    Current Gene Expression Studies in Esophageal Carcinoma

    Esophageal carcinoma is one of the deadliest cancers with highly aggressive potency, ranking as the sixth most common cancer among males and ninth most common cancer among females globally. Due to metastasis and invasion of surrounding tissues in early stage, the 5-year overall survival rate (14%) of esophageal cancer remains poor, even in comparison with the dismal survival rates (4%) from the 1970s. Numerous genes and proteins with abnormal expression and function involve in the pathogenesis of esophageal cancer, but the concrete process remains unclear. Microarray technique has been applied to investigating esophageal cancer. Many gene expression studies have been undertaken to look at the specific patterns of gene transcript levels in esophageal cancer. Human tissues and cell lines were used in these geneprofiling studies and a very valuable and interesting set of data has resulted from various microarray experiments. These expression studies have provided increased understanding of the complex pathological mechanisms involved in esophageal cancer. The eventual goal of microarray is to discover new markers for therapy and to customize therapy based on an individual tumor genetic composition.     IMP3 signatures of fallopian tube: a risk for pelvic serous cancers

    BACKGROUND:Recent advances suggest fallopian tube as the main cellular source for women's pelvic serous carcinoma (PSC). In addition to TP53 mutations, many other genetic changes are involved in pelvic serous carcinogenesis. IMP3 is an oncofetal protein which has recently been observed to be overexpressed in benign-looking tubal epithelia. Such findings prompted us to examine the relationship between IMP3 over-expression, patient age and the likelihood of development of PSC.METHODS:Fallopian tubes from three groups (low-risk, high-risk, and PSC) of patients with matched ages were studied. Age was recorded in 10years intervals ranging from age 20 to older than 80. The number of IMP3 signatures (defined by 10 or more tubal secretory cells stained positively and continuously in benign appearing tubal mucosa) from both tubal fimbria and ampulla segments was measured. The data was analyzed by standard contingency table and Poisson distribution methods after age adjustment. IMP3 overexpression was also examined in serous tubal intraepithelial carcinoma and PSC.RESULTS:The positive IMP3-stained cells are mainly tubal secretory cells. The absolute number of tubal IMP3 signatures increased significantly within each age group. Age remained a significant risk factor for serous neoplasia after age adjustment. IMP3 signatures were more frequent in the patients of both high-risk and PSC groups. The presence of IMP3 signatures in tubal mucosa was significantly associated with tubal or pelvic serous carcinogenesis (p<0.001).CONCLUSIONS:The findings suggest that tubal secretory cells with IMP3 signatures showing growth advantage could potentially serve as a latent precancer biomarker for tubal or pelvic serous carcinomas in women.This item is part of the UA Faculty Publications collection.     Up-regulation of fas reverses cisplatin resistance of human small cell lung cancer cells

    <p>Abstract</p> <p>Background/Aim</p> <p>Fas/FasL system is a major regulator of apoptosis. The mechanisms by which Fas mediates cisplatin resistance remain unclear. The aim of this study is to explore the effect of Fas over-expression on cisplatin resistance of small cell lung cancer cells and its possible mechanisms.</p> <p>Materials and methods</p> <p>Fas was over-expressed in H446/CDDP cells by infection with the adenoviruses containing Fas. Sensitivity of Fas-overexpressed H446/CDDP cells to cisplatin was evaluated using MTT assay. Expressions of Fas, GST-Ď€ and ERCC1 were detected by RT-PCR and Western blot analysis. Apoptosis rate was examined by FACS.</p> <p>Results</p> <p>Over-expression of Fas in H446/CDDP cells significantly decreased the expressions of GST-Ď€ and ERCC1 at mRNA and protein levels, and increased the cell apoptosis. Furthermore, up-regulation of Fas significantly decreased the tolerance of H446/CDDP cells to cisplatin.</p> <p>Conclusion</p> <p>Over-expression of Fas reverses drug resistance of H446/CDDP cells, possibly due to the increased cell sensitivity to apoptosis and the decreased expressions of GST-Ď€ and ERCC1.</p

    Retrospective analysis for thirty-nine patients with solitary fibrous tumor of pleura and review of the literature

    <p>Abstract</p> <p>Background</p> <p>Solitary fibrous tumor of the pleura (SFTP) is an uncommon neoplasm arising from mesenchymal cells. The aim of this study is to summarize the experience and the outcome of the surgical treatment for 39 cases of SFTP.</p> <p>Methods</p> <p>From January 2004 to December 2008, 39 patients underwent surgical resection of SFTP in our department. All patients had clinical follow-up by the same team of surgeons. The mean follow-up was 40.3 months.</p> <p>Results</p> <p>A local removal of the neoplasm was accomplished by video-assisted thoracic surgery (VATS) in 9 patients (group A) and by thoracotomy in 30 patients (group B) respectively. Comparing with group B, operations in group A took significantly less operative time, blood loss and spent less time in the intensive care unit and hospital. All specimens were positive for CD34 and Bcl-2. One patient developed recurrence, and the remaining 38 patients are alive and disease free at the end of follow-up.</p> <p>Conclusions</p> <p>Malignant SFTP still had the potential recurrence. VATS represents the more acceptable choice for the selected patients with SFTP.</p

    Gemcitabine enhances cell invasion via activating HAb18G/CD147-EGFR-pSTAT3 signaling

    Pancreatic cancer, one of the most lethal cancers, has very poor 5-year survival partly due to gemcitabine resistance. Recently, it was reported that chemotherapeutic agents may act as stressors to induce adaptive responses and to promote chemoresistance in cancer cells. During long-term drug treatment, the minority of cancer cells survive and acquire an epithelial-mesenchymal transition phenotype with increased chemo-resistance and metastasis. However, the short-term response of most cancer cells remains unclear. This study aimed to investigate the short-term response of pancreatic cancer cells to gemcitabine stress and to explore the corresponding mechanism. Our results showed that gemcitabine treatment for 24 hours enhanced pancreatic cancer cell invasion. In gemcitabine-treated cells, HAb18G/CD147 was up-regulated; and HAb18G/CD147 down-regulation or inhibition attenuated gemcitabine-enhanced invasion. Mechanistically, HAb18G/CD147 promoted gemcitabine-enhanced invasion by activating the EGFR (epidermal growth factor receptor)-STAT3 (signal transducer and activator of transcription 3) signaling pathway. Inhibition of EGFR-STAT3 signaling counteracted gemcitabine-enhanced invasion, and which relied on HAb18G/CD147 levels. In pancreatic cancer tissues, EGFR was highly expressed and positively correlated with HAb18G/CD147. These data indicate that pancreatic cancer cells enhance cell invasion via activating HAb18G/CD147-EGFR-pSTAT3 signaling. Our findings suggest that inhibiting HAb18G/CD147 is a potential strategy for overcoming drug stress-associated resistance in pancreatic cancer

    Bilateral transfer of motor performance as a function of motor imagery training: a systematic review and meta-analysis

    ObjectiveThe objective of this review was to evaluate the efficacy of mental imagery training (MIT) in promoting bilateral transfer (BT) of motor performance for healthy subjects.Data sourcesWe searched 6 online-databases (Jul-Dec 2022) using terms: “mental practice,” “motor imagery training,” “motor imagery practice,” “mental training,” “movement imagery,” “cognitive training,” “bilateral transfer,” “interlimb transfer,” “cross education,” “motor learning,” “strength,” “force” and “motor performance.”Study selection and data extractionWe selected randomized-controlled studies that examined the effect of MIT on BT. Two reviewers independently determined if each study met the inclusion criteria for the review. Disagreements were resolved through discussion and, if necessary, by a third reviewer. A total of 9 articles out of 728 initially identified studies were chosen for the meta-analysis.Data synthesisThe meta-analysis included 14 studies for the comparison between MIT and no-exercise control (CTR) and 15 studies for the comparison between MIT and physical training (PT).ResultsMIT showed significant benefit in inducing BT compared to CTR (ES = 0.78, 95% CI = 0.57–0.98). The effect of MIT on BT was similar to that of PT (ES = –0.02, 95% CI = –0.15–0.17). Subgroup analyses showed that internal MIT (IMIT) was more effective (ES = 2.17, 95% CI = 1.57–2.76) than external MIT (EMIT) (ES = 0.95, 95% CI = 0.74–1.17), and mixed-task (ES = 1.68, 95% CI = 1.26–2.11) was more effective than mirror-task (ES = 0.46, 95% CI = 0.14–0.78) and normal-task (ES = 0.56, 95% CI = 0.23–0.90). No significant difference was found between transfer from dominant limb (DL) to non-dominant limb (NDL) (ES = 0.67, 95% CI = 0.37–0.97) and NDL to DL (ES = 0.87, 95% CI = 0.59–1.15).ConclusionThis review concludes that MIT can serve as a valuable alternative or supplement to PT in facilitating BT effects. Notably, IMIT is preferable to EMIT, and interventions incorporating tasks that have access to both intrinsic and extrinsic coordinates (mixed-task) are preferred over those that involve only one of the two coordinates (mirror-task or normal-task). These findings have implications for rehabilitation of patients such as stroke survivors

    CROP: Conservative Reward for Model-based Offline Policy Optimization

    Offline reinforcement learning (RL) aims to optimize policy using collected data without online interactions. Model-based approaches are particularly appealing for addressing offline RL challenges due to their capability to mitigate the limitations of offline data through data generation using models. Prior research has demonstrated that introducing conservatism into the model or Q-function during policy optimization can effectively alleviate the prevalent distribution drift problem in offline RL. However, the investigation into the impacts of conservatism in reward estimation is still lacking. This paper proposes a novel model-based offline RL algorithm, Conservative Reward for model-based Offline Policy optimization (CROP), which conservatively estimates the reward in model training. To achieve a conservative reward estimation, CROP simultaneously minimizes the estimation error and the reward of random actions. Theoretical analysis shows that this conservative reward mechanism leads to a conservative policy evaluation and helps mitigate distribution drift. Experiments on D4RL benchmarks showcase that the performance of CROP is comparable to the state-of-the-art baselines. Notably, CROP establishes an innovative connection between offline and online RL, highlighting that offline RL problems can be tackled by adopting online RL techniques to the empirical Markov decision process trained with a conservative reward.     Preparation of Ag/HBP/PAN Nanofiber Web and Its Antimicrobial and Filtration Property

    To widen the application of nanofibers web in the field of medical health materials, a new Ag/amino-terminated hyperbranched polymer (HBP)/polyacrylonitrile (PAN) nanofiber web with excellent antimicrobial activity and filtration property was produced with Ag/HBP dispersion solution and PAN nanofiber. Ag/HBP dispersion solution was prepared with HBP as reducer and stabilizer, and Ag/HBP/PAN nanofiber was prepared by modifying electrospun PAN nanofiber with Ag/HBP aqueous solution. The characterization results showed that spherical Ag nanoparticles were prepared and they had a narrow distribution in HBP aqueous solution. The results of Ag/HBP/PAN nanofiber characterized with SEM and EDS showed that the content of silver nanoparticles on the surface of PAN nanofiber was on the increase when the treating temperature rose. The bacterial reduction rates of HBP-treated PAN nanofiber against S. aureus and E. coli were about 89%, while those of the Ag/HBP/PAN nanofiber against S. aureus and E. coli were 99.9% and 99.96%, respectively, due to the cooperative effects from the amino groups in HBP and Ag nanoparticles. Moreover, the small pores and high porosity in Ag/HBP/PAN nanofiber web resulted in high filtration efficiency (99.9%) for removing smaller particles (0.1 μm~0.7 μm), which was much higher than that of the gauze mask
