    Simple and Multistate Survival Curves

    Objective and Sample: This investigation assessed the comprehension of survival curves in a community sample of 88 young and middle-aged adults when several aspects of good practice for graphical communication were implemented, and it compared comprehension for alternative presentation formats. Design, Method, and Measurements: After reading worked examples of using survival curves that provided explanation and answers, participants answered questions on survival data for pairs of treatments. Study 1 compared presenting survival curves for both treatments on the same figure against presentation via 2 separate figures. Study 2 compared presenting data for 3 possible outcome states via a single “multistate” figure for each treatment against presenting each outcome on a separate figure (with both treatments on the same figure). Both studies compared alternative forms of questioning (e.g., “number alive” versus “number dead”). Numeracy levels (self-rated and objective measures) were also assessed. Results: Comprehension was generally good—exceeding 90% correct answers on half the questions—and was similar across alternative graphical formats. Lower accuracy was observed for questions requiring a calculation but was significantly lower only when the requirement for calculation was not explicit (13%–28% decrements in performance). In study 1, this effect was most acute for those with lower levels of numeracy. Subjective (self-rated) numeracy and objective (measured) numeracy were both moderate positive predictors of overall task accuracy (r ≈ 0.3). Conclusions: A high degree of accuracy in extracting information from survival curves is possible, as long as any calculations that are required are made explicit (e.g., finding differences between 2 survival rates). Therefore, practitioners need not avoid using survival curves in discussions with patients, although clear and explicit explanations are important </jats:p

While the obvious solution is to adopt electronic charting systems, this can prove challenging in practice. This article describes the key issues to consider in planning this transition and the charting options available, ranging from bespoke local systems to commercial packages and a new initiative by the Royal College of Paediatrics and Child Health

    Visualisation of BioPAX Networks using BioLayout Express (3D).

    BioLayout Express (3D) is a network analysis tool designed for the visualisation and analysis of graphs derived from biological data. It has proved to be powerful in the analysis of gene expression data, biological pathways and in a range of other applications. In version 3.2 of the tool we have introduced the ability to import, merge and display pathways and protein interaction networks available in the BioPAX Level 3 standard exchange format. A graphical interface allows users to search for pathways or interaction data stored in the Pathway Commons database. Queries using either gene/protein or pathway names are made via the cPath2 client and users can also define the source and/or species of information that they wish to examine. Data matching a query are listed and individual records may be viewed in isolation or merged using an 'Advanced' query tab. A visualisation scheme has been defined by mapping BioPAX entity types to a range of glyphs. Graphs of these data can be viewed and explored within BioLayout as 2D or 3D graph layouts, where they can be edited and/or exported for visualisation and editing within other tools

    Current plate boundary deformation of the Afar rift from a 3-D velocity field inversion of InSAR and GPS

    Extension, faulting, and magmatism are the main controls on the magnitude and localization of strain at mid‐ocean ridges. However, the temporal and spatial patterns of such processes are not clear since the strain distribution has not been resolved in the past at sufficient spatial resolution and over extended areas. Interferometric synthetic aperture radar (InSAR) and GPS data with unprecedented resolution are now available to us from the Afar rift of Ethiopia. Here we use a velocity field method to combine InSAR and GPS to form the first high‐resolution continuous three‐dimensional velocity field of Afar. We study an area that is 500 km wide and 700 km long, covering three branches of the Afar continental rift and their triple junctions. Our velocity field shows that plate spreading is currently achieved in Afar in contrasting modes. A transient postdiking deformation is focused at the Dabbahu rift segment, while in central Afar, spreading is distributed over several overlapping segments and southern Afar exhibits an interdiking deformation pattern focused at the Asal–Ghoubbet segment. We find that current spreading rates at Dabbahu, following the 2005–2010 intrusions, are up to 110 mm/yr, 6 times larger than the long‐term plate divergence. A segment‐centered uplift of up to 80 mm/yr also occurs, indicating that magma flow is still a primary mechanism of deformation during postdiking. On the other hand, no vertical displacements are currently observed in central and southern Afar, suggesting lack of significant magmatic activity at shallow levels

    All-fibre wavefront sensor

    We report on a tapered three-core optical fibre that can be used as a tip-tilt wavefront sensor. In this device, a coupled region of a few millimetres at the sensing tip of the fibre converts fragile phase information from an incoming wavefront into robust intensity information within each of the cores. The intensity information can be easily converted to linear wavefront error over small ranges, making it ideal for closed loop systems. The sensor uses minimal information to infer tip-tilt and is compatible with remote detector arrays. We explore its application within adaptive optics and present a validation case to show its applicability to astronomy.Comment: 7 pages, 6 figure

    The application of AEM to mapping the aquifer and groundwater characteristics of the La Grange groundwater area, WA

    This report describes the interpretation of a large airborne electromagnetic data survey that was designed to aid characterisation of groundwater systems in the La Grange groundwater allocation area which lies south of Broome, Western Australia. Characterisation of these systems is critical to developing policy around water use in the region, especially for the development of agriculture. This report is the second of two reports and it is designed to refine surfaces which are used as inputs to groundwater models. The volume defined by these surfaces viz. the seawater intrusion in the west, the top of the Jarlemai siltstone and the top of the Broome sandstone describes the portion of the Broome sandstone aquifer that is able to store water that can be used for human purposes, agriculture and mining. With the identification of palaeochannels in the La Grange groundwater allocation area and an improved analysis of geological structure from AEM results, most objectives of the survey have been realised. Palaeochannels were identified in the north of the La Grange groundwater allocation area, around La Grange Bay, and also in the south. Also in the south, faults were identified. It is hypothesised that these faults permit groundwater movement between the Broome sandstone, which is the important aquifer in the La Grange groundwater allocation area, and the Wallal sandstone which is the important aquifer south of the allocation area. Objectives relating to delineation of freshwater zones, and of water quality were not addressed. Although the top of the seawater intrusion was mapped, as was the top of the Broome sandstone near the coast, because freshwater and the sandstone matrix have similar electrical resistivities, it is difficult to map freshwater specifically

    Detection of a Nearby Halo Debris Stream in the WISE and 2MASS Surveys

    Combining the Wide-Field Infrared Survey Explorer All-Sky Release with the Two Micron All Sky Survey Point Source Catalog, we detect a nearby, moderately metal-poor stellar debris stream spanning 24° across the southern sky. The stream, which we designate Alpheus, is at an estimated distance of ~1.9 kpc. Its position, orientation, width, estimated metallicity, and, to some extent, its distance, are in approximate agreement with what one might expect of the leading tidal tail of the southern globular cluster NGC 288

    Stimulation of TLR4 by recombinant HSP70 requires structural integrity of the HSP70 protein itself

    <p>Abstract</p> <p>Background</p> <p>Toll-like receptor 4 (TLR4) is activated by bacterial endotoxin, a prototypical pathogen-associated molecular pattern (PAMP). It has been suggested that TLR4 can also be activated by damage-associated molecular pattern (DAMP) proteins such as HSP70. It remains a challenge to provide unequivocal evidence that DAMP proteins themselves play a role in TLR4 activation, as the DAMP proteins used are often contaminated with endotoxin and other TLR ligands introduced during protein expression and/or purification.</p> <p>Results</p> <p>Here we report that the activation of TLR4 on primary human macrophage cultures by recombinant HSP70 is not solely due to contaminating endotoxin. Polymyxin B pretreatment of HSP70 preparations to neutralize contaminating endotoxin caused significant reductions in the amount of TNF-α induced by the recombinant protein as determined by ELISA. However, digestion of HSP70 with Proteinase K-agarose beads also dramatically reduced the TNF-α response of macrophages to HSP70, while leaving levels of contaminating endotoxin largely unchanged relative to controls.</p> <p>Conclusions</p> <p>These results indicate that the stimulatory effect of recombinant HSP70 requires both the presence of endotoxin and structural integrity of the heat shock protein itself.</p
