    Chronic administration of aripiprazole activates GSK3β-dependent signalling pathways, and up-regulates GABA\u3csub\u3eA\u3c/sub\u3e receptor expression and CREB1 activity in rats

    Aripiprazole is a D2-like receptor (D2R) partial agonist with a favourable clinical profile. Previous investigations indicated that acute and short-term administration of aripiprazole had effects on PKA activity, GSK3β-dependent pathways, GABAA receptors, NMDA receptor and CREB1 in the brain. Since antipsychotics are used chronically in clinics, the present study investigated the long-term effects of chronic oral aripiprazole treatment on these cellular signalling pathways, in comparison with haloperidol (a D2R antagonist) and bifeprunox (a potent D2R partial agonist). We found that the Akt-GSK3β pathway was activated by aripiprazole and bifeprunox in the prefrontal cortex; NMDA NR2A levels were reduced by aripiprazole and haloperidol. In the nucleus accumbens, all three drugs increased Akt-GSK3β signalling; in addition, both aripiprazole and haloperidol, but not bifeprunox, increased the expression of Dvl-3, β-catenin and GABAA receptors, NMDA receptor subunits, as well as CREB1 phosphorylation levels. The results suggest that chronic oral administration of aripiprazole affects schizophrenia-related cellular signalling pathways and markers (including Akt-GSK3β signalling, Dvl-GSK3β-β-catenin signalling, GABAA receptor, NMDA receptor and CREB1) in a brain-region-dependent manner; the selective effects of aripiprazole on these signalling pathways might be associated with its unique clinical effects

    Mobility analysis of generalized angulated scissor-like elements with the reciprocal screw theory

    This paper deals with the planar closed loop linkage consisting of a series of scissor-like elements connected by revolute joints. Because every generalized angulated element (GAE) subtends a constant angle during the motion, every angulated link was assumed as a PRRP linkage which has two prismatic joints and two revolute joints. Therefore, the two PRRP linkages of the GAE are individually movable with a single degree of freedom. The mobility of two types of GAEs was investigated with the method based on the screw theory. It has been proven that both types of GAEs are movable because the terminal constraints exerted to the common joint by the two linkages are equal

    Motion analysis of a foldable barrel vault based on regular and irregular Yoshimura Origami

    This paper investigates the geometry of a foldable barrel vault with Yoshimura Origami patterns during the motion. On the base of the geometry analysis of the origami unit, the radius, span, rise, and longitudinal length of the foldable barrel vault with regular Yoshimura Origami pattern in all configurations throughout the motion are determined. The results show that the radius of curvature and the span increase during deployment. But the rise increases first, followed by a decrease with increasing fold angle. Furthermore, the influence of the apex angle of the origami unit and the numbers of triangular plates in the span direction on the geometric parameters is also investigated. Finally, the method to obtain the rise and span of the barrel vault with irregular origami pattern is also given

    MyHealthAvatar lifestyle management support for cancer patients

    MyHealthAvatar (MHA) is built on the latest information and communications technology with the aim of collecting lifestyle and health data to promote citizen's wellbeing. According to the collected data, MHA offers visual analytics of lifestyle data, contributes to individualised disease prediction and prevention, and supports healthy lifestyles and independent living. The iManageCancer project aims to empower patients and strengthen self-management in cancer diseases. Therefore, MHA has contributed to the iManageCancer scenario and provides functionality to the iManageCancer platform in terms of its support of lifestyle management of cancer patients by providing them with services to help their cancer management. This paper presents two different MHA-based Android applications for breast and prostate cancer patients. The components in these apps facilitate health and lifestyle data presentation and analysis, including weight control, activity, mood and sleep data collection, promotion of physical exercise after surgery, questionnaires and helpful information. These components can be used cooperatively to achieve flexible visual analysis of spatiotemporal lifestyle and health data and can also help patients discover information about their disease and its management

    Decreased density of serotonin 2A receptors in the superior temporal gyrus in schizophrenia - a postmortem study

    The superior temporal gyrus (STG) is strongly implicated in the pathophysiology of schizophrenia,particularly with regards to auditory hallucinations. In this study, using in situ quantitative autoradiography in postmortem tissue, we investigated the binding of the [3H]ketanserin to 5-HT2A receptors and [3H] mesulergine to 5-HT2C receptors in the left STG of 8 male schizophrenic patients compared to 8 control subjects. A strong [3H]ketanserin binding was observed in the STG, however there was a very weak [3H] mesulergine binding in the STG. A significant decrease in binding of [3H]ketanserin was clearly observed in schizophrenia patients in comparison with control subjects. There were no significant correlations between 5-HT2A binding density and age, postmortem intervals, or brain pH. These results suggest that the alterations of the 5-HT2A receptors contribute to the pathophysiology of the STG in schizophrenia. Furthermore, there is a clear tendency for a positive correlation between 5-HT2A and muscarinic M1 receptor bindings, and for negative correlations between 5-HT2A and GABAA receptor bindings and between muscarinic M1 and GABAA receptor bindings. This provides a possible mechanism of auditory hallucinations through interactions between 5-HT2A, acetylcholine muscarinic and GABA transmissions in the STG in schizophrenia
