    Linkage analysis of longitudinal data

    BACKGROUND: We propose a statistical model for linkage analysis of the longitudinal data. The proposed model is a mixed model based on the new Haseman and Elston model and allows several random effects. Specifically, the proposed model includes a random effect for correlation among sib pairs having one sibling in common, and one for the correlation among siblings from the same parents. RESULTS: The proposed model was applied to the analysis of the Genetic Analysis Workshop 13 simulated data set for a quantitative trait of the systolic blood pressure. A simple independence model and two kinds of random effects models yielded good power for detecting linkage for these data sets, while the random effects models performed slightly better than the independence model. Both random effects models showed similar performance. CONCLUSIONS: The proposed models seem not only quite useful in detecting linkage with the longitudinal data for the trait but also quite flexible. They can handle a wide class of correlation structures. Models with a more general class of covariance structure are desirable

    Additive Value of B-Type Natriuretic Peptide on Rest 201Tl-Dipyridamole Stress 99mTc-Sestamibi Gated Myocardial SPECT in Patients with Normal Left Ventricular Systolic Function

    We evaluated whether BNP has additive value to SPECT in patients with normal left ventricular (LV) systolic function. Data from 224 consecutive patients who underwent rest 201Tl-dipyridamole stress 99mTc-sestamibi gated SPECT and coronary angiography due to chest pain were analyzed. Patients with true positive SPECT showed significant higher BNP level than those with false positive defect (38.5 (19.0–79.8) versus 19.0 (9.3–35.8), P = .01). Patients with true negative SPECT also showed significantly lower BNP level than those with false negative SPECT (39.0 (23.0–77.0) versus 22.0 (15.0–43.0), P = .002). In multivariate analyses, elevated BNP level (using a cut-off value of 23.0 pg/mL) was the strongest and independent predictor of CAD in overall patients (OR 2.75, 95% CI: 1.50–5.023, P = .001) and patients with positive SPECT (OR 3.34, 95% CI: 1.51–7.37, P = .003). The area under the receiver-operating characteristic curve for CAD in overall patients and patients with positive SPECT was 0.673 (95% CI: 0.603–0.743, P < .001) and 0.694 (95% CI: 0.602–0.786, P < .001), respectively. This study suggests that BNP level has additive diagnostic value to SPECT findings in predicting CAD in patients with normal LV systolic function

    Histidine 454 plays an important role in polymerization of human glutamate dehydrogenase

    AbstractAlthough previous chemical modification studies have suggested several residues to be involved in the maintenance of the quaternary structure of glutamate dehydrogenase (GDH), there are conflicting views for the polymerization process and no clear evidence has been reported yet. In the present study, cassette mutagenesis at seven putative positions (Lys333, Lys337, Lys344, Lys346, Ser445, Gly446, and His454) was performed using a synthetic human GDH gene to examine the polymerization process. Of the mutations at the seven different sites, only the mutagenesis at His454 results in depolymerization of the hexameric GDH into active trimers as determined by HPLC gel filtration analysis and native gradient polyacrylamide gel electrophoresis. The mutagenesis at His454 has no effects on expression or stability of the protein. The KM values for NADH and 2-oxoglutarate were 1.5-fold and 2.5-fold greater, respectively, for the mutant GDH than for wild-type GDH, indicating that substitution at position 454 had appreciable effects on the affinity of the enzyme for both NADH and 2-oxoglutarate. The Vmax values were similar for wild-type and mutant GDH. The kcat/KM value of the mutant GDH was reduced up to 2.8-fold. The decreased efficiency of the mutant, therefore, results from the increase in KM values for NADH and 2-oxoglutarate. The results with cassette mutagenesis and HPLC gel filtration analysis suggest that His454 is involved in the polymerization process of human GDH

    THE EFFECTS OF WEARING SPANDEX WEAR WITH COMPRESSION BAND ON MUSCLE ACTIVITY DURING A GOLF SWING

    The purpose of this study was to determine how spandex wear with compressive band affects muscle activities during a golf swing. This study showed that average and maximum nEMG (normalized EMG) values of left AO (external abdominal oblique) were less in EG (experimental group) compared with CG (control group) during back swing, whereas those of left PM (pectoralis major) in EG were greater than CG. It is more likely that EG performed effectively golf swing without excess muscle activity. Thus, the spandex wear with compressive band played an important role in improving swing performance with injury prevention. This has led to suggestions of the need for further kinetic and kinematic analyses to evaluate its function

    AbstractThe structural gene (vmhA) of hemolysin from Vibrio mimicus (ATCC33653) was cloned and sequenced. The vmhA gene contains an open reading frame consisting of 2232 nucleotides which can code for a protein of 744 amino acids with a predicted molecular mass of 83 059. The similarity of amino acid sequence shows 81.6% identity with Vibrio cholerae El Tor hemolysin
