    Exploring PANoptosis in breast cancer based on scRNA-seq and bulk-seq

    BackgroundPANoptosis, a cell death pathway involving pyroptosis, apoptosis, and necroptosis, is pivotal in the development of malignancy. However, in the field of breast cancer, the interaction between PANoptosis and tumor cells has not been thoroughly explored.MethodsWe downloaded breast cancer data and GSE176078 single-cell sequencing dataset from Gene Expression Omnibus (GEO) and The Cancer Genome Atlas (TCGA) databases to obtain PANoptosis-associated genes. To construct prognostic models, COX and LASSO regression was used to identify PANoptosis-associated genes with prognostic value. Finally, immune infiltration analysis and differential analysis of biological functions were performed.ResultsRisk grouping was performed according to the prognostic model constructed by COX regression and LASSO regression. The low-risk group showed a better prognosis (P < 0.05) and possessed higher levels of immune infiltration and expression of immune checkpoint-related genes. In addition, the lower the risk score, the higher the degree of microsatellite instability (MSI). Meanwhile, radixin (RDX), the gene with the highest hazard ratio (HR) value among PANoptosis prognosis-related genes, was explicitly expressed in artery Iendothelial cells (ECs) and was widely involved in signaling pathways such as immune response and cell proliferation, possessing rich biological functions.ConclusionWe demonstrated the potential of PANoptosis-based molecular clustering and prognostic features in predicting the survival of breast cancer patients. Furthermore, this study has led to a deeper understanding of the role of PANoptosis in breast cancer and has the potential to provide new directions for immunotherapy of breast cancer

    Effect of Clogging on the Permeability of Porous Asphalt Pavement

    The purpose of this paper is to report on the drainage of porous asphalt pavement evaluation method suited for use in analyzing clogging effect. To preliminarily reveal the decrease in permeability caused by clogging of permeable asphalt pavement, an innovative device was proposed to evaluate the anisotropy of permeability influenced by clogging, and the maximum drainage capacity without surface ponding can be obtained when the supplied water was controlled. Then, finite element models for asphalt pavements with hydromechanical coupling were proposed based on porous media theory and Biot’s theory. The variation in pore water pressure was simulated by considering the decrease in voids and the increase in clogging grains. The results indicate that the internally retained water should not be ignored because the semiconnected voids were filled with water rapidly at the beginning of permeability tests. To avoid surface ponding, the drainage capacity coefficient (DCC) can be used to evaluate the maximum drainage capacity (MDC) influenced by clogging. Moreover, the pore water pressure increased due to the reduction in voids and a high level of clogging. In addition, the peak value of pore water pressure is also affected by the upper-layer height of the pavement. Under the action of clogging and driving load, a reasonable thickness of the upper layer and a drainage evaluation should be considered to improve road safety

    Green Manure Crops as Food Source: Impact on the Performance of the Migratory Beet Webworm, <i>Loxostege sticticalis</i> (Lepidoptera: Pyralidae)

    The application of green manure is crucial for achieving sustainable agriculture and animal husbandry, but pest management is often overlooked. Conducting a risk assessment for insect pests in green manure is essential. The beet webworm, Loxostege sticticalis, a polyphagous insect, is currently experiencing an outbreak in northern China, and represents a significant migratory pest. A two-sex life table and flight mill test approach was used to comprehensively evaluate the effects of three major legume green manure crops (Pisum sativam, Vicia sativa, and Vicia villosa) on the growth, development, fecundity, and flight ability of L. sticticalis in China. Our findings indicate that L. sticticalis cannot utilize V. villosa for generational development. L. sticticalis shows reduced performance on P. sativam and V. sativa compared to its suitable host Chenopodium album. However, both the population parameters (R0, r, λ, and T) and the population prediction results suggest that L. sticticalis can adapt to P. sativam and V. sativa. In the process of promoting green manure, careful consideration should be given to the selection of appropriate green manure varieties and the implementation of effective pest control measures during their planting. Our findings lay the groundwork for the promotion of green manure and implementation of an ecological management plan for L. sticticalis

    Evaluation of Pavement Runoff and Driving Safety on Highway Curve Segment

    Runoff depth distribution on the concave and circular curve sections is obtained from a two-dimensional numerical simulating model in order to analyze the temporal and spatial variation of the pavement runoff on the curve section. The two-dimensional model verified by the field data can depict the alignment of pavement more accurately as compared to the empirical equation and a one-dimensional model. The runoff on the concave section and circular curve section is compared for the free water drainage and centerline drainage. Results show that a two-dimensional model is essential for the analysis of the centerline drainage. The runoff depth can be controlled by a reasonable curb height and location interval. The drainage type affects the variation of the runoff depth on the nearside lane, and the maximum water depth can be up to more than 80 mm on the concave section and nearly 60 mm on the circular curve section under centerline drainage. Besides the existing hydroplaning results, the runoff depth difference of the wheel trace should be considered to evaluate driving safety. Sideslip will occur when the depth difference becomes more than 6 mm under condition that the runoff depth is less than the tread depth (7 mm). When the runoff depth is more than the tread depth, sideslip will occur once the depth difference exceeds 4 mm

    Ferroptosis participates in dibutyl phthalate-aggravated allergic asthma in ovalbumin-sensitized mice

    Dibutyl phthalate (DBP), used as a plasticizer, is of wide concern as an environmental pollutant since it has certain immunotoxicity. Although there is growing evidence supporting a link between DBP exposure and allergic airway inflammation, there is less information concerned with whether the ferroptosis pathway is involved in DBP-aggravated allergic asthma in ovalbumin (OVA)-sensitized mice. This study aimed to investigate the role and underlying mechanisms of ferroptosis in DBP-exposed allergic asthmatic mice. Balb/c mice were orally exposed to 40 mg/kg−1 DBP for 28 days, followed by sensitization with OVA and seven consecutive challenges with nebulized OVA. We analyzed airway hyperresponsiveness (AHR), immunoglobulins, inflammation and pulmonary histopathology, to investigate whether DBP exacerbates allergic asthma in OVA-induced mice. We also measured the biomarkers of ferroptosis (Fe2+, GPX4, PTGS2), proteins related to the ferroptosis pathway (VEGF, IL-33, HMGB1, SLC7A11, ALOX15, PEBP1), and indices of lipid peroxidation (ROS, Lipid ROS, GSH, MDA, 4-HNE), to explore the role of ferroptosis in DBP+OVA mice. Finally, we used ferrostatin-1 (Fer-1) as an antagonist against the harmful effects of DBP. The results showed that, DBP+OVA mice had a significant increase in AHR, airway wall remodeling and airway inflammation. Further, we showed that DBP aggravated allergic asthma via ferroptosis and lipid peroxidation, and that Fer-1 inhibited ferroptosis and alleviated the pulmonary toxicity of DBP. These results suggest that ferroptosis participates in the exacerbation of allergic asthma resulting from oral exposure to DBP, highlighting a novel pathway for the connection between DBP and allergic asthma
