    Collagen type III biosynthesis by cultured pubocervical fascia fibroblasts surrounding mono and multifilament polypropylene mesh after estrogens and tamoxifen treatment

    Abstract Aim: Surgical procedures using synthetic implants are currently considered as the most efficient therapy for stress urinary incontinence (SUI) and pelvic organ prolapse (POP). Insertion of the tape or mesh causes enhanced collagen synthesis that largely affects the biomechanical property of the implant. This process is significantly modulated by estrogens and improper wound healing and treatment failure may result in hypoestrogenism. The aim of the study was to assess the rate of collagen type III synthesis by pubocervical fascia fibroblasts cultured with polypropylene meshes in the presence of estrogens and tamoxifen. Material and Methods: Fibroblasts were obtained from pubo-cervical fascia sampled from a 52-year-old premenopausal woman who underwent surgical treatment for SUI and cultured with monofilament or multifilament polypropylene meshes in the presence of 17β-estradiol, estriol, daidzein or tamoxifen. The cultures were run for 216hr and the media were replaced every 72hr. N-terminal propeptide of type III procollagen (PIIINP) was used as a marker of collagen type III synthesis. Its concentration in the media was measured by radioimmunoassay. Pubocervical fascia fibroblast cultured with monofilament or multifilament meshes are capable of collagen type III synthesis. Following treatment with estradiol or tamoxifen, the highest PIIINP concentrations were observed after 72hr, whereas in case of estriol, daidzein or no treatment after 144hr of culture, regardless of the type of mesh used. Results: Only in cultures containing monofilament mesh and stimulated with estriol the high rate of collagen type III synthesis persisted until the end of the experiment. Paradoxically, the highest total production of PIIINP was observed in culture treated with tamoxifen, both for multifilament and monofilament meshes. Conclusion: The rate of collagen type III synthesis by pubocervical fascia fibroblast cultured with polypropylene meshes is subjected to modulation by estrogens and antiestrogens

    Different types of canine lymphoma respond differently to chemotherapy and have different prognoses. Diffuse large B-cell lymphoma (DLBCL) is the most common lymphoma in dogs. Topoisomerase II alpha (TOPIIα) protein has been shown to be a proliferation marker associated with prognostic significance. The aim of the study was to determine the relationship between TOPIIα expression, mitotic count (MC), and Ki67 antigen index in DLBCL in dogs, taking into account the applicability of these parameters to select the chemotherapy protocol with emphasis on the use of anthracycline drugs. Samples of formalin-fixed paraffin-embedded lymph nodes from 34 dogs with DLBCL were immunohistochemically labelled with anti-TOPIIα and Ki67. The number of positive cells and the intensity of the reaction were taken into account in order to assess TOPIIα expression. MC was estimated in the hematoxylin and eosin-stained slides in the area of 2.37 mm2. Positive association between TOPIIα and MC, but no association between TOPIIα and Ki67 was found. It can be concluded that the immunohistochemical determination of TOPIIα as a molecular target for drugs from the anthracycline group may be used in association with MC to establish a diagnostic-clinical protocol for selecting dogs with DLBCL for treatment with anthracycline drugs

    Study on the concentration of airbone respirable asbestos fibres in rural areas of the Lublin region in south-east Poland

    Objective. The objective of the study was measurement of the concentrations of airborne asbestos fibres in the rural environment of the Lublin Region in south-east Poland. Methods. Measurements of concentrations of respirable asbestos fibres were carried out in the rural areas of the Lublin Region (Lublin and Włodawa counties) for a period of 24 months. The studies were conducted on 3 farms with various technical conditions of asbestos-containing materials: Farm A – good technical condition of asbestos products, Farm B – poor technical condition, and Farm C – with no asbestos containing products and no such products in its direct vicinity (up to 500 m). On the selected farms, 3 samples on each were simultaneously collected at 3 measuring sites. During the period 2009–2011, a total number of 216 samples were collected on all farms. Sampling was performed using JSH 16,000 stationary aspirators, with air flow velocity of 16 l/min. and sampling time 60–80 minutes. The number of fibres on filters was determined using an optical phase contrast microscope. Results. The study showed that the mean concentration of respirable asbestos fibres on the farms examined was 296 fibres•m [sup]-3[/sup]. The highest concentrations were noted on Farm B was 529 fibres•m [sup]-3[/sup], on average; on farm A the mean concentration of respirable fibres was 328 fibres•m [sup]-3[/sup] , whereas the lowest mean concentration of airborne respirable asbestos fibres was noted on farm C, where there were no asbestos products (30 fibres•m [sup]-3[/sup] )

    Study of DNA topoisomerase IIa expression in canine lymphomas and its potential role as a marker of sensitivity to anthracycline-based chemotherapy in dogs

    Introduction. Canine lymphoma remains one of the most chemotherapy-responsive neoplasia in dogs. Many factors affect the prognosis in dogs treated for lymphoma, but indications for a specific treatment regimen in individual animals with lymphoma are poorly defined. Topoisomerase IIα (TOPIIα) is a key enzyme in DNA replication and a molecular target for TOPIIα inhibitors, including anthracyclines. The aim of this study was to determine the expression of TOPIIα in canine malignant lymphomas. The relationship between TOPIIα expression in canine lymphomas and potential sensitivity of neoplastic cells to anthracycline-based chemotherapy is discussed. Materials and method. Samples of formalin-fixed paraffin-embedded lymph nodes from 47 dogs with different subtypes of non-Hodgkin’s (34 B-cell and 13 T-cell) lymphoma were immunohistochemically labeled with anti-TOPIIα. The number of positive cells and the intensity of the reaction were taken into account in order to assess TOPIIα expression. Results. TOPIIα expression was evident in all cases, although differences in the number of positive cells and intensity of the reaction were demonstrated between B-cell and T-cell lymphoma groups as well as within individual groups. Based on the established scoring system, in the B-cell lymphoma group statistically higher expression of TOPIIα was found compared to the T-cell lymphoma group (P = 0.006). In B-cell lymphoma group moderate (41.18%) and strong (32.35%) TOPIIα expression predominated, whereas among T-cell lymphoma group the majority were cases with a weak (46.15%) TOPIIα expression. Conclusion. These preliminary results indicate that further studies are needed to determine the prognostic value of TOPIIα expression with regard to the sensitivity of canine B-cell lymphomas to anthracycline-based chemotherapy regimen. Nevertheless, this study indicates the possibility of choosing the appropriate treatment of canine lymphoma based on TOPIIa expression

    Objectives: Polypropylene meshes are widely used for surgical treatment of stress urinary incontinence(SUI) and pelvic organ prolapse (POP). Synthesis and deposition of collagen induced by an inserted implant arelargely controlled by oestrogens. The aim of the study was to assess the rate of collagen type I (Col I) synthesisby pubo-cervical fascia (PCF) fibroblasts cultured with mono- or multifilament polypropylene meshes in thepresence of oestrogens. Material and Methods: Specimens of PCF were obtained during a surgical procedure from a 56-year-old womansuffering from SUI and POP. Fibroblasts were cultured with mono- or multifilament meshes and exposed to17β-oestradiol, oestriol or phytoestrogen daidzein. The cultures were run for 216 hrs and the media were replaced every 72 hrs. Procollagen type 1 N-terminal propeptide (PINP), a marker of Col I biosynthesis, was assessedin culture media by radioimmunoassay. Results: The biosynthesis of Col I was more abundant in the presence of monofilament than multifilamentmeshes. Fibroblasts exposed to oestriol or daidzein produced more Col I than those treated with oestradiol,regardless of the mesh applied. In the presence of monofilament mesh the rate of Col I synthesis induced byoestriol and daidzein increased persistently until the end of the experiment, whereas the peak concentration ofPINP in cultures treated with oestradiol was observed between 72 hrs and 144 hrs. In the presence of multifilamentmesh the rate of Col I production dropped after 144 hrs in all cultures. Conclusions: PCF fibroblasts produce more Col I when cultured on monofilament than on multifilamentmesh. This process may be enhanced by oestriol and phytoestrogens
