The negative effect of visual acuity for growing children led by widely use of IC device such as mobile phone is concerned today. Heavy use of electronic device such as PC in early age will bring poor visual acuity, and an inconvenience in exercise caused by wearing glasses. In this study, we used wearable devise to search relations between daily lives of children and a decrease in vision in add to a preexisting questionnaire. Compared with the duration of watching TV and reading books of PVA( Poor visual acuity) and NPVA(Non poor visual acuity), PVA students were turned out to be using more time in doing, which supports previous studies. The relevance to visual acuity and sleep was clarified to be significantly shorter in NPVA in terms of nocturnal awakening, and also the average of a sleep and a deep sleep duration were longer in NPVA. No significant difference was observed due to the small parameter. From now on, the further work is needed to examine child's sleep conscientiously, including enriching health guidance for sight maintenance

In the past several years, a number of studies have been carried out on the behavioral and reproductive ecology of the Japanese giant salamander Andrias japonicus, but the age and longevity of A. japonicus has not yet been studied. In this study, we attempted to establish an age determination method using specimens (age: 1 to 11 years old) from Hiroshima City Asa Zoological Park that lived and died in captivity. The cross sections of phalangeal bones were nearly circular in shape, and hematoxylinophilic lines that were interpreted as lines of arrested growth (LAGs) were observed in the periosteal tissue; this suggests that this technique can be used to estimate the age of A. japonicus. The number of LAGs was one less than the number of winters that each individual experienced. We could observe LAGs in both frozen and 10% formalin specimens. LAGs could be confirmed even for specimens that had been fixed in formalin for up to 30 years. By using this method, it was suggested that the lifespan of this species could be determined from specimens existing in museums, zoos, and aquariums worldwide. It also showed potential for providing important conservation information, such as generation time and age structure of populations in the field. This work supported by JSPS KAKENHI Grant Numbers JP 26870400, JP 17K 01209

miR-155, a Modulator of FOXO3a Protein Expression, Is Underexpressed and Cannot Be Upregulated by Stimulation of HOZOT, a Line of Multifunctional Treg

MicroRNAs (miRNAs) play important roles in regulating post-transcriptional gene repression in a variety of immunological processes. In particular, much attention has been focused on their roles in regulatory T (Treg) cells which are crucial for maintaining peripheral tolerance and controlling T cell responses. Recently, we established a novel type of human Treg cell line, termed HOZOT, multifunctional cells exhibiting a CD4+CD8+ phenotype. In this study, we performed miRNA profiling to identify signature miRNAs of HOZOT, and therein identified miR-155. Although miR-155 has also been characterized as a signature miRNA for FOXP3+ natural Treg (nTreg) cells, it was expressed quite differently in HOZOT cells. Under both stimulatory and non-stimulatory conditions, miR-155 expression remained at low levels in HOZOT, while its expression in nTreg and conventional T cells remarkably increased after stimulation. We next searched candidate target genes of miR-155 through bioinformatics, and identified FOXO3a, a negative regulator of Akt signaling, as a miR-155 target gene. Further studies by gain- and loss-of-function experiments supported a role for miR-155 in the regulation of FOXO3a protein expression in conventional T and HOZOT cells

The deterioration of children’s eyesight is on the rise. Children's lives have changed in a short period of time, such as an increase in blue light due to the use of electronic devices. It is necessary to consider non-traditional content of health guidance for prevention. The accurate visual acuity test is important to examine the effects of visual acuity and environmental or living factors. We examined the accuracy of the visual acuity test with the three stage Landolt ring for children from the relationship with the eye axial length. After performing a normal detailed visual acuity test of the children and the correlation analysis with the eye axial length, we examined the distribution of the eye axis according to the results of the three-stage visual acuity test by the Landolt ring. The number of children measured eye axial length was 13. The correlation coefficient between detailed visual acuity test and eye axial length was negative. According to the results of the three-stage visual acuity test, the distribution of the eye axis is good for the right eye: the average eye axis of “A” was 23.44 mm, moderate: the average of “B” was 22.58 mm. The average of the left eye axis for “A” children is 23.34 mm, for “B” is 23.08 mm, and for “C” is 23.7 mm. The correlation coefficient between the detailed visual acuity test and the eye axial length was negative on both sides, namely a negative correlation between the visual acuity and the eye axial length. There was a possibility of not correspondence between the three-stage visual test and the eye axial length. In the case of children, it was considered that their visual performance and answers were different because of their mood and physical condition on the day

Structural Basis for Receptor-Mediated Selective Autophagy of Aminopeptidase I Aggregates

SummarySelective autophagy mediates the degradation of various cargoes, including protein aggregates and organelles, thereby contributing to cellular homeostasis. Cargo receptors ensure selectivity by tethering specific cargo to lipidated Atg8 at the isolation membrane. However, little is known about the structural requirements underlying receptor-mediated cargo recognition. Here, we report structural, biochemical, and cell biological analysis of the major selective cargo protein in budding yeast, aminopeptidase I (Ape1), and its complex with the receptor Atg19. The Ape1 propeptide has a trimeric coiled-coil structure, which tethers dodecameric Ape1 bodies together to form large aggregates. Atg19 disassembles the propeptide trimer and forms a 2:1 heterotrimer, which not only blankets the Ape1 aggregates but also regulates their size. These receptor activities may promote elongation of the isolation membrane along the aggregate surface, enabling sequestration of the cargo with high specificity

The giant salamander Andrias japonicas, which is endemic to Japan, is ranked as vulnerable. It is designated as a national special natural monument. Information on the giant salamander in Higashi-Hiroshima city is extremely scarce. To determine the current habitat of A. japonicas, we surveyed the Mukunashi River including areas where the giant salamander had lived in the past. Our surveys did find 21 adults and 10 larvae of A. japonicas within a two year period (from 2011 to 2013). However, we did not discover any young individuals of total length of 50 mm - 600 mm. This suggests that larval recruitment is not successful. This paper specifically reports that a critical situation exists for the giant salamander in the Mukunashi River in Higashi-Hiroshima City. And the publicity work aiming at conservation and its result are also reported
