Genome shotgun sequencing and development of microsatellite markers for gerbera (Gerbera hybrida H.) by 454 GS-FLX
The objective of this research was to develop and characterize microsatellite markers for gerbera. We used shotgun sequencing with Roche 454 GS-FLX Titanium technology to identify microsatellite loci in gerbera genomic DNA (Gerbera hybrida). The total length of non-redundant sequences obtained was 22,527,019 bp, which consisted of 3,085 contigs and 28,249 singletons. We assembled 61,958 reads into 3,085 contigs, of which 114 (3.70%) contained microsatellite repeats. The average G+C content was 39.3%. Functional annotation to known sequences yielded 14.7% unigenes in the ‘Raon’ cultivar. Analysis of the gerbera genome DNA (‘Raon’) general library showed that sequences of (AT), (AG), (AAG) and (AAT) repeats appeared most often, whereas (AC), (AAC) and (ACC) were the least frequent. Primer pairs were designed for 80 loci. Only eight primer pairs produced reproducible polymorphic bands in the 28 gerbera accessions analyzed. A total of 30 alleles were identified from the eight polymorphic SSR loci, with two to eight alleles per locus (average level of 3.75). Tokyo Guidelines 2018 management bundles for acute cholangitis and cholecystitis
Management bundles that define items or procedures strongly recommended in clinical practice have been used in many guidelines in recent years. Application of these bundles facilitates the adaptation of guidelines and helps improve the prognosis of target diseases. In Tokyo Guidelines 2013 (TG13), we proposed management bundles for acute cholangitis and cholecystitis. Here, in Tokyo Guidelines 2018 (TG18), we redefine the management bundles for acute cholangitis and cholecystitis. Critical parts of the bundles in TG18 include the diagnostic process, severity assessment, transfer of patients if necessary, and therapeutic approach at each time point. Observance of these items and procedures should improve the prognosis of acute cholangitis and cholecystitis. Superior outcomes and high-risk features with carfilzomib, lenalidomide, and dexamethasone combination therapy for patients with relapsed and refractory multiple myeloma: Results of the multicenter KMMWP2201 study
Carfilzomib, lenalidomide, and dexamethasone (KRd) combination therapy improves the survival of patients with relapsed and/or refractory multiple myeloma (RRMM). Nonetheless, evidence on the use of KRd in Asian populations remains scarce. Accordingly, this study aimed at investigating this regimen’s efficacy in a large group of patients. This retrospective study included patients with RRMM who were treated with KRd at 21 centers between February 2018 and October 2020. Overall, 364 patients were included (median age: 63 years). The overall response rate was 90% in responseevaluable patients, including 69% who achieved a very good partial response or deeper responses. With a median follow-up duration of 34.8 months, the median progression-free survival (PFS) was 23.4 months and overall survival (OS) was 59.5 months. Among adverse factors affecting PFS, highrisk cytogenetics, extramedullary disease, and doubling of monoclonal protein within 2 to 3 months prior to start of KRd treatment significantly decreased PFS and overall survival (OS) in multivariate analyses. Patients who underwent post-KRd stem cell transplantation (i.e.delayed transplant) showed prolonged PFS and OS. Grade 3 or higher adverse events (AEs) were observed in 56% of the patients, and non-fatal or fatal AE’s that resulted in discontinuation of KRd were reported in 7% and 2% of patients, respectively. Cardiovascular toxicity was comparable to that reported in the ASPIRE study. In summary, KRd was effective in a large real-world cohort of patients with RRMM with long-term follow-up. These findings may further inform treatment choices in the treatment of patients with RRMM
Delphi consensus on bile duct injuries during laparoscopic cholecystectomy:An evolutionary cul-de-sac or the birth pangs of a new technical framework?
Bile duct injury (BDI) during laparoscopic cholecystectomy remains a serious iatrogenic surgical complication. BDI most often occurs as a result of misidentification of the anatomy; however, clinical evidence on its precise mechanism and surgeons' perceptions is scarce. Surgeons from Japan, Korea, Taiwan, and the USA, etc. (n=614) participated in a questionnaire regarding their BDI experience and near-misses; and perceptions on landmarks, intraoperative findings, and surgical techniques. Respondents voted for a Delphi process and graded each item on a five-point scale. The consensus was built when 80% of overall responses were 4 or 5. Response rates for the first- and second-round Delphi were 60.6% and 74.9%, respectively. Misidentification of local anatomy accounted for 76.2% of BDI. Final consensus was reached on: (1) Effective retraction of the gallbladder, (2) Always obtaining critical view of safety, and (3) Avoiding excessive use of electrocautery/clipping as vital procedures; and (4) Calot's triangle area and (5) Critical view of safety as important landmarks. Behavior Signature for Fine-grained Traffic Identification
Abstract: With the rapid development of the Internet and a vigorous emergence of new applications, traffic identification has become a key issue for efficient network management. Although various methods have been proposed, there are still several limitations to achieving fine-grained and application-level traffic identification. In this paper, we propose a new signature model called a behavior signature for Internet traffic identification that utilizes the inter-flow relation of application traffic. The proposed behavior signature is a unique traffic behavior pattern appearing in the first few packets of plural traffic flows when a specific function is conducted by an application with a combination of various optional traffic features. This is in contrast to other existing signature models that usually focus on a singular packet or flow for feature extraction and traffic identification. We proved the feasibility and applicability of the proposed behavior signature by developing an extraction and identification algorithm and by conducting experiments on several popular applications
Feasibility Study on Variable-Speed Air Conditioner under Hot Climate based on Real-Scale Experiment and Energy Simulation
It is well known that inverter-driven variable-speed compressor (or inverter) air conditioners are more efficient than constant-speed compressor air conditioners. Therefore, most countries have employed part-load assessment test standards such as ISO 16358, EN 14825 and ASHRAE 116 in addition to the conventional ISO 5151 full-load test standard to appropriately evaluate the part-load efficiencies of variable-speed air conditioners. However, many countries in the Middle East and South America still use the ISO 5151 standard owing to policy or high- temperature environmental considerations. In this study, we experimentally verify the energy saving effect of the inverter air conditioner with respect to the constant-speed air conditioner under the Korean climate with distinct temperature changes of four seasons and under the hot climate of Saudi Arabia throughout the year. ISO 5151 defines test conditions for a single temperature, whereas ISO 16358, EN 14825 and ASHRAE 116 simulate seasonal efficiencies using interpolation of several climate test results. Herein, we directly employ the environmental changes during a day or season in a qualified test room with specific dimension. Using extensive regional and seasonal climate data for Saudi Arabia and South Korea, the changes in temperature conditions are applied directly to the outdoor side and appropriate building cooling load conditions are applied to the indoor side of the air-enthalpy-type test room. The energy savings of the inverter air conditioner were analyzed experimentally according to the spatial and temporal temperature changes. The energy reduction effects of the inverter air conditioner largely depended on the temperature and cooling load changes for a day or season. Furthermore, a feasibility study based on an energy simulation showed that the variable-speed air conditioner could be economical even in hot climates
