    Synthesis of Oleoylethanolamide Using Lipase

    An effective process for the enzymatic synthesis of oleoylethanolamide is described in this study. The process included purification of a commercial oleic acid product and then optimization of the reaction between the purified oleic acid and ethanolamine in the presence of hexane and a lipase. Under the optimal amidation reaction conditions identified, oleoylethanolamide was obtained with 96.6% purity. The synthesis was also conducted on a large scale (50 mmol of each of the reactants), and oleoylethanolamide purity and yield after crystallization purification were 96.1 and 73.5%, respectively. Compared to the previous studies, the current method of preparing high-purity oleoylethanolamide is more effective and economically feasible. The scalability and ease for such synthesis make it possible to study the biological and nutritional functions of the cannabinoid-like oleoylethanolamide in animal or human subjects

    The purposes are to monitor the nitrogen utilization efficiency of crops and intelligently evaluate the absorption of nutrients by crops during the production process. The research object is Chinese cabbage. The Chinese cabbage population with different agricultural parameters is constructed through different densities and nitrogen fertilizer application rates based on digital image processing technology, and an estimation NC (Nitrogen Content) model is established. The population is classified through the K-Means Clustering algorithm using the feature extraction method, and the Chinese cabbage population quality BPNN (Backpropagation Neural Network) model is constructed. The nonlinear mapping relationship between different agricultural parameters and population quality, and the contribution rate of each indicator, are studied. The nitrogen utilization of Chinese cabbage is monitored effectively. Results demonstrate that the proposed NC estimation model has correlation coefficients above 0.70 in different growth stages. This model can accurately estimate the NC of the Chinese cabbage population. The results of the Chinese cabbage population quality BPNN model show that the population planting density based on the seedling number is reasonable. The constructed population quality evaluation model has a high R2 value and a comparatively low RMSE (Root Mean Square Error) value for the quality evaluation of Chinese cabbage in different periods, showing that it applies to evaluate the population quality of Chinese cabbage in different growth stages. The constructed nitrogen utilization model and quality evaluation model can monitor the nutrient utilization of crops in different growth stages, ascertain the agricultural characteristics of other yield groups in different growth stages, and clarify the performance of agricultural parameters in different growth stages. The above results can provide some ideas for crop growth intelligent detection

    Non-Fickian Solute Transport in a Single Fracture of Marble Parallel Plate

    Accurate prediction of solute transport processes in a fracture aquifer is an important task not only for proper management of the groundwater but also for pollution control. A key issue of this task is how to accurately obtain the experimental data and to analyze the solute transport in fracture in subsurface hydrology, which would greatly help us to understand the releasing mechanism and transport of the solute in a fracture. In this study, a fracture experiment is conducted in a laboratory based on previous studies. The fracture used with a length of 60 cm and a width of 10 cm is sealed with glass glue to avoid leakage of tracer due to uneven fracture walls. The sodium chloride (NaCl) solute is injected from the left of the fracture. And an electrical conductivity monitoring system is installed on the right of the fracture. Then breakthrough curves (BTCs) of solute transport are fitted using the classical advection-dispersion equation (ADE) and the truncation power-law function (TPL) model in the package of the continuous time random walk (CTRW). The results show that the flow satisfies non-Darcian law in the experimental conditions, which can be better fitted using the Forchheimer equation and Izbash equation. The solute transport presents non-Fickian phenomena and shows a long tailing. The fitting results of the TPL model are far better than ADE in fitting the long tailing at three different flow velocities. Furthermore, electrical conductivity monitoring method not only is effective but also has an advantage of no disturbance to water and concentration fields in a fracture

    Diverse Krill Lipid Fractions Differentially Reduce LPS-Induced Inflammatory Markers in RAW264.7 Macrophages In Vitro

    Antarctic krill oil is an emerging marine lipid and expected to be a potential functional food due to its diverse nutrients, such as eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), phospholipids, astaxanthin and tocopherols. Although krill oil has been previously proved to have anti-inflammatory activity, there is little information about the relationship between its chemical compositions and anti-inflammatory activity. In this study, the RAW264.7 macrophages model was used to elucidate and compare the anti-inflammatory potential of different krill lipid fractions: KLF-A, KLF-H and KLF-E, which have increasing phospholipids, EPA and DHA contents but decreasing astaxanthin and tocopherols levels. Results showed that all the krill lipid fractions alleviated the inflammatory reaction by inhibition of production of nitric oxide (NO), release of tumor necrosis factor-&alpha; (TNF-&alpha;), interleukin (IL)-1&beta; and IL-6 and gene expression of proinflammatory mediators including TNF-&alpha;, IL-1&beta;, IL-6, cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS). In addition, KLF-E with the highest phospholipids, EPA and DHA contents showed the strongest inhibition effect on the LPS-induced proinflammatory mediator release and their gene expressions. The results would be helpful to provide powerful insights into the underlying anti-inflammatory mechanism of krill lipid and guiding the production of krill oil products with tailor-made anti-inflammatory activity

    Hsa_circ_0046264 up-regulated BRCA2 to suppress lung cancer through targeting hsa-miR-1245

    Abstract Objective Lung cancer had been leading mounts of deaths worldwide. Advances in genes microarray had helped human further understand genes and identify novel circular RNAs. This study aimed at investigating the biological functions and molecular mechanisms of hsa_circ_0046264 in lung cancer which may be helpful in lung cancer early diagnosis and clinical treatment. Methods Gene microarray data screened the differential gene of hsa_circ_0046264 and its downstream genes were found by bioinformatics analysis and verified by luciferase reporter assay. QRT-PCR and Western blot was used to detect the RNA and protein levels respectively. RNase R digestion confirmed the existences of circular RNA. Cell viability, invasion and apoptosis were determined by MTT assay, flow cytometry and DNA damage assay. Tumor formation in nude mice and immunohistochemistry proved the functions of hsa_circ_0046264 in vivo. Results Hsa_circ_0046264 and BRCA2 were down-regulated in lung cancer tissues while miR-1245 was up-regulated. Hsa_circ_0046264 induced apoptosis but inhibited proliferation and invasion of lung cancer cells through targeting miR-1245 to up-regulate BRCA2. Hsa_circ_0046264 inhibited the tumor growth in vivo. Conclusion Hsa_circ_0046264 was a tumor suppressor in lung cancer. Overexpression of hsa_circ_0046264 could up-regulate BRCA2 expression through down-regulating of miR-1245

    Knowledge of and Attitude to Nuclear Power among Residents around Tianwan Nuclear Power Plant in Jiangsu of China

    <p><b>Aims:</b> The aims of this paper were to determine the level of knowledge of and attitude to nuclear power among residents around Tianwan Nuclear power plant in Jiangsu of China.</p><p><b>Design:</b> A descriptive, cross-sectional design was adopted.</p><p><b>Participants:</b> 1,616 eligible participants who lived around the Tianwan nuclear power plant within a radius of 30km and at least 18 years old were recruited into our study and accepted epidemiological survey.</p><p><b>Methods:</b> Data were collected through self-administered questionnaires consisting of a socio-demographic sheet. Inferential statistics, <i>t</i>-test, ANOVA test and multivariate regression analysis were used to compare the differences between each subgroup and correlation analysis was conducted to understand the relationship between different factors and dependent variables.</p><p><b>Results:</b> Our investigation found that the level of awareness and acceptance of nuclear power was generally not high. Respondents' gender, age, marital status, residence, educational level, family income and the distance away from the nuclear power plant are important effect factors to the knowledge of and attitude to nuclear power.</p><p><b>Conclusions:</b> The public concerns about nuclear energy's impact are widespread. The level of awareness and acceptance of nuclear power needs to be improved urgently.</p
