    Interleukin-37 is increased in ankylosing spondylitis patients and associated with disease activity

    Background: Interleukin-37 (IL-37) has been known to play an immunosuppressive role in various inflammatory disorders, but whether it participates in the regulation of pathogenesis of ankylosing spondylitis (AS) has not been investigated. Here, we examined the serum levels of IL-37 and its clinical association in AS, and explored the anti-inflammatory effects of IL-37 on peripheral blood mononuclear cells (PBMCs) from AS patients. Methods: The mRNA levels of IL-37, TNF-alpha, IL-6, IL-17, and IL-23 in PBMCs and their serum concentrations from 46 AS patients were examined by real-time polymerase chain reaction (RT-PCR) and enzyme-linked immunoassay (ELISA), respectively. The correlations between serum IL-37 levels with disease activity, laboratory values and pro-inflammatory cytokines in AS were analyzed by Spearman correlation test. PBMCs from 46 AS patients were stimulated with recombinant IL-37 protein, expressions of TNF-alpha, IL-6, IL-17 and IL-23 were determined by RT-PCR and ELISA. Results: Compared to healthy controls (HC), AS patients and active AS patients showed higher levels of IL-37 in PBMCs and serum respectively. Strikingly, serum IL-37 levels were higher in AS patients with osteoporosis than those without. Serum levels of IL-37 were correlated with laboratory values as well as TNF-alpha, IL-6 and IL-17, but not IL-23 in patients with AS. The productions of pro-inflammatory cytokines such as TNF-alpha, IL-6, IL-17, IL-23 in PBMCs from AS patients were obviously attenuated after recombinant IL-37 stimulation, but not in the HC. Conclusion: The higher levels of IL-37 were found in AS patients, which were correlated with disease activity and AS related pro-inflammatory cytokines.     The infiltrative growth and malignant biological behavior of glioma make it one of the most challenging malignant tumors in the brain, and how to maximize the extent of resection (EOR) while minimizing the impact on normal brain tissue is the pursuit of neurosurgeons. The current intraoperative visualization assistance techniques applied in clinical practice suffer from low specificity, slow detection speed and low accuracy, while Raman spectroscopy (RS) is a novel spectroscopy technique gradually developed and applied to clinical practice in recent years, which has the advantages of being non-destructive, rapid and accurate at the same time, allowing excellent intraoperative identification of gliomas.     Nuclear mass table in deformed relativistic Hartree-Bogoliubov theory in continuum, II: Even-ZZ nuclei

    The mass table in the deformed relativistic Hartree-Bogoliubov theory in continuum (DRHBc) with the PC-PK1 density functional has been established for even-ZZ nuclei with 8Z1208\le Z\le120, extended from the previous work for even-even nuclei [Zhang et. al.\it{et.~al.} (DRHBc Mass Table Collaboration), At. Data Nucl. Data Tables 144, 101488 (2022)]. The calculated binding energies, two-nucleon and one-neutron separation energies, root-mean-square (rms) radii of neutron, proton, matter, and charge distributions, quadrupole deformations, and neutron and proton Fermi surfaces are tabulated and compared with available experimental data. A total of 4829 even-ZZ nuclei are predicted to be bound, with an rms deviation of 1.477 MeV from the 1244 mass data. Good agreement with the available experimental odd-even mass differences, α\alpha decay energies, and charge radii is also achieved. The description accuracy for nuclear masses and nucleon separation energies as well as the prediction for drip lines is compared with the results obtained from other relativistic and nonrelativistic density functional. The comparison shows that the DRHBc theory with PC-PK1 provides an excellent microscopic description for the masses of even-ZZ nuclei. The systematics of the nucleon separation energies, odd-even mass differences, pairing energies, two-nucleon gaps, α\alpha decay energies, rms radii, quadrupole deformations, potential energy curves, neutron density distributions, and neutron mean-field potentials are discussed.Comment: 394 pages, 17 figures, 2 tables, published in Atomic Data and Nuclear Data Tables, data file in the TXT form is available for download under "Ancillary files

    Effect of Enhanced UV-B Radiation on Fruit Maturity and Quality, and Leaf Photosynthesis in ' Guifei ' Mango

    (1) Background: Investigating the characteristics of photosynthetic physiological changes of leaves in Mangifera indica L. cv. 'GuIfei' under enhanced UV-B radiation, natural light exposed trees were regarded as control, and 96 kJ&amp;middot;m-2&amp;middot;d-1enhanced UV-B radiation was artificially simulated in the field; (2) Methods: The changes of fruit maturity and fruit quality, leaf net photosynthetic rate (Pn), photosynthetic pigments contents, photochemical reaction, activities of photosynthetic enzymes and their genes expressions were determined; (3) Results: Compared with CK, the percentage of mature fruits of the treatment was significantly increased, and fruit quality was better. The net photosynthetic rate (Pn), the contents of photosynthetic pigment, Hill reaction activity and photochemical quenching coefficient (qP) of the treatment leaves showed a significantly higher trend than CK. The activities of Rubisco and RCA, and the expression of Rubisco genes rbcL and rbcS were significantly increased; (4) Conclusions: 96 kJ&amp;middot;m-2&amp;middot;d-1 enhanced UV-B radiation treatment improved Rubisco activity through increasing the expression of Rubisco genes rbcL and rbcS, thereby enhancing the CO2-fixing capacity and dark reaction capacity of leaves. Based on this, it raised the net photosynthetic rate of leaves, which promoted the early maturity of 'Guifei' mango by the fast accumulating photosynthetic products.</jats:p

    LINC00665 regulates hepatocellular carcinoma by modulating mRNA via the m6A enzyme

    Abstract This study aimed to reveal the mechanism by which long noncoding RNAs (lncRNAs) modulate hepatocellular carcinoma (HCC) by regulating mRNA via the N6-methyladenosine (m6A) enzyme. The expression and clinical data of 365 HCC patients and 50 healthy control samples were downloaded from the the Cancer Genome Atlas (TCGA) database. Differentially expressed lncRNAs (DElncRNAs) and differentially expressed mRNAs (DEmRNAs) screened using limma packages from the R. m6A2Target database were used to predict the relationship between m6A enzyme-lncRNA and m6A enzyme-mRNA. The mRNAs in the lncRNA-m6A enzyme-mRNA network were subjected to enrichment analysis. Cox regression analysis was used to screen for RNAs significantly related to HCC prognosis. The expression of differentially expressed RNAs (DERs) was verified using the TCGA dataset and GSE55092. Eighty-five DElncRNAs and 747 DEmRNAs were identified. The mRNAs in the lncRNA-m6A enzyme-mRNA network were primarily involved in mitotic cell division, the p53 signaling pathway, and the cell cycle. Three lncRNAs and 14 mRNAs were significantly associated with HCC prognosis. Furthermore, the expression of 12 DERs differed significantly between patients in the early and advanced stages. LINC00665 was predicted to regulate 11 mRNAs by modulating IGF2BP1, IGF2BP2, and YTHDF1. Thus, this study provides new insights into the roles of lncRNA and m6A enzymes in HCC.</jats:p

    Transcriptomics-based Analysis of the Response of Sugar Content in Litchi Pulp to Foliar Calcium Fertilizer Treatment

    Ten 16-year-old trees were used as test materials to investigate the effect of foliar calcium fertilizer on the sugar content of ‘Feizixiao’ litchi (Litchi chinensis Sonn.) pulp. The experiment began 35 days after anthesis (DAA) in 2020 and 2021, and the treatment was a foliar spray application of 0.3% CaCl2 aqueous solution, whereas the control was a foliar spray application of water. The sugar content, sucrose-metabolizing enzymes, and ATP-dependent phosphofructokinase (PFK) activities in pulp were measured in 2020 and 2021. Transcriptome sequencing (RNA-seq) was performed on RNA samples from treatment and control fruit pulps at 35, 63, and 69 DAA (full mature stage) in 2020, and 10 genes were chosen for confirmation by real-time polymerase chain reaction (PCR) in 2020 and 2021. At full maturity, the soluble sugar content in the calcium-treated group was extremely significantly or significantly higher than that in the control group. After 63 DAA, the net sucrose-metabolizing enzyme activity in the calcium-treated group was significantly higher than that in the control group. Furthermore, at full maturity, the calcium-treated group had significantly higher sucrose synthase cleavage activity and significantly lower PFK activity than the control group. Fifty-four highly expressed genes in the glycolytic pathway (EMP) were screened from transcriptome data, including hexokinase, PFK, and pyruvate kinase genes; 87% of these genes were downregulated in the treatment group compared with the control group at 69 DAA in 2020. The linear regression between RNA-seq and real-time PCR results was significant in 2020 (r = 0.9292) and 2021 (r = 0.8889). When the fruit is fully ripe, calcium treatment increases net sucrose-metabolizing enzyme activity by increasing sucrose synthase cleavage activity, promoting the accumulation of reducing sugars, and it downregulates phosphofructokinase gene expression in EMP, promoting sugar accumulation.</jats:p
