    Role of MicroRNA-26b in Glioma Development and Its Mediated Regulation on EphA2

    BACKGROUND: MicroRNAs (miRNAs) are short, non-coding RNAs that regulate the expression of multiple target genes. Deregulation of miRNAs is common in human tumorigenesis. Low level expression of miR-26b has been found in glioma cells. However, its underlying mechanism of action has not been determined. METHODOLOGY/PRINCIPAL FINDINGS: Real-time PCR was employed to measure the expression level of miR-26b in glioma patients and cells. The level of miR-26b was inversely correlated with the grade of glioma. Ectopic expression of miR-26b inhibited the proliferation, migration and invasion of human glioma cells. A binding site for miR-26b was identified in the 3'UTR of EphA2. Over-expression of miR-26b in glioma cells repressed the endogenous level of EphA2 protein. Vasculogenic mimicry (VM) experiments were performed to further confirm the effects of miR-26b on the regulation of EphA2, and the results showed that miR-26b inhibited the VM processes which regulated by EphA2. SIGNIFICANCE: This study demonstrated that miR-26b may act as a tumor suppressor in glioma and it directly regulates EphA2 expression. EphA2 is a direct target of miR-26b, and the down-regulation of EphA2 mediated by miR-26b is dependent on the binding of miR-26b to a specific response element of microRNA in the 3'UTR region of EphA2 mRNA

    Plant Phenotypic Traits Eventually Shape Its Microbiota: A Common Garden Test

    Plant genotype drives the development of plant phenotypes and the assembly of plant microbiota. The potential influence of the plant phenotypic characters on its microbiota is not well characterized and the co-occurrence interrelations for specific microbial taxa and plant phenotypic characters are poorly understood. We established a common garden experiment, which quantifies prokaryotic and fungal communities in the phyllosphere and rhizosphere of six spruce (Picea spp.) tree species, through Illumina amplicon sequencing. We tested for relationships between bacterial/archaeal and fungal communities and for the phenotypic characters of their plant hosts. Host phenotypic characters including leaf length, leaf water content, leaf water storage capacity, leaf dry mass per area, leaf nitrogen content, leaf phosphorous content, leaf potassium content, leaf δ13C values, stomatal conductance, net photosynthetic rate, intercellular carbon dioxide concentration, and transpiration rate were significantly correlated with the diversity and composition of the bacterial/archaeal and fungal communities. These correlations between plant microbiota and suites of host plant phenotypic characters suggest that plant genotype shape its microbiota by driving the development of plant phenotypes. This will advance our understanding of plant-microbe associations and the drivers of variation in plant and ecosystem function

    At present, corporate social responsibility has been widely mentioned by the international society, especially platform enterprises. For a platform that assumes social responsibilities, consumer surplus is a rather critical aspect, and product quality is one of the most important factors directly related to consumers. This paper studies a supply chain consisting of a manufacturer and a retailing platform, in which the retailing platform procures products from the manufacturer. The manufacturer produces the products and decides the product quality. We consider two channel structures of the manufacturer and the retailing platform in the reseller mode and marketplace mode. Based on the model analysis and discussions, we obtain some managerial insights that are helpful in commercial practice. For the retailing platform, it has to suffer a loss in economic profit to care more about consumer surplus and become a social responsibility platform. In addition, its social responsibility plays different roles in different channel structures. In the marketplace mode, a social responsibility retailing platform helps to improve product quality. In the reseller mode, the retailing platform’s social responsibility does not make a change in product quality. Furthermore, the product quality in the reseller mode is always higher than that in the marketplace mode. From the perspective of economic profits, the manufacturer obtains higher profits in the reseller mode than the marketplace mode.     Finite volume element methods for two-dimensional three-temperature radiation diffusion equations

    \u3cp\u3eTwo-dimensional three-temperature (2-D 3-T) radiation diffusion equations are widely used to approximately describe the evolution of radiation energy within a multi-material system and explain the exchange of energy among electrons, ions and photons. Their highly nonlinear, strong discontinuous and tightly coupled phenomena always make the numerical solution of such equations extremely challenging. In this paper, we construct two finite volume element schemes both satisfying the discrete conservation property. One of them can well preserve the positivity of analytical solutions, while the other one does not satisfy this property. To fix this defect, two as repair techniques are designed. In addition, as the numerical simulation of 2-D 3-T equations is very time consuming, we also devise a mesh adaptation algorithm to reduce the cost. Numerical results show that these new methods are practical and efficient in solving this kind of problems.\u3c/p\u3

    Data-Driven Sustainable Supply Chain Decision Making in the Presence of Low Carbon Awareness

    Low-carbon production is a vital solution for many environmental problems, as is consumers’ consciousness about carbon playing a more important role and ultimately passing upstream. Supply chains are eager to seek sustainable development via appropriate decision making with data-driven methods. Consistent with this aim, we investigated decisions toward lower carbon efforts and prices in a two-echelon supply chain via a game theoretical approach. The decision-making scenarios of decentralized, centralized, and cost-sharing contracts were investigated and compared. The results show that the level of improvement in environmental performance is positively correlated with the degree of cooperation between partners. Cooperation between partners would be even more significant with an increase in consumers’ low carbon awareness. Furthermore, cost-sharing contracts improve the performance of the entire supply chain compared with decentralized cases. Finally, we implemented numerical experiments to verify the modeling results. Therefore, this study provides theoretical support toward sustainable operations for supply chains concerning low carbon awareness

    Sand Blocking Mechanisms and Productivity Analysis of Slant Well in Siltstone Reservoirs

    The siltstone reservoir with many small layers of pay zones is usually produced by slant holes. However, severe sand blockages take place when the siltstone reservoir is developed by slant holes. Currently, the sand blocking mechanisms and the effect of sand blocking on productivity of slant well are still challenges for engineers and scholars. In this paper, based on the existing productivity model of inclined slant wells, the mechanical skin factor, which describes the effect of sand blocking on productivity, is proposed. Meanwhile, many experimental works have been done to investigate the sand blocking mechanisms in siltstone reservoirs. From the experimental work, it is concluded that with the increase of displacement PV number and displacement flow rate, the permeability of sand control system decreases by 40%. When solid particles enter the casing and block the gravel and sand control screen in the annulus, the skin factor increases sharply and the productivity decreases by more than 80%. Through the productivity calculation of multilayer sand control wells, it is considered that larger gravel packing radius can keep particles away from the well bore, which is helpful to ensure oil well productivity. Furthermore, the influence of differential filling radius on the fluid production capacity of each layer of sand control well is analyzed. It is proved that optimizing the filling radius of each layer can improve the production effect of mediate- and low-permeability layers. This method has been applied in Kendong #12 block. The daily oil production rate is increased by 9.61 t/day, and the oil recovery of this block is increased by 2.12%

    Pricing and Allotment in a Sea-Cargo Supply Chain with Reference Effect: A Dynamic Game Approach

    The contract between the carrier and forwarder is a long-term issue, and the repeated contract business makes the forwarder develop a reference point based on the contract prices, and this reference effect, to a large extent, affects the forwarder’s contract purchasing decisions. Based on that, this paper introduces the reference effect in the sea-cargo supply chain and studies a multiple-period contract problem between the carrier and the forwarder. It is found that when the capacity price in the spot market is less than the forwarder’s willingness-to-pay, the forwarder’s contract purchasing decision is not affected by the reference effect, only by the capacity price in the spot market, and the multiple-period contract problem can be simplified into a single-period game. In addition, the carrier’s optimal contract wholesale price approaches the capacity price in the spot market. Although, the forwarder’s contract purchasing decision depends upon the reference effect, it is difficult to derive the closed-form solution. Moreover, because of the risk in the spot market, the carrier tends to sell his/her capacity in the contract market. Finally, we employ the numerical simulation to study the carrier’s contract pricing decisions and the forwarder’s capacity purchasing decisions in two cases
