    Synergistic Effects of Clopidogrel and Fufang Danshen Dripping Pills by Modulation of the Metabolism Target and Pharmacokinetics

    Background and Objective. The aim was to evaluate the synergistic effects of clopidogrel and FDDP by modulating the metabolism target and the pharmacokinetics. Methods. The inhibition effect of FDDP on the CES1 was first investigated by the molecular simulation method, and the synergistic effects on the pharmacokinetics of CPGS were studied as follows: SD rats were treated with oral clopidogrel alone at a dosage of 30 mg/kg or the combination of clopidogrel and FDDP at dosages of 30 mg/kg and 324 mg/kg, respectively, for 21 days. The concentrations of CPGS in the blood plasma samples were determined and the calculated concentrations were used to determine the pharmacokinetic parameters. Results. 20 compounds in FDDP potentially interacted with CES1 target. The CPGS showed a two-compartment model pharmacokinetic profile. The concentration-time course of CPGS was not changed by FDDP, but FDDP decreased the peak plasma concentration and area under the curve of CPGS. Conclusion. The CES1’s activity could be partly inhibited by FDDP through the molecular simulation investigation. The concentration-time course of CPGS was altered slightly by FDDP. The results demonstrated the synergistic effects of clopidogrel and FDDP by modulating both the pharmacokinetics and the target metabolism

    Fluid mineralization of the Dongtongyu gold deposit in the southern margin of North China craton: Evidence from microthermometry and composition of fluid inclusions

    In this study, petrographic, microthermometric, and synchrotron radiation X-ray fluorescence (SRXRF) analyses of fluid inclusions were conducted to shed light on the mineralization mechanism of the Dongtongyu deposit and provide some evidence of the relationship among CO2, Au, and other ore elements (e.g., Cu, Fe, Zn, and Pb) in ore-forming fluids. The ore-forming fluid is characterized as the H2O–CO2–NaCl system with medium–high temperatures and low salinities. Four structural mineralization stages are distinguished: Pyrite-quartz (Stage 1), gold-quartz-pyrite (Stage 2), gold-quartz-polymetallic sulfide (Stage 3), and quartz-calcite (Stage 4). Fluid inclusions in Stages 1–3 are dominated by the H2O–CO2 type, and most of them contain liquid H2O and liquid CO2 at room temperature. The melting temperatures (Tm-CO2 = −82.1°C to −57.5°C) of solid CO2 in Stage 1 are relatively low. The values of Tm-CO2 in Stages 2–3 are quite close, with ranges of −60.5°C to −56.5°C and −59.2°C to −58.6°C, respectively. The melting temperatures of clathrate in Stages 1–3 are −2.7°C to +7.8°C, −5.5°C to +7.8°C, and +3.7°C to +7.2°C. The homogenization temperatures of the CO2 phase in the H2O–CO2 inclusions of the three stages are measured as −7.5°C to +31.7°C, −7.5°C to +29.3°C, and 7.1°C to +24.1°C. The total homogenization temperatures in Stages 1–3 are 180°C–394°C, 202°C–305°C, and 224°C–271°C, with salinities of 4.3 wt.%–18.2 wt% NaCl, 4.3 wt.%–20.0 wt% NaCl, and 5.3 wt.%–11.0 wt% NaCl, respectively. The laser Raman spectroscopy results show that the CO2–H2O inclusions in the quartz veins contain abundant CO2 and CH4. The SRXFR results show that most of the elements, especially As, Te, and Cu, are more enriched in liquid CO2 than in liquid H2O. The elements of Au, Fe, Ni, Cu, and Pb have higher concentrations in H2O–CO2-type fluid inclusions in Stage 2 than other fluid inclusions in Stages 1–2, suggesting that gold mineralization is closely related to CO2-rich fluids. During the fluid evolution process, fluid immiscibility is an important mineralization mechanism of gold. The increase in CO2 and CH4 and the decrease in the fluid temperature might promote fluid immiscibility

    Comparative expression profiles of carboxylesterase orthologous CXE14 in two closely related tea geometrid species, Ectropis obliqua Prout and Ectropis grisescens Warren

    Insect carboxylesterases (CXEs) can be expressed in multiple tissues and play crucial roles in detoxifying xenobiotic insecticides and degrading olfactory cues. Therefore, they have been considered as an important target for development of eco-friendly insect pest management strategies. Despite extensive investigation in most insect species, limited information on CXEs in sibling moth species is currently available. The Ectropis obliqua Prout and Ectropis grisescens Warren are two closely related tea geometrid species, which share the same host of tea plant but differ in geographical distribution, sex pheromone composition, and symbiotic bacteria abundance, providing an excellent mode species for studies of functional diversity of orthologous CXEs. In this study, we focused on EoblCXE14 due to its previously reported non-chemosensory organs-biased expression. First, the EoblCXE14 orthologous gene EgriCXE14 was cloned and sequence characteristics analysis showed that they share a conserved motif and phylogenetic relationship. Quantitative real-time polymerase chain reaction (qRT-PCR) was then used to compare the expression profiles between two Ectropis spp. The results showed that EoblCXE14 was predominately expressed in E. obliqua larvae, whereas EgriCXE14 was abundant in E. grisescens at multiple developmental stages. Interestingly, both orthologous CXEs were highly expressed in larval midgut, but the expression level of EoblCXE14 in E. obliqua midgut was significantly higher than that of EgriCXE14 in E. grisescens midgut. In addition, the potential effect of symbiotic bacteria Wolbachia on the CXE14 was examined. This study is the first to provide comparative expression profiles of orthologous CXE genes in two sibling geometrid moth species and the results will help further elucidate CXEs functions and identify a potential target for tea geometrid pest control

    Microstructural Modeling and Strengthening Mechanism of TiB/Ti-6Al-4V Discontinuously-Reinforced Titanium Matrix Composite

    A novel modeling method was proposed to provide an improved representation of the actual microstructure of TiB/Ti-6Al-4V discontinuously-reinforced titanium matrix composite (DRTMC). Based on the Thiessen polygon structure, the representative volume element (RVE) containing the complex microstructures of the DRTMC was first generated. Thereafter, by using multiple user-defined subroutines in the commercial finite element software ABAQUS, the application of asymmetric mesh periodic boundary conditions on the RVE was realized, and the equivalent elastic modulus of the DRTMC was determined according to the homogenization method. Through error analyses on the experimental and calculated results regarding the equivalent elastic parameters of the DRTMC, the rationality of generating the DRTMC finite element model by using the present method was validated. Finally, simulations based on four types of network-like models revealed that the present simplifications to the particle shape of the reinforcement phase had less of an influence on the overall composite strength. Moreover, the present study demonstrates that the DRTMC enhancement is mainly attributed to the matrix strengthening, rather than the load-transferring mechanism. The strengthening influences of the distribution forms of the reinforcement phases, including their distribution density and orientation, were studied further. It was found that both the higher distribution density and limited distribution orientation of the particles would increase the probability of overlapping and merging between particles, and; therefore, higher strength could be yielded when the volume fraction of the reinforcement phase reached a certain threshold. Owing to the versatility of the developed methods and programs, this work can provide a useful reference for the characterization of the mechanical properties of other composites types

    The mitochondrial genome of Ectropis dentilineata Moore, 1868 (Lepidoptera: Geometridae) from tea plantations in Guizhou province, China

    Ectropis dentilineata Moore, 1868 has a wide geographic distribution while its genome data are largely unknown. In this study, we collected samples of E. dentilineata from a tea plantation in Guizhou province, China. Illumina sequencing showed that the mitochondrial genome of E. dentilineata is 15,356 bp in length, containing the entire set of 37 mitochondrial genes. Several tRNAs show gene arrangements compared with the ancestral gene order, mainly involving the tRNA cluster (M-I-Q). These data will facilitate a deeper understanding and exploitation of E. dentilineata

    Clinical Characteristics and Prognosis of Adult Orbital Cellulitis in a Tertiary General Hospital

    Purpose. Adult orbital cellulitis (OC) occurs relatively rarely, and comprehensive studies that retrospectively evaluate OC are lacking. Here, we aimed to examine the clinical characteristics and prognosis of OC in a tertiary general hospital. Methods. Between October 2010 and May 2019, patients presenting with clinical symptoms of OC in a tertiary general hospital were analyzed in this retrospective study. Twenty-six cases were identified for a detailed review. In these cases, 16 males and 10 females were diagnosed with orbital cellulitis by clinical characteristics and multimodal examinations. We divided patients into three groups: (1) patients secondary to rhinosinusitis, (2) patients secondary to endogenous infection(s) without endophthalmitis, and (3) patients secondary to endophthalmitis. For each group, age, gender, eye type, combined systemic diseases, clinical presentation, leukocyte count, blood culture, diagnostic imaging, therapeutic methods, length of stay, time of postoperation, and patient prognosis were analyzed in detail. Results. There were no significant differences regarding age, gender, preoperative leukocyte count, exophthalmia, blood culture, treatment, or visual changes within the three groups (P<0.05). There were significant differences, however, in postoperative leukocyte count and ophthalmoplegia between the three groups (P<0.05). The preoperative and postoperative logarithms of the minimum resolution angle scored by the best-corrected visual acuity (LogMAR BCVA) of group 3 were statistically significant compared to group 1 and group 2 (P<0.05). Conclusion. We confirmed that the prognosis of OC patients combined with systemic diseases was poor. Patients with OC secondary to endophthalmitis infrequently experience ophthalmoplegia; however, these patients still have poor visual outcomes. Patients cultured positive for Klebsiella pneumoniae infection may not be associated with liver abscess. The level of leukocytes may indicate the condition of the disease

    The Mitochondrial Genomes of Two Parasitoid Wasps <i>Protapanteles immunis</i> and <i>Parapanteles hyposidrae</i> (Hymenoptera: Braconidae) with Phylogenetic Implications and Novel Gene Rearrangements

    Parapanteles hypsidrae (Wilkinson, 1928) and Protapanteles immunis (Haliday, 1834) are the most important parasitic wasps of Ectropis grisescens Warren and Ectropis obliqua (Prout). We sequenced and annotated the mitochondrial genomes of Pa. hyposidrae and Pr. immunis, which are 17,063 bp and 16,397 bp in length, respectively, and possess 37 mitochondrial genes. We discovered two novel types of gene rearrangement, the local inversion of nad4L in Pa. hyposidrae and the remote inversion of the block cox3-nad3-nad5-nad4 in Pr. immunis, within the mitogenomes of Braconidae. The phylogenetic analysis supported the subfamily Microgastrinae is a monophyletic group, but the tribes Apantelini and Cotesiini within this subfamily are paraphyletic groups

    An Evaluation Method of Brittleness Characteristics of Shale Based on the Unloading Experiment

    Shale reservoir has an initial unloading effect during the natural uplift and erosion process, which causes the shale brittleness to change, affecting the design of the fracturing scheme. To consider this, the axial compression loading and confining pressure unloading experiment of shale is carried out, and then the influence of unloading rate on the mechanical parameters, failure characteristics, and the brittleness of rock are analyzed. What is more, a new evaluation method of brittleness characteristics that take the unloading effect into consideration is proposed. The conclusions are as follows: (1) The unloading rate has a weakening effect on the mechanical parameters, such as the destructive confining pressure and the residual strength of the samples. (2) The failure characteristics of shale specimens are a single shear failure in an oblique section under low unloading rate, and multiple shear zones accompanied with bedding fracture under high unloading rate. (3) The brittleness of shale samples is well verified by the brittleness index B d 1 and B d 2 during the loading path; nevertheless, it has shortage at the unloading path. This paper proposes a new brittleness evaluation method which can consider the influence of the different unloading rates and unloading points.
