    1-D modelling and 3-D simulation of confined bubble formation and formation and pressure fluctuations during flow boiling in a microchannel with a rectangular cross-section of high aspect ratio

    Copyright @ 2009 ASMEA simple 1-D model with low requirements for computing time is required to investigate parametric influences on the potentially adverse effects of pressure fluctuations driven by confined vapour bubble growth in microchannel evaporative cooling systems operating at high heat fluxes. A model is developed in this paper for the particular conditions of a channel of rectangular cross-section with high aspect ratio with a constant inlet flow rate (zero upstream compressibility). (The model will later be extended to the conditions of finite upstream compressibility that lead to transient flow reversal). Some parametric trends predicted by the model are presented. The simplifying assumptions in the model are examined in the light of a 3-D simulation by a commercial CFD code, described in an accompanying paper by the same authors. The predictions of pressure changes are in reasonable agreement. Fattyacid-binding protein 4 (FABP4) and adiponectin are both adipocyte-derived cytokines closely related with inflammation pathways. The FABP4 exhibits a pro-inflammatory property while adiponectin shows anti-inflammatory effect. The study aimed to assess the expression of FABP4 and adiponectin in paired blood and synovial fluid from OA patients, and to examine whether the infrapatellar fat pad (IPFP) from OA subjects would release FABP4 and adiponectin similar to that of donor-matched subcutaneous adipose tissue (ScAT). Materials and Methods: Plasma, synovial fluid, ScAT, and IPFP of 20 OA patients (4 males and 16 females) were collected during total knee arthroplasty. Levels of FABP4 and adiponectin were measured in plasma, synovial fluid, and fat conditioned media using ELISA (AIS, HKU). Results: Levels of FABP4 were significantly higher in IPFP media than ScAT (p = 0.015), while the same were significantly higher in synovial fluid than in plasma (p < 0.001). There were no significant differences of adiponectin levels between IPFP and ScAT media (p = 0.737). Plasma levels of adiponectin were significantly higher than synovial fluid (p < 0.001). Discussion and Conclusion: The OA patients exhibit an inflammatory state in local environment (IPFP and synovial fluid) compared with systemic environment (ScAT and plasma). The IPFP may play a key role in OA inflammation and may be an important resource of inflammatory mediators in synovial fluid.postprin

    Novel OCRL mutations in children with Lowe Syndrome

    Background: Lowe syndrome is a rare X-linked recessive hereditary disease caused by mutations of the OCRL gene, which encodes an inositol polyphosphate-5-phosphatase. The disease is clinically characterized by congenital cataracts, psychomotor retardation, and proximal tubulopathy. Methods: We retrospectively reviewed three unrelated Chinese patients with Lowe syndrome, clinically diagnosed by the abnormalities of eyes, nervous system, and kidneys. Genetic analysis of the OCRL gene was done for the three patients as well as their family members. Results: Three OCRL gene mutations were detected in our study. Two of the mutations, g.1897delT in exon 18 (patient 1) and g.1470delG in exon 15 (patient 2), were novel. A missense mutation (p.Y513C) in exon 15, which had been reported previously, was found in patient 3. The mothers of all patients were heterozygous carriers of the respective mutations. Conclusion

    Platelet-activating factor acetylhydrolase (PAF-AH) is an enzyme that catalyzes the hydrolysis of platelet-activating factor (PAF). A homolog of alpha subunit of PAF-AH(Ib) from Antheraea pernyi (GuΓ©rin-MΓ©neville) (Lepidoptera: Saturniidae) (ApPAFAHIbΞ±) was isolated and characterized. The obtained cDNA sequence was 1843 base pairs (bp) long with an open reading frame (ORE) of 678 bp encoding 225 amino acids. The predicted amino acid sequence shared several conserved features of PAF-AHs of other organisms, and revealed 88, 60, and 46% identity with the homologues of Bombyx mori, Drosophila melanogaster, and Homo sapiens, respectively. Phylogenetic analysis indicated that lepidopteran PAFAHIbΞ±s including ApPAFAHIbΞ± might be a new member of the PAF-AHs family of insects. Reverse transcriptase polymerase chain reaction (RT-PCR) analysis showed that the ApPAFAHIbΞ± gene was transcribed at four developmental stages and expressed in all tissues tested

    Recepteur d'Origine Nantais (RON) is a distinct receptor tyrosine kinase in the c-met proto-oncogene family. We examined the mutational and expression patterns of RON in eight human uroepithelial cell lines. Biological effects of RON overexpression on cancer cells were investigated in vitro, and the prognostic significance of RON and/or c-met protein (MET) expression was analysed in a bladder cancer cohort (n=183). There was no evidence of mutation in the kinase domain of RON. Overexpression of RON using an inducible Tet-off system induced increased cell proliferation, motility, and antiapoptosis. Immunohistochemical analysis showed that RON was overexpressed in 60 cases (32.8%) of primary tumours, with 14 (23.3%) showing a high level of expression. Recepteur d'Origine Nantais expression was positively associated with histological grading, larger size, nonpapillary contour, and tumour stage (all P<0.01). In addition, MET was overexpressed in 82 cases (44.8%). Co-expressed RON and MET was significantly associated with decreased overall survival (P=0.005) or metastasis-free survival (P=0.01) in 35 cases (19.1%). Recepteur d'Origine Nantais-associated signalling may play an important role in the progression of human bladder cancer. Evaluation of RON and MET expression status may identify a subset of bladder-cancer patients who require more intensive treatment

    Mitotic Illegitimate Recombination Is a Mechanism for Novel Changes in High-Molecular-Weight Glutenin Subunits in Wheat-Rye Hybrids

    Wide hybrids can have novel traits or changed expression of a quantitative trait that their parents do not have. These phenomena have long been noticed, yet the mechanisms are poorly understood. High-molecular-weight glutenin subunits (HMW-GS) are seed storage proteins encoded by Glu-1 genes that only express in endosperm in wheat and its related species. Novel HMW-GS compositions have been observed in their hybrids. This research elucidated the molecular mechanisms by investigating the causative factors of novel HMW-GS changes in wheat-rye hybrids. HMW-GS compositions in the endosperm and their coding sequences in the leaves of F1 and F2 hybrids between wheat landrace Shinchunaga and rye landrace Qinling were investigated. Missing and/or additional novel HMW-GSs were observed in the endosperm of 0.5% of the 2078 F1 and 22% of 36 F2 hybrid seeds. The wildtype Glu-1Ax null allele was found to have 42 types of short repeat sequences of 3-60 bp long that appeared 2 to 100 times. It also has an in-frame stop codon in the central repetitive region. Analyzing cloned allele sequences of HMW-GS coding gene Glu-1 revealed that deletions involving the in-frame stop codon had happened, resulting in novel ∼1.8-kb Glu-1Ax alleles in some F1 and F2 plants. The cloned mutant Glu-1Ax alleles were expressed in Escherichia coli, and the HMW-GSs produced matched the novel HMW-GSs found in the hybrids. The differential changes between the endosperm and the plant of the same hybrids and the data of E. coli expression of the cloned deletion alleles both suggested that mitotic illegitimate recombination between two copies of a short repeat sequence had resulted in the deletions and thus the changed HMW-GS compositions. Our experiments have provided the first direct evidence to show that mitotic illegitimate recombination is a mechanism that produces novel phenotypes in wide hybrids

    Strain induced exciton fine-structure splitting and shift in bent ZnO microwires

    Lattice strain is a useful and economic way to tune the device performance and is commonly present in nanostructures. Here, we investigated for the first time the exciton spectra evolution in bent ZnO microwires along the radial direction via high spatial/energy resolution cathodeluminescence spectroscopy at 5.5β€…K. Our experiments show that the exciton peak splits into multi fine peaks towards the compressive part while retains one peak in the tensile part and the emission peak displays a continuous blue-shift from tensile to compressive edges. In combination with first-principles calculations, we show that the observed NBE emission splitting is due to the valence band splitting and the absence of peak splitting in the tensile part maybe due to the highly localized holes in the A band and the carrier density distribution across the microwire. Our studies may pave the way to design nanophotonic and electronic devices using bent ZnO nanowires

    BMI-1 Autoantibody as a New Potential Biomarker for Cervical Carcinoma

    BMI-1 is overexpressed in a variety of cancers, which can elicit an immune response leading to the induction of autoantibodies. However, BMI-1 autoantibody as a biomarker has seldom been studied with the exception of nasopharyngeal carcinoma. Whether BMI-1 autoantibodies can be used as a biomarker for cervical carcinoma is unclear. In this study,BMI-1 proteins were isolated by screening of a T7 phage cDNA library from mixed cervical carcinoma tissues. We analyzed BMI-1 autoantibody levels in serum samples from 67 patients with cervical carcinoma and 65 controls using ELISA and immunoblot. BMI-1 mRNA or protein levels were over-expressed in cervical carcinoma cell lines. Immunoblot results exhibited increased BMI-1 autoantibody levels in patient sera compared to normal sera. Additionally, the results for antibody affinity assay showed that there was no difference between cervical polyps and normal sera of BMI-1 autoantibody levels, but it was significantly greater in patient sera than that in normal controls (patient 0.827Β±0.043 and normal 0.445Β±0.023; P<0.001). What's more, the levels of BMI-1 autoantibody increased significantly at stage I (0.672Β±0.019) compared to normal sera (P<0.001), and levels of BMI-1 autoantibodies were increased gradually during the tumor progression (stage I 0.672Β±0.019; stage II 0.775 Β±0.019; stage III 0.890 Β±0.027; stage IV 1.043Β±0.041), which were significantly correlated with disease progression of cervical carcer (P<0.001). Statistical analyses using logistic regression and receiver operating characteristics (ROC) curves indicated that the BMI-1 autoantibody level can be used as a biomarker for cervical carcinoma (sensitivity 0.78 and specificity 0.76; AUCβ€Š=β€Š0.922). In conclusion, measuring BMI-1 autoantibody levels of patients with cervical cancer could have clinical prognostic value as well as a non-tissue specific biomarker for neoplasms expressing BMI-1
