Recent evidence indicates that Gram-negative bacteria-derived extracellular vesicles (EVs) in indoor dust can evoke neutrophilic pulmonary inflammation, which is a key pathology of chronic obstructive pulmonary disease (COPD). Escherichia coli is a ubiquitous bacterium present in indoor dust and secretes nanometer-sized vesicles into the extracellular milieu. In the current study, we evaluated the role of E. coli-derived EVs on the development of COPD, such as emphysema. E. coli EVs were prepared by sequential ultrafiltration and ultracentrifugation. COPD phenotypes and immune responses were evaluated in C57BL/6 wild-type (WT), IFN-gamma-deficient, or IL-17A-deficient mice after airway exposure to E. coli EVs. The present study showed that indoor dust from a bed mattress harbors E. coli EVs. Airway exposure to E. coli EVs increased the production of proinflammatory cytokines, such as TNF-alpha and IL-6. In addition, the repeated inhalation of E. coli EVs for 4 wk induced neutrophilic inflammation and emphysema, which are associated with enhanced elastase activity. Emphysema and elastase activity enhanced by E. coli EVs were reversed by the absence of IFN-gamma or IL-17A genes. In addition, during the early period, lung inflammation is dependent on IL-17A and TNF-alpha, but not on IFN-gamma, and also on TLR4. Moreover, the production of IFN-gamma is eliminated by the absence of IL-17A, whereas IL-17A production is not abolished by IFN-gamma absence. Taken together, the present data suggest that E. coli-derived EVs induce IL-17A-dependent neutrophilic inflammation and thereby emphysema, possibly via upregulation of elastase activity.X111613Ysciescopu

Effect of different seeding methods on green manure biomass, soil properties and rice yield in rice-based cropping systems

The cultivation of green manure crops plays an important role in soil quality and the sustainability of agricultural systems. Field experiments were conducted during one season (2007/2008) to evaluate the effects of different seeding methods on the biomass and N production of hairy vetch (Vicia villosa) and barley (Hordeum vuglare). The effects of treatments on rice yield and its components were also investigated. Specifically, the following four treatments were evaluated: broadcasting before rice harvesting (BBRH), partial tillage seeding (PTS), group seeding (GS) and drill seeding (DS). The overall results showed the following ranking of biomass and nitrogen production of hairy vetch by different seeding methods: BBRH > PTS > DS > GS. Additionally, biomass and nitrogen production of barley was lower than those of hairy vetch at all tested seeding treatments. BBRH and PTS treatments led to an increase in soil bulk density, porosity and soil organic content. The application of green manure with BBRH and PTS treatments led to a significant increase in rice yield (5,330 and 5,320 kg ha-1) when compared to conventional fertilization. Based on the results, BBRH and PTS are good practices for production of green manure in paddy soil. Chemical fertilizers can be replaced with hairy vetch as green manure in rice-based cropping systems.Key words: Green manure, rice, biomass, soil property, seeding method

VIABILITY, FATTY ACID COMPOSITION, AND STRUCTURE OF THE CORALLINE ALGA CORALLINA PILULIFERA

The decrease in the seaweed flora in some rocky areas, known as algal whitening or barren ground, is associated with some species of coralline algae. To determine the biological characteristics of a representative species of branched coralline alga, the number of medullary tiers was counted and ranged from 12 to 16. The 18S rDNA, psbA, and rbcL genes were used to confirm the identification of Corallina pilulifera. Measuring viability using triphenyl tetrazolium chloride showed highly viability from December to January. Cultural conditions of 16 C, 16 h light:8 h dark cycle, and 40 mu E m(-2) s(-1) light intensity were optimal for maintaining the viability of the coralline alga for up to three days. The fatty acids included 31.4% omega-3 eicosapentaenoic acid. Scanning electron microscopy of the surface structure revealed unique round wells about 7.9 +/- 1.3 mu m in diameter. The coralline alga, preventing fleshy seaweeds, may be used as a potential template for the creation of new environmentally friendly biomimetic antifouling material against the attachment of soft foulants, especially micro- and macroalgae.X111Ysciescopu

Exosomes known as nano-sized extracellular vesicles attracted recent interests due to their potential usefulness in drug delivery. Amid remarkable advances in biomedical applications of exosomes, it is crucial to understand in vivo distribution and behavior of exosomes. Here, we developed a simple method for radiolabeling of macrophage-derived exosome-mimetic nanovesicles (ENVs) with Tc-99m-HMPAO under physiologic conditions and monitored in vivo distribution of Tc-99m-HMPAO-ENVs using SPECT/CT in living mice. ENVs were produced from the mouse RAW264.7 macrophage cell line and labeled with Tc-99m-HMPAO for 1 hr incubation, followed by removal of free Tc-99m-HMPAO. SPECT/CT images were serially acquired after intravenous injection to BALB/c mouse. When ENVs were labeled with Tc-99m-HMPAO, the radiochemical purity of Tc-99m-HMPAO-ENVs was higher than 90% and the expression of exosome specific protein (CD63) did not change in Tc-99m-HMPAO-ENVs. Tc-99m-HMPAOENVs showed high serum stability (90%) which was similar to that in phosphate buffered saline until 5 hr. SPECT/CT images of the mice injected with Tc-99m-HMPAO-ENVs exhibited higher uptake in liver and no uptake in brain, whereas mice injected with Tc-99m-HMPAO showed high brain uptake until 5 hr. Our noninvasive imaging of radiolabeled-ENVs promises better understanding of the in vivo behavior of exosomes for upcoming biomedical application.114327Ysciescopu

The enhanced expression of IL-17-secreting T cells during the early progression of atherosclerosis in ApoE-deficient mice fed on a western-type diet

Atherosclerosis is a chronic progressive inflammatory disorder and the leading cause of cardiovascular mortality. Here we assessed the dynamic changes of T-cell-derived cytokines, such as inteferon (IFN)-gamma, interleukin (IL)-17 and IL-4, during the progression of atherosclerosis in apolipoprotein E-null (ApoE(-/-)) mice, to understand the role of immune responses in different stages of atherosclerosis. Male ApoE(-/-) mice were fed a high-fat, western-type diet (WD: 21% lipid, 1.5% cholesterol) after 5 weeks of age and were compared with C57BL/6 wild-type control mice fed a standard chow diet. Atherosclerotic lesions appeared in the aortic sinus of ApoE(-/-) mice 4 weeks after WD and the lesions progressed and occupied > 50% of the total sinus area 16 weeks after WD. Aortic IL-17 mRNA and protein expression started to increase in ApoE(-/-) mice after 4 weeks on the WD and peaked at around 8-12 weeks on the WD. In terms of systemic expression of T-cell-derived cytokines, IL-17 production from splenocytes after anti-CD3/CD28 stimuli increased from 4 weeks on the WD, peaked at 12 weeks and returned to control levels at 16 weeks. The production of IFN-gamma and IL-4 (Th1 and Th2 cytokines, respectively) from splenocytes was delayed compared with IL-17. Taken together, the present data indicate that Th17 cell response may be involved at an early stage in the development of atherosclerosis.11911Ysciescopu

The Bishop-Phelps-Bollobas theorem for operators on L-1(mu)

    Get PDF
In this paper we show that the Bishop-Phelps-Bollobas theorem holds for L(L-1(mu), L-1(v)) for all measures and v and also holds for L(L-1(mu), L-infinity(nu)) for every arbitrary measure mu and every localizable measure nu Finally, we show that the Bishop-Phelps-Bollobas theorem holds for two classes of bounded linear operators from a real L-1(mu) into a real C(K) if mu is a finite measure and K is a compact Hausdorff space. In particular, one of the classes includes all Bochner representable operators and all weakly compact operators. (c) 2014 Elsevier Inc. All rights reserved.X1174Ysciescopu
