    Ethnopharmacokinetic- and Activity-Guided Isolation of a New Antidepressive Compound from Fructus Aurantii Found in the Traditional Chinese Medicine Chaihu-Shugan-San: A New Approach and Its Application

    Aims. We aimed to identify an antidepressive compound found in traditional Chinese medicine (TCM) by a new approach called ethnopharmacokinetic- and activity-guided isolation (EAGI). Methods. The new approach targets an unknown chromatographic peak produced by an absorbed compound found in oral Chaihu-Shugan-San (CSS) taken by patients with depression. Once the compound was isolated from Fructus Aurantii (FA), spectral data was employed to identify the compound. The effects of this compound, FA, and CSS on depressive behaviors were investigated. Results. The identified compound was merazin hydrate (MH) according to the new approach. MH, FA, and CSS significantly reduced immobility time and increased locomotor activity. The effects of MH, FA and CSS were similar to Fluoxetine at high doses. Conclusion. MH, a compound whose antidepressive effect is similar to FA and CSS, was isolated for the first time from FA via targeting its corresponding unknown chromatographic peak, and its antidepressive effect was compared with FA or CSS. These findings highlight the potential for drug R&D and pharmacological research of ∼100,000 TCMs

    Formation and characteristic corrosion behavior of alternately lamellar arranged α and β in as-cast AZ91 Mg alloy

    Formation and characteristic corrosion resistance of alternately lamellar arranged α and β in as-cast AZ91 Mg alloy were investigated as an independent micro-constituent identity. As-cast AZ91 presented three microstructural entities, i.e. α-Mg grain, (α+β) lamellae and coarse β particle, and each had its own Al content and microstructural morphology. The lamellae occurrence was due to the precipitation of β particle from the divorced eutectic Al-rich-α phase during solidification, because the Al composition can not exceed its maximum solubility. The evidences that were obtained from electrochemical tests, micro-corrosion morphology and hydrogen evolution rate certified that the (α+β) lamellae was beneficial to corrosion resistance, which was different from the reported deleterious influence for its original eutectic Al-rich-α phase. This different corrosion behavior was explained to be ascribed to the changes in fine structure and local composition that resulted in combined electrochemical effects of the changes in α and β phases on the corrosion

    Regulation of LYRM1 Gene Expression by Free Fatty Acids, Adipokines, and Rosiglitazone in 3T3-L1 Adipocytes

    LYR motif containing 1 (LYRM1) is a novel gene that is abundantly expressed in the adipose tissue of obese subjects and is involved in insulin resistance. In this study, free fatty acids (FFAs) and tumor necrosis factor-α (TNF-α) are shown to upregulate LYRM1 mRNA expression in 3T3-L1 adipocytes. Conversely, resistin and rosiglitazone exert an inhibitory effect on LYRM1 mRNA expression. These results suggest that the expression of LYRM1 mRNA is affected by a variety of factors that are related to insulin sensitivity. LYRM1 may be an important mediator in the development of obesity-related insulin resistance

    Effective components screening and anti-myocardial infarction mechanism study of the Chinese medicine NSLF6 based on "system to system" mode

    <p>Abstract</p> <p>Background</p> <p><it>Shuanglong </it>formula (SLF), a Chinese medicine composed of <it>panax ginseng </it>and <it>salvia miltiorrhiza </it>exhibited significant effect in the treatment of myocardial infarction (MI) in clinical. Because of the complex nature and lack of stringent quality control, it's difficult to explain the action mechanism of SLF.</p> <p>Method</p> <p>In this study, we present a "system to system" (S2S) mode. Based on this mode, SLF was simplified successively through bioactivity-guided screening to achieve an optimized minimal phytochemical composition (new formula NSLF6) while maintaining its curative effect for MI.</p> <p>Results</p> <p>Pharmacological test combining with the study of systems biology show that NSLF6 has activity for treatment MI through synergistic therapeutic efficacies between total ginsenosides and total salvianolic acids via promoting cardiac cell regeneration and myocardial angiogenesis, antagonistic myocardial cell oxidative damage.</p> <p>Conclusions</p> <p>The present S2S mode may be an effective way for the discovery of new composite drugs from traditional medicines.</p
