    Chemical composition of the essential oil of whole plant of Elsholtizia dense Benth and its anti-tumor effect on human hepatoma cells

    Purpose: To determine the chemical components of the essential oil of Elsholtizia dense in Sichuan Province and evaluate the effect of the oil on human hepatoma cells (SMMC-7721) in vitro.Methods: The essential oil was extracted using the modified steam-distillation  extraction method, and its chemical components were determined by gas  chromatography-mass spectrometry (GC-MS). The effect of the essential oil on proliferation of SMMC-7721 cells was studied by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) assay, with L02 and HeLa cells serving as control groups.Results: GC-MS results show that the essential oil of E. dense contains 40  components. Thirty seven components were identified and accounted for 98.39 % of the essential oil. The two main components were rosefuran epoxide (53.12 %) and 2-ethyl imidazole (29.8 %). The oil significantly inhibited cell proliferation in a concentration- and time-dependent manner (p < 0.05). SMMC-7721 cells were more inhibited than L02 and HeLa cells by the oil, with half maximal inhibitory concentration (IC50) values of 26.23 and 25.46 μg/mL after 8-h and 24-h treatments, respectively.Conclusion: Out of the 40 chemical components of the essential oil of E. dense, rosefuran epoxide and 2-ethyl imidazole were the most abundant.     α-Mangostin protects against myocardial ischemia reperfusion injury by suppressing the activation of HIF-1α

    Purpose: To investigate the cytoprotective effect of α-mangostin on myocardial tissues in ischemic rats, and the underlying mechanism.Methods: Histopathological changes in myocardial tissues were determined using inverted microscope. Protein expressions were measured by western blotting, while enzyme-linked immunosorbent assay (ELISA) was used to assay the expression levels of caspase-3, caspase-9 and caspase-8.Results: Treatment with α-mangostin (20 mg/kg) suppressed production of reactive oxygen species (ROS) and lipid peroxides in myocardial tissues of MI/R rats, and significantly alleviated MI/R injurymediated reduction in ATP levels in cardiac tissues (p < 0.05). α-Mangostin treatment of MI/R injury rats suppressed HIF-1α activation, and markedly elevated BNIP3 levels, relative to model group. Moreover, MI/R-induced cardiomyocyte apoptosis was significantly alleviated by α-mangostin treatment (p < 0.05). Treatment with α-mangostin also suppressed I/R-induced increases in caspase-8 and caspase-3 activation in myocardial tissues, improved Nrf-2 activation, and promoted HO-1 and GST levels in MI/R injury rats (p < 0.05).Conclusion: These results suggest that α-mangostin protects rat cardiac tissues from MI/R-induced oxidative damage via reduction of HIF-1α expression, inhibition of ROS generation and suppression of apoptosis.     Rare Copy Number Variants Identify Novel Genes in Sporadic Total Anomalous Pulmonary Vein Connection

    Total anomalous pulmonary venous connection (TAPVC) is a rare congenital heart anomaly. Several genes have been associated TAPVC but the mechanisms remain elusive. To search novel CNVs and candidate genes, we screened a cohort of 78 TAPVC cases and 100 healthy controls for rare copy number variants (CNVs) using whole exome sequencing (WES). Then we identified pathogenic CNVs by statistical comparisons between case and control groups. After that, we identified altogether eight pathogenic CNVs of seven candidate genes (PCSK7, RRP7A, SERHL, TARP, TTN, SERHL2, and NBPF3). All these seven genes have not been described previously to be related to TAPVC. After network analysis of these candidate genes and 27 known pathogenic genes derived from the literature and publicly database, PCSK7 and TTN were the most important genes for TAPVC than other genes. Our study provides novel candidate genes potentially related to this rare congenital birth defect (CHD) which should be further fundamentally researched and discloses the possible molecular pathogenesis of TAPVC

    Description of a new species of the genus Cultellus Schumacher, 1817 (Bivalvia, Pharidae) from the South China Sea, based on integrative taxonomy

    The present study describes a new species within the genus Cultellus Schumacher, 1817 collected from the South China Sea. An integrative taxonomic approach incorporating morphological comparisons, geometric morphometrics and genetic analyses was used to identify and differentiate the new species. Cultellus exilis sp. nov. is distinguished from its congeners by its slender, fragile and translucent valves, curved posteroventral margins and relatively large protractor scars. The geometric morphometric analyses, based on outlines data, indicated that samples of Cultellus exilis sp. nov. clustered together and were distinctly separated from other species. Multiple species delimitation results, based on the mitochondrial COI gene, support the separation of Cultellus exilis sp. nov. from its related congeners. Phylogenetic analyses of a nuclear (28S rRNA) and two mitochondrial (COI, 16S rRNA) genes using Maximum Likelihood and Bayesian Inference methods revealed that the species belongs to the genus Cultellus. The superfamily Solenoidea Lamarck, 1809, which includes the families Solenidae Lamarck, 1809 and Pharidae H. Adams & A. Adams, 1856, exhibits closer affinity to the family Hiatellidae Gray, 1824 than to Solecurtidae d’Orbigny, 1846. Furthermore, we found that the genus Siliqua was clustered alongside the genera Ensiculus and Phaxas as a sister clade, which contradicts the current systematics of the subfamily within the family Pharidae. It is used as food and medicine alone or in combination with other herbs in the early days. Its seeds have become one of the most popular Chinese herbal medicines. The coixan was extracted from Coix seed, and it was administered to diabetic mice. After five weeks of treatment, it was found to have some role in hypoglycemic. This study provides a good application prospect for Chinese medicine treatment of diabetes

    Bus ridership and its determinants in Beijing: A spatial econometric perspective

    Understanding the temporal and spatial dynamics and determinants of public transport ridership play an important role in urban planning. Previous studies have focused on exploring the determinants at the station level using global models, or a local model, geographically weighted regression (GWR), which cannot reveal spatial autocorrelation at the global level. This study explores the factors affecting bus ridership considering spatial autocorrelation using the spatial Durbin model (SDM). Taking the community in Beijing as the basic study unit, this study aims to explore the temporal and spatial dynamics of bus ridership and identify its key determinants considering neighboring effects. The results show the following: (1) The temporal dynamics are quite distinct on weekdays and weekends as well as at different time slots of the day. (2) The spatial patterns of bus ridership varied across different time slots, and the hot areas are mainly located near the central business district (CBD), transport hubs, and residential areas. (3) Key determinants of bus ridership varied across weekends and weekdays and varied at different time slots per day. (4) The spatial neighboring effects had been verified. This study provides a common analytical framework for analyzing the spatiotemporal dynamics and determinants of bus ridership at the community level

    Analysis of Interpolation Methods in the Validation of Backscattering Coefficient Products

    Validation is the basis of synthetic aperture radar (SAR) image quantification applications. Based on the point target of the field site, the radiation characteristics of the backscattering coefficient image can be used to optimize the SAR imaging, and the product production system can be more closely targeted, to ensure the image product accuracy in the actual quantification application. In this study, the validation of the backscattering coefficient image was examined using calibrators, and the radiometric properties of the image were evaluated by extracting the radar cross-section of each point target. Bilinear interpolation and fast Fourier transform (FFT) interpolation methods were introduced for the local area interpolation of point targets, and the two methods were compared from the perspective of response function imaging and validation accuracy. The results show that the FFT interpolation method is more favorable for validating the backscattering coefficient

    Integrated analysis reveals potential significance of FKBP5 in the prognosis and immunity of osteoarthritis and pan-cancer

    Background: The high disability rate of osteoarthritis (OA), a joint disease with an insidious onset and widespread effects, places a heavy financial burden on patients, families, and society. Traditional diagnostic approaches, including radiology and physical examination, cannot achieve early-stage screening of OA and thus, miss early intervention for patients. Therefore, the need of biomarkers for the early diagnosis of OA is crucial. Results: A total of 390 differentially expressed genes (DEGs) were identified from the training set, and 1077 key module genes were found by constructing a weighted gene co-expression network, and 161 key genes were obtained as a result. Four diagnostic marker genes highly associated with OA were screened for key genes using machine learning algorithms, and the resulting nomogram model showed excellent predictive power and clinical value. After further background studies, immune infiltration and functional enrichment analysis, we found that FKBP5 may play an important role in the prognosis and immune infiltration of multiple cancers, and this hypothesis was verified by pan-cancer analysis. Conclusions: We screened four diagnostic marker genes (FKBP5, EPYC, KLF9 and PDZRN4) that are highly associated with OA. And this led to a diagnostic model, which was assessed to have good predictive power and clinical value.
