 different volatile compounds were detected through Gas Chromatography-Mass Spectrometry (GC-MS) from three species of Atractylodes, namely Atractylodes lancea, Atractylodes japonica, and Atractylodes chinensis. Thirteen-volatile flavor compounds i.e., acid, alcohol, aldehyde, alkane, alkene, alkyne, ester, ketone, monoterpene, oxygenated monoterpene, sesquiterpene, oxygenated sesquiterpene, and oxygenated triterpenoid detected from different species of Atractylodes. It was observed that all the species contained 38 common compounds, while A. lancea contained 7 unique compounds, A. japonica has 4 unique compounds, and A. chinensis hold 6 compounds not detected in the other extracts. In addition, essential oils from A. lancea and A. japonica possessed 11 compounds in common, and A. lancea and A. chinensis possessed 19 compounds in common. The remaining 14 compounds were detected only in A. japonica and A. chinensis. The total content of all components in the species was comparable, with 82.528%, 81.766%, and 81.799% of volatile components being detected for A. lancea, A. japonica, and A. chinensis, respectively. Curzerene was found to be the most predominant compound in both A. lancea (14.1%) and A. chinensis (16.7%), while murolan-3,9(11)-diene-10-peroxy was found predominantly in A. japonica (16.8%). The present study suggests that the identified volatile compounds may possess important biological properties, and could be suitable for application in both oriental medicines and the pharmaceutical industry

    Floristic study of Cheondeungsan Mountain in Korea

    AbstractThe distribution of native plants of Cheondeungsan Mountain (807 m, N 37°05'00“–37°05'30”, E 128°00'0“–128°02'0”) in Chungcheongbuk-do was determined and the major flora were identified. During field investigations carried out from May 2011 to October 2011, 87 families, 254 genera, and 369 taxonomic groups (327 species, 4 subspecies, 33 varieties, and 5 forms) were confirmed, and the distribution of 219 taxonomic groups was discovered for the first time. The distribution of four endemic plants of Korea, including Ajuga spectabilis Nakai and Salvia chanryoenica Nakai, and that of Penthorum chinense Pursh, a Grade V specific plant species, was found. There were 20 taxa of naturalized plants at Cheondeungsan; the growth and development of plants that are harmful to the ecosystem, such as Ambrosia artemisiifolia L., Ambrosia trifida L., Eupatorium rugosum Houtt., and Aster pilosus Willd., was observed around the forest paths and lowlands

    Serotonin-Norepinephrine Reuptake Inhibitors for Pain Control: Premise and Promise

    The precise mechanisms of pain perception and transmission in the central nervous system have not been fully elucidated. However, extensive data support a role for the monoamine neurotransmitters, serotonin and norepinephrine, in the modulation of pain. Experiments with animal models of pain indicate that noradrenergic interventions, and to a lesser extent serotonergic interventions, reduce pain-related behavior. This is supported by data from clinical trials in humans in which antidepressants have been shown to reduce pain and functional impairment in central and neuropathic pain conditions. These effects are particularly well-studied in trials with serotonin-norepinephrine reuptake inhibitors (SNRIs), which have provided a useful tool in the clinician’s arsenal, particularly considering the limitations of other classes of pain medications such as opioids, anti-inflammatories, and anticonvulsants (i.e., limited efficacy, safety and tolerability issues). Moreover, painful physical symptoms are frequently comorbid with major psychiatric disorders such as major depressive disorder and anxiety disorders. This paper reviewed and summarized the rationale and potential role of SNRIs for the control of pain including clinical and preclinical background. Currently evidence does not definitely support a role of the SNRIs, while limited data propose a putative promise of SNRIs in the treatment of pain related disorders including fibromyalgia and depressed patients with multiple somatic complaints. More researches are warranted to generalize currently available preliminary evidences
