    Failure to detect Xenotropic murine leukaemia virus-related virus in Chinese patients with chronic fatigue syndrome

    <p>Abstract</p> <p>Background</p> <p>Recent controversy has surrounded the question of whether xenotropic murine leukaemia virus-related virus (XMRV) contributes to the pathogenesis of chronic fatigue syndrome (CFS). To investigate the question in a Chinese population, 65 CFS patients and 85 blood donor controls were enrolled and multiplex real-time PCR or reverse transcriptase PCR (RT-PCR) was developed to analyze the XMRV infection status of the study participants. The assay was standardized by constructing plasmid DNAs and armored RNAs as XMRV standards and competitive internal controls (CICs), respectively.</p> <p>Results</p> <p>The sensitivities of the multiplex real-time PCR and RT-PCR assays were 20 copies/reaction and 10 IU/ml, respectively, with 100% specificity. The within-run precision coefficient of variation (CV) ranged from 1.76% to 2.80% and 1.70% to 2.59%, while the between-run CV ranged from 1.07% to 2.56% and 1.06% to 2.74%. XMRV was not detected in the 65 CFS patients and 65 normal individuals out of 85 controls.</p> <p>Conclusions</p> <p>This study failed to show XMRV in peripheral blood mononuclear cells (PBMCs) and plasma of Chinese patients with CFS. The absence of XMRV nucleic acids does not support an association between XMRV infection and the development of CFS in Chinese.</p

    Chemokines and chemokine receptors in Behçet’s disease

    Behçet’s disease (BD), a chronic vascular inflammatory disease, is characterized by the symptoms of ocular lesions, recurrent genital and oral ulcers, skin symptoms and arthritis in addition to neurological, intestinal and vascular involvement. The pathogenesis of BD is poorly understood, and there are no effective laboratory markers for the diagnosis of BD. In addition, BD is presently incurable. Chemokines, a family of small secreted chemotactic cytokines, interact with chemokine receptors and mediate the migration, localization and cellular interactions of inflammatory cells. Several studies have suggested that chemokines and their receptors play an important role in the occurrence and development of BD and that these chemokines along with their receptors can be utilized as biomarkers and therapeutic targets.     Quasiphoton at the Subcycle Level in Strong-Field Ionization

    Photon is an energy quanta of light that does not exist at the sub-optical-cycle level. Exploiting the dynamical rotational symmetry of circularly or elliptically polarized light pulses, however, we demonstrate the existence of quasiphotons down to the subcycle level. We illustrate the concept of quasiphotons in strong-field ionization through the correlated spectrum of angular momentum and energy (SAME) of photoelectrons, both at the tunnel exit and in the asymptotic region. Moreover, we propose a protocol based on electron vortices to directly visualize the existence of quasiphotons. Our work paves the pathway towards a deeper understanding of fundamental light-matter interactions with photonic characteristics on the subcycle scale.Comment: 6 pages, 4 figure

    Comparative proteomics study on liver mitochondria of primary biliary cirrhosis mouse model

    BACKGROUND: Primary biliary cirrhosis (PBC) is a liver specific chronic disease with unclear pathogenesis, especially for the early stage molecular events. The mitochondrion is a multi-functional organelle associated with various diseases including PBC. The purpose of this study was to discover the alterations in the mitochondria proteome using an early stage PBC mouse model for revealing the possible pathogenesis mechanisms in the early stages of PBC. METHODS: Mouse model of early stage of PBC was constructed by consecutive administration of poly I:C. Mitochondria of mouse models and controls were purified and comparative proteomics was performed by iTRAQ technology. Then, differentially expressed proteins were validated by western blotting. RESULTS: In total 354 proteins that satisfied the criteria for comparative proteomics study were identified. Of them, nine proteins were downregulated and 20 were up-regulated in liver mitochondria of PBC mouse model. Most differentially expressed proteins are associated with oxidation-reduction and lipid metabolism, and some are involved in the biosynthesis of steroid hormone and primary bile acid. Interestingly, four proteins (HCDH, CPT I, DECR, ECHDC2) involved in the fatty acid beta-oxidation were all upregulated. CONCLUSIONS: iTRAQ is a powerful tool for comparative proteomics study of PBC mouse model and differentially expressed proteins in mitochondria proteome of PBC mouse model provide insights for the pathogenesis mechanism at early stage of PBC

    COVID-19 vaccination willingness among people living with HIV in Shijiazhuang, China: a cross-sectional survey

    ObjectivesThe COVID-19 pandemic imposed an enormous disease and economic burden worldwide. SARS-CoV-2 vaccination is essential to containing the pandemic. People living with HIV (PLWH) may be more vulnerable to severe COVID-19 outcomes; thus, understanding their vaccination willingness and influencing factors is helpful in developing targeted vaccination strategies.MethodsA cross-sectional study was conducted between 15 June and 30 August 2022 in Shijiazhuang, China. Variables included socio-demographic characteristics, health status characteristics, HIV-related characteristics, knowledge, and attitudes toward COVID-19 vaccination and COVID-19 vaccination status. Multivariable logistic regression was used to confirm factors associated with COVID-19 vaccination willingness among PLWH.ResultsA total of 1,428 PLWH were included, with a 90.48% willingness to receive the COVID-19 vaccination. PLWH were more unwilling to receive COVID-19 vaccination for those who were female or had a fair/poor health status, had an allergic history and comorbidities, were unconvinced and unsure about the effectiveness of vaccines, were unconvinced and unsure about the safety of vaccines, were convinced and unsure about whether COVID-19 vaccination would affect ART efficacy, or did not know at least a type of domestic COVID-19 vaccine. Approximately 93.00% of PLWH have received at least one dose of the COVID-19 vaccine among PLWH, and 213 PLWH (14.92%) reported at least one adverse reaction within 7 days.ConclusionIn conclusion, our study reported a relatively high willingness to receive the COVID-19 vaccination among PLWH in Shijiazhuang. However, a small number of PLWH still held hesitancy; thus, more tailored policies or guidelines from the government should be performed to enhance the COVID-19 vaccination rate among PLWH

    Different responses of soil element contents and their stoichiometry (C: N: P) to different grazing intensity on the Tibetan Plateau shrublands

    Potentilla fruticosa, a major alpine shrubland type, is widely distributed across the Tibetan Plateau, and grazing is the most common disturbance in the shrublands of P. fruticosa. However, soil organic carbon (SOC), soil total nitrogen (STN), soil total phosphorus (STP), and their stoichiometry under different grazing intensities were unclear. In our study, we explored SOC, STN, STP, their stoichiometry, and their controlling factors in the grazing disturbance of heavy grazing (HG), moderate grazing (MG), light grazing (LG), and no grazing (NG) conditions in the Tibetan Plateau P. fruticosa shrublands. The grazing intensities were mainly assessed by considering the shrublands’ ground cover, the indicators of the road density, the distance between sampling sites and cowshed or sheep shed, the amounts of cow and sheep dung, and vegetation that had been gnawed and stampeded. Our results indicated that soil physical properties of soil temperature and bulk density have decreasing trends with decreasing grazing intensities from HG to NG. The SOC, STN, STP, and soil C:N and C:P ratios have increasing trends with decreasing grazing intensities from HG to NG, while the changes in soil N:P ratio were relatively stable along grazing intensities. Our results indicated that HG generally had stronger effects on SOC, STN, and soil C:N and C:P ratios than NG, indicating substantial effects of grazing disturbance on biogeochemical cycles of SOC and STN in the shrubland ecosystems. Therefore, for the alpine shrubland of P. fruticosa, the HG should be avoided for sustainable cycling of soil nutrients and the balance of soil nutrient stoichiometry. The grazing types can directly affect plant conditions, and plant conditions can directly affect soil physical and chemical properties and litter standing crops. Finally, soil physicochemical properties and litter standing crop resulting from different grazing intensities directly control SOC, STN, and STP. For the soil stoichiometry, the soil’s physical and chemical properties resulting from different grazing intensities have direct impacts on soil C:P and N:P ratios
