    Multifunctional Polymeric Enveloped Nanocarriers: Targeting Extracellular and Intracellular Barriers

    Over the past several years, employment of multifunctional polymeric excipients-based nanoparticles for controlled and targeted drug delivery of therapeutic modalities to mucosal membrane-based organelles and systemic circulation has gained enormous interest. Because they promise to resolve numerous key therapeutical issues associated with current clinical practice including low treatment efficacy and significant side effects. Potential controlled and targeted drug delivery systems, therefore, should be able to overcome not only extracellular barriers but also intracellular barriers. Extracellularly, targeted nanocarriers ought to provide extended circulation time, selective binding to the targeted mucosal tissues, long residence time at the site of absorption, and controlled drug release. Intracellularly, the targeted nanocarriers should offer cellular uptake, cellular localization, and endosomal release. Hence, this chapter will provide an overview of the unique chemistry of multifunctional polymeric enveloped diverse nanocarriers such as dendrimers, semiconducting polymer dots, quantum dots, carbon dots, and magnetic as versatile platform addressing both extracellular and intracellular barriers

    Crossing Biological Barriers for Leishmaniasis Therapy: From Nanomedicinal Targeting Perspective

    Despite past 60 years of extensive research in antileishmanial drug development, the successful therapy of this disease cannot be achieved at full potential. The biological barriers encountered by the therapeutic modalities favor the disseminations of the disease like intramacrophage location of parasite, lack of oral bioavailability, permeability across the cutaneous tissue, and active efflux of the drug. Nanomedicines are specifically engineered nano-sized delivery systems. The goal of designing a nanomedicine is to achieve the specific therapeutic objective via targeting the specific cells and intracellular locations, pharmacological receptors, enzymes and proteins, crossing biological barriers, and navigation through endocytic pathways. This chapter will cover various nanomedicinal approaches like targeting the macrophages, pathological organs, efflux pumps, metabolic enzymes, redox biology of Leishmania by using polymeric and metal nanocarriers to overcome all the biological barriers thus providing a successful alternative over the conventional therapies

    ABSTRACT A placebo-controlled trial compared 6% hexadecyl-phosphorylcholine (HePC) and 12% benzethonium chloride ointment with placebo ointment for treatment of cutaneous leishmaniasis. Cuta--neous lesions were experimentally induced by inoculation with leishmania promastigotes in 60 golden hamsters. Forty (40) animals were treated with drug and 20 with placebo ointment applied twice daily for 15 days. After treatment, all lesions were significantly reduced in size in the treatment group compared with the placebo ointment. No parasites were detected in smears from 35/40 of the drug-treated lesions and no relapses occurred over 120 days of observation. 5 Efficacy of hexadecyl-phosphorylcholine ointment for treatment of cutaneous La pommade d'héxadécylphosphocholine pour le traitement de la leishmaniose cutanée : essai sur l'animal RÉSUMÉ Un essai contrôlé contre placebo a comparé une pommade contenant 6 % d'héxadécyl--phosphocholine et 12 % de chlorure de benzéthonium avec une pommade placebo pour le traitement de la leishmaniose cutanée. Les lésions cutanées ont été induites expérimentalement par inoculation de promastigotes de leishmania sur 60 hamsters dorés. Quarante (40) animaux ont été traités avec le médicament et 20 avec la pommade placebo appliquée deux fois par jour pendant 15 jours. Après le traitement, toutes les lésions ont significativement diminué en taille dans le groupe du traitement par rapport à la pommade placebo. Aucun parasite n'a été détecté dans les frottis de 35 des 40 lésions traitées par le médicament et aucune rechute ne s'est produite pendant 120 jours d'observation

    Latent activity of curcumin against leishmaniasis in vitro

    In this study the anti-proliferative effect of curcumin (curcuma longa) that is the active ingredient of ground dried rhizome has been studied against three local and three reference leishmanial strains, Leishmania major, Leishmania tropica and Leishmania infantum (Pakistani isolate). Curcumin has shown an average IC50 of 5.3mM against promastigotes of various leishmanial strains which is much lower as compared with pentamidine that is one of the basic treatments against leishmaniasis. The main draw back attributed to these assays performed on promastigotes is the heterogeneity of results compared with those obtained with intracellular amastigotes or with in vivo effect. We also tested activity of curcumin against axenic amastigote like cells (AALC) of L. major strain (MHOM/PK/88/DESTO). Curcumin proves to be far more potent then pentamidine against AALC which further strengthens the fact about its leishmaniacidal activity

    Antispasmodic and vasodilator activities of Morinda citrifolia root extract are mediated through blockade of voltage dependent calcium channels

    <p>Abstract</p> <p>Background</p> <p><it>Morinda citrifolia </it>(Noni) is an edible plant with wide range of medicinal uses. It occurs exclusively in tropical climate zone from India through Southeast Asia and Australia to Eastern Polynesia and Hawaii. The objective of this study was to explore the possible mode(s) of action for its antispasmodic, vasodilator and cardio-suppressant effects to rationalize its medicinal use in gut and cardiovascular disorders.</p> <p>Methods</p> <p>Isolated tissue preparations such as, rabbit jejunum, rat and rabbit aorta and guinea pig atria were used to test the antispasmodic and cardiovascular relaxant effects and the possible mode of action(s) of the 70% aqueous-ethanolic extract of <it>Morinda citrifolia </it>roots (Mc.Cr).</p> <p>Results</p> <p>The Mc.Cr produced a concentration-dependent relaxation of spontaneous and high K<sup>+ </sup>induced contractions in isolated rabbit jejunum preparations. It also caused right ward shift in the concentration response curves of Ca<sup>++</sup>, similar to that of verapamil. In guinea-pig right atria, Mc.Cr caused inhibition of both atrial force and rate of spontaneous contractions. In rabbit thoracic aortic preparations, Mc.Cr also suppressed contractions induced by phenylephrine (1.0 μM) in normal- Ca<sup>++ </sup>and Ca<sup>++</sup>-free Kerb's solutions and by high K<sup>+</sup>, similar to that of verapamil. In rat thoracic aortic preparations, Mc.Cr also relaxed the phenylephrine (1.0 μM)-induced contractions. The vasodilatory responses were not altered in the presence of L-NAME (0.1 mM) or atropine (1.0 μM) and removal of endothelium.</p> <p>Conclusions</p> <p>These results suggest that the spasmolytic and vasodilator effects of Mc.Cr root extract are mediated possibly through blockade of voltage-dependent calcium channels and release of intracellular calcium, which may explain the medicinal use of <it>Morinda citrifolia </it>in diarrhea and hypertension. However, more detailed studies are required to assess the safety and efficacy of this plant.</p

    Antileishmanial activity of aqueous onion extract in vitro

    Onion has had an important dietary and medicinal role for centuries. In this study the antileishmanial effect of aqueous onion extract (AOE) was investigated. Five leishmanial strains in the promastigote stage were studied in vitro. Seventy-two hour inoculation of AOE gave an IC100 and average IC50 values of 1.25 mg/ml and 0.376 mg/ml, respectively, against all leishmanial strains tested
