    Total and horizontal distances of the foveal stereotaxic displacement can be prognostic indicators for patients with idiopathic epiretinal membrane

    IntroductionThis study aimed to examine the foveal stereo deviations in the different ectopic inner foveal layer (EIFL) stages of idiopathic epiretinal membrane (iERM) and assess its predictive utility for the baseline and postoperative best-corrected visual acuity (BCVA).MethodsBased on the calculational combination of foveal displacements in the horizontal and vertical axial optical coherence tomography (OCT) images, the foveal stereotaxic displacement was estimated through the total distance (TD, the distance from the foveal bottom to the inner edge of displaced central foveal) and horizontal distance (HD, projection of the TD in the retinal plane). The preoperative TD, HD, and other OCT- and OCT angiography (OCTA)-related indicators were obtained. The correlations between structural parameters and baseline and postoperative BCVA were evaluated through correlation and multiple linear regression analyses.ResultsIn patients with advanced EIFL stage, there was a significant increase in the HD, TD, baseline log of the minimum angle of resolution unit for BCVA, central macular thickness (CMT), acircularity index, and incidence of microcystic macular edema (MME; p < 0.05). Further, they showed a decreased foveal avascular zone (FAZ) area and perimeter (p < 0.001). HD, TD, CMT, MME, FAZ area, and FAZ perimeter were significantly correlated with the baseline and postoperative BCVA (p < 0.05). TD had the highest correlation indexic and was an individual predictor of the baseline and postoperative BCVA. Moreover, FD-300 and MME were individual predictors of postoperative BCVA.DiscussionStereoscopic foveal deviations significantly correlated with the baseline and postoperative visual acuity. TD may be used as an independent prognostic factor for BCVA

    NF45/NF90-mediated rDNA transcription provides a novel target for immunosuppressant development

    Herein, we demonstrate that NFAT, a key regulator of the immune response, translocates from cytoplasm to nucleolus and interacts with NF45/NF90 complex to collaboratively promote rDNA transcription via triggering the directly binding of NF45/NF90 to the ARRE2-like sequences in rDNA promoter upon T-cell activation in vitro. The elevated pre-rRNA level of T cells is also observed in both mouse heart or skin transplantation models and in kidney transplanted patients. Importantly, T-cell activation can be significantly suppressed by inhibiting NF45/NF90-dependent rDNA transcription. Amazingly, CX5461, a rDNA transcription-specific inhibitor, outperformed FK506, the most commonly used immunosuppressant, both in terms of potency and off-target activity (i.e., toxicity), as demonstrated by a series of skin and heart allograft models. Collectively, this reveals NF45/NF90-mediated rDNA transcription as a novel signaling pathway essential for T-cell activation and as a new target for the development of safe and effective immunosuppressants

    Joint effect of BMI and metabolic status on mortality among adults: a population-based longitudinal study in United States

    Abstract We explored the joint effects of different metabolic obesity phenotypes on all-cause and disease-specific mortality risk among the American population. Data were obtained from the National Health and Nutrition Examination Survey (NHANES) 1999–2018. Mortality outcome data were from mortality files linked to National Death Index record and follow-up information was up to December 31, 2019. 50,013 participants were finally included. Four metabolic obesity phenotypes were defined based on obesity and metabolic status: metabolically healthy obese (MHO), metabolically unhealthy obese (MUO), metabolically healthy non-obese (MHNO), and metabolically unhealthy non-obese (MUNO). Population-weighted Cox proportional hazards models were used to explore the all-cause and disease-specific mortality risk of metabolic obesity phenotypes. The all-cause mortality risk of MUO and MUNO was significantly higher than MHNO. MUNO was associated with a significantly increased risk of death from heart disease (HR: 1.40, 95% CI 1.16–1.70), hypertension (HR: 1.68, 95% CI 1.34–2.12), diabetes (HR: 2.29, 95% CI 1.67–3.15), and malignant neoplasms (HR:1.29, 95% CI 1.09–1.53). Metabolic unhealth significantly increased the risk of all-cause mortality, regardless of obesity status. Among individuals with metabolic unhealthy status, obesity significantly reduced the risk of all-cause mortality (HR: 0.91, 95% CI 0.85–0.98). Our study highlights the importance of identifying and characterizing metabolic obesity phenotypes in obese and metabolically abnormal patients, as well as healthy adults. Comprehensive evaluation of obesity and metabolic status is necessary to adopt appropriate interventions and treatment measures and maximize patient benefit

    Analysis of the Effect of Thickness on the Performance of Polymeric Heart Valves

    Polymeric heart valves (PHVs) are a promising and more affordable alternative to mechanical heart valves (MHVs) and bioprosthetic heart valves (BHVs). Materials with good durability and biocompatibility used for PHVs have always been the research focus in the field of prosthetic heart valves for many years, and leaflet thickness is a major design parameter for PHVs. The study aims to discuss the relationship between material properties and valve thickness, provided that the basic functions of PHVs are qualified. The fluid−structure interaction (FSI) approach was employed to obtain a more reliable solution of the effective orifice area (EOA), regurgitant fraction (RF), and stress and strain distribution of the valves with different thicknesses under three materials: Carbothane PC−3585A, xSIBS and SIBS−CNTs. This study demonstrates that the smaller elastic modulus of Carbothane PC−3585A allowed for a thicker valve (>0.3 mm) to be produced, while for materials with an elastic modulus higher than that of xSIBS (2.8 MPa), a thickness less than 0.2 mm would be a good attempt to meet the RF standard. What is more, when the elastic modulus is higher than 23.9 MPa, the thickness of the PHV is recommended to be 0.l–0.15 mm. Reducing the RF is one of the directions of PHV optimization in the future. Reducing the thickness and improving other design parameters are reliable means to reduce the RF for materials with high and low elastic modulus, respectively

    The Therapeutic Effects of EFNB2-Fc in a Cell Model of Kawasaki Disease

    The EphrinB2/EphB4 signaling pathway involves the regulation of vascular morphogenesis and angiogenesis. However, little is known about EphrinB2/EphB4 in the pathogenesis of Kawasaki disease (KD) and coronary artery aneurysm formation. Hence, this study aimed to explore the role of EphrinB2/EphB4 and the potential therapeutic effect of EphrinB2-Fc in the coronary arterial endothelial injury of KD. The levels of EphB4 were compared between KD patients and healthy children. Human coronary artery endothelial cells (HCAECs) were stimulated with sera from acute KD patients to establish the KD cell model. The overexpression of EphB4 or treatment with EphrinB2-Fc was found to intervene in the cell model. The cell migration, angiogenesis, and proliferation ability were assessed, and the expression of inflammation-related factors was measured. Our study showed that EphB4 showed low expression in both KD patients and the cell model of KD. The EphB4 protein levels in the CECs of CAA+ KD patients were much lower than those in healthy children. EphrinB2-Fc treatment of KD sera-activated HCAECs suppressed cell proliferation, reduced the expression of inflammation-related factors (such as IL-6 and P-selectin), and elevated cell angiogenesis ability. The results reveal that EphrinB2-Fc has a protective function in endothelial cells and has promising clinical applications for protecting vascular endothelium in patients with KD

    Characteristics of Atmospheric Deposition during the Period of Algal Bloom Formation in Urban Water Bodies

    Urban water bodies are limited by poor mobility, small surface areas, and little water supply; thus, they are sensitive to atmospheric nutrient inputs, especially during the optimal period of algae growth. This study investigated the impact of atmospheric deposition on the Quyang urban water body in Shanghai. Observations that coupled atmospheric organic matter, nitrogen and phosphorous and the actual urban water body (nutrient availability and Chlorophyll-a concentrations (Chl-a)) were conducted during spring and summer. Atmospheric total organic carbon (TOC), total nitrogen (TN), ammonia (N-NH4+) and total phosphorus (TP) deposition ranged from 35–81, 3–40, 0.79–20.40 and 0.78–0.25 mg m−2 d−1, respectively. The soluble N/P molar ratios of the bulk deposition (ranging from 56–636) were well above the Redfield ratio (N/P = 16). Nutrient inputs from atmospheric deposition have been suggested to be a strong factor for increasing the likelihood of P limitation in the water bodies. The actual loads to small, shallow urban water bodies were assessed and found to be ~50, 130, 130 (the N-fixation contributes to the atmospheric deposition inputs especially during the spring), and 80% of TOC, TN, N-NH4+, and TP, respectively, representing nutrients transferred into the water phase. The maximum primary production (evaluated as Chl-a) stock resulting in a 2-m-deep water column from the above inputs ranged from 2.54–7.98 mg Chl-a m−3. As a continuous source of nutrients, atmospheric deposition should not be underestimated as a driving force for urban water body eutrophication, and it potentially influences primary production, especially during the optimal algae growth period

    Impact-resistant supercapacitor by hydrogel-infused lattice

    Abstract The safety of energy storage devices is increasingly crucial due to the growing requirements for application under harsh conditions. Effective methods for enhancing robustness without compromising functionality are necessary. Here we present an impact-resistant, ready-to-use supercapacitor constructed from self-healable hydrogel electrolyte-infused lattice electrodes. Three-dimensional-printed carbon-coated silicon oxycarbide current collectors provide mechanical protection, with compressive stress, Young’s modulus, and energy absorption up to 70.61 MPa, 2.75 GPa, and 92.15 kJ/m3, respectively. Commercially viable polyaniline and self-healable polyvinyl alcohol hydrogel are used as active coatings and electrolytes. I-wrapped package structured supercapacitor electrode exhibits a static specific capacitance of 585.51 mF/cm3 at 3 mA/cm3, with an energy density of 97.63 μWh/cm3 at a power density of 0.5 mW/cm3. It maintains operational integrity under extreme conditions, including post-impact with energy of 0.3 J/cm3, dynamic loading ranging from 0 to 18.83 MPa, and self-healing after electrolyte damage, demonstrating its promise for applications in extreme environments

    Long-Term Trends of Asthma Mortality in China from 2000 to 2019: A Joinpoint Regression and Age-Period-Cohort Analysis

    Background: Trends of asthma mortality vary widely all over the world, while the trends in China over the past 15 years are unknown. The aim of this study was to assess the trends of asthma mortality in China. Methods: Asthma deaths and demographic characteristics were collected from National Death Cause Datasets of Disease Surveillance System between 2004 and 2019. The data were analyzed with joinpoint regression analysis and age-period-cohort (APC) analysis for the mortality rate due to asthma in China. Results: Asthma mortality declined from 2.4 (95% confidence interval (CI): 2.3–2.5) per 100,000 in 2004 to 1.6 (95% CI: 1.5–1.7) per 100,000 in 2019. Age-adjusted asthma mortality rates decreased for men and women in urban and rural areas from 2004 through 2019. The decreasing trend of the mortality rate has slowed down substantially during 2007 and 2009. After that, the decreasing trend has stabilized. The asthma mortality rates generally have a positive relationship with the age of the population when controlling for period and cohort. The period trend decreased and then increased when controlling for age and cohort. Conclusions: We should pay more attention to asthma management plans or treatment for aging people who are facing higher risk of asthma death
