    Phenotypic Characterization of the Arabidopsis ufm1 (Ubiquitin Fold Modifier) Gene Involved in Seed Development

    Background and Objective: With the completion of the Arabidopsis genome sequencing, the next challenge is the determination ofgene function. Post-translational modifications of proteins by small polypeptide are implicated in plant growth and development.Ubiquitin fold modifier 1 is a member of the ubiquitin like protein family. While, the enzymatic conjugation cascade of ubiquitin foldmodifier 1 has been elucidated in recent years, its biological role is still unknown. The present study focuses in elucidate the role of theufm1 in plants development. Materials and Methods: The researchers performed analyses of the development of wild-type Columbiaplants and mutants of the ubiquitin fold modifier 1 gene to identify and interpret phenotypic changes in plants and seeds. Data werestatistically analyzed with the Info Stat software. Results: In this study, evidence suggesting that ubiquitin fold modifier 1 is involved inthe normal development of the seeds. Conclusion: The ufm1 gene would affect the normal development of the seed, particularly of theembryo, causing high percentage of seed abortion.Fil: Cornejo, Paula.     Visual Perceptual Difficulties and Under-Achievement at School in a Large Community-Based Sample of Children

    Difficulties with visual perception (VP) are often described in children with neurological or developmental problems. However, there are few data regarding the range of visual perceptual abilities in populations of normal children, or on the impact of these abilities on children's day-to-day functioning. Methods Data were obtained for 4512 participants in an ongoing birth cohort study (Avon Longitudinal Study of Parents and Children; ALSPAC). The children's mothers responded to questions designed to elicit indications of visual perceptual difficulties or immaturity, when their children were aged 13 years. We examined associations with standardised school test results in reading and in mathematics at age 1314 years (SATS-KS3), accounting for potential confounders including IQ. Results Three underlying factors explained half the variance in the VP question responses. These correlated best with questions on interpreting cluttered scenes; guidance of movement and face recognition. The adjusted parameter estimates (95% CI) for the cluttered-scenes factor (0.05; 0.02 to 0.08; p<0.001) suggested positive associations with the reading test results whilst that for the guidance-of-movement factor (0.03; 0.00 to 0.06; p = 0.026) suggested positive association with the mathematics results. The raw scores were associated with both test results. Discussion VP abilities were widely distributed in this sample of 13-year old children. Lower levels of VP function were associated with under-achievement in reading and in mathematics. Simple interventions can help children with VP difficulties, so research is needed into practicable, cost-effective strategies for identification and assessment, so that support can be targeted appropriately

    Epigenetic consequences of interploidal hybridisation in synthetic and natural interspecific potato hybrids

    Interploidal hybridisation can generate changes in plant chromosome numbers, which might exert effects additional to the expected due to genome merger per se (i.e., genetic, epigenetic and phenotypic novelties).Wild potatoes are suitable to address this question in an evolutionary context. To this end, we performed genetic (AFLP and SSR), epigenetic (MSAP), and cytological comparisons in: i) natural populations of the diploid cytotype of the hybrid taxonomic species Solanum x rechei (2n=2x, 3x) and its parental species, the triploid cytotype of Solanum microdontum (2n=2x, 3x) and Solanum kurtzianum (2n=2x); and ii) newly synthesised intraploidal (2x x 2x) and interploidal (3x x 2x) S. microdontum x S. kurtzianum hybrids.Aneuploidy was detected in S. x rechei and the synthetic interploidal progeny; this phenomenon might have originated the significantly higher number of methylation changes observed in the interploidal vs. the intraploidal hybrids. The wide epigenetic variability induced by interploidal hybridisation is consistent with the novel epigenetic pattern established in S. x rechei compared to its parental species in nature.These results suggest that aneuploid potato lineages can persist throughout the short term, and possibly medium term, and that differences in parental ploidy resulting in aneuploidy are an additional source of epigenetic variation.Fil: Cara, Nicolás.     Advances in genetic breeding for processing tomatoes

    de mejora genética en tomate para industria, asistido por marcadores moleculares, con el objetivo principal de generar cultivares autopolinizadas, especialmente orientadas a pequeños productores, como opción al uso de cultivares híbridas, resistentes a nematodos (Meloidogyne incognita, M. arenaria y M. javanica), peste negra (TSWV) y peca del tomate (Pseudomonas tomato syringae pv. tomato). En el proceso de mejora se siguió el método de retrocruza y selección genealógica. Las determinaciones de marcadores moleculares fueron realizadas por el Laboratorio de Biología Molecular INTA-Facultad de Ciencias Agrarias (UNCuyo). Las líneas avanzadas fueron evaluadas en el Programa Tomate 2000, con testigos híbridos difundidos en el gran cultivo. De los materiales evaluados a 2010-11, se han identificado homocigotos para resistencia combinada a nematodos y peste negra; igualmente para nematodos, peste negra y peca del tomate y homocigotos a nematodos, peste negra y peca del tomate. También se han determinado materiales heterocigotos en distintas combinaciones de los genes bajo estudio. Líneas avanzadas del programa de mejora, no arrojaron diferencias significativas con los testigos híbridos, en ensayos comparativos de rendimiento (kg·ha-1). Se analizan recomendaciones de cultivo y destino de los materiales.At the EEA La Consulta, INTA, a molecular assisted breeding program for processing tomatoes started in 2001 with the aim to generate self-pollinated cultivars, specially oriented for small growers. One of the main objectives of breeding program is the introduction of resistance to nematodes (Meloidoyne incognita, M. arenaria y M. javanica), tomato spotted wild virus (TSWV) and, tomato speck (Pseudomonas tomato syringae pv. tomato) as an alternative to use foreign and expensive hybrids cultivars. Breeding lines were obtained by pedigree and backcross selection methods.fake watches Early selection of resistant plants was done using molecular markers linked to nematode and TSWV. Molecular marker determinations were performed by the Laboratory of Molecular Biology of the INTA-Faculty of Agricultural Sciences (UNCuyo). Advanced breeding lines were tested against commercial hybrids in the Tomato 2000 Program. From all the materials evaluated up to 2010-11,a cartier replica we have identified homozygous lines that combine resistance to nematodes and tomato spotted wilt virus; homozygous lines that combine resistance to nematodes, tomato spotted wilt virus and tomato speck and homozygous lines resistant to nematodes, tomato spotted wilt virus and tomato speck (in separate genotypes). Also, heterozygous lines in different combinations for the genes under study.     Exploring the therapeutic alliance and race from sports psychologists' and athletes' lived experiences: A pilot study

    The therapeutic alliance has been explored widely within therapy literature but more research is necessary on the sport and exercise psychology client dyad. The racial/cultural identity development model and Rogers's (1957) six core conditions provide the conceptual and theoretical framework for this pilot qualitative study, which seeks to explore the building process in therapeutic relationships within cross-racial sport and exercise psychology. The perspectives of four black athletes of mixed descent and seven sport and exercise psychologists from diverse backgrounds (Arab = 1, Black British = 3, White British = 3) were considered. One-hour semi-structured interviews were analysed using an inductive thematic analysis. This yielded the following themes for athletes: lack of disclosure, racial impact on alliance, desired characteristics and experience. For sport and exercise psychologists these themes included therapeutic alliance building blocks, creating safe spaces and the racial impact of disclosure. Recommendations for building the process in cross-racial dyads are explored based on the participants' comments. The findings are discussed and areas for future research are explored based on these main themes.publishedVersio

    Parental style in families of origin and anxiety among batterers

    The present study was aimed at investigating male batterers' family of origin's parental style, and its relationship with anxiety symptoms in such men, by comparing these variables with men without a history of domestic violence with similar age, socio-economic status, education and marital status. A sample of 20 batterers and 20 men without a violence history was utilized. The instruments in Portuguese included: a Batterer's Family of Origin Semi-Structured Interview, The Parental Style Inventory, and the Beck Anxiety Inventory (BAI). Both groups did not differ statistically in regards to age, socio-economic status, education and marital status. However, the groups were statistically different in terms of the variables family of origin's parental style and anxiety. It is suggested that anxiety, combined with family of origin's inappropriate parenting, may contribute to intimate partner violence. Future studies could further investigate this possibility using larger samples.O presente estudo investigou o estilo parental da família de origem do agressor conjugal e o nível de ansiedade de tal homem, comparando-os com dados de homens não agressores de idade, nível sócio-econômico, escolaridade e estado civil semelhantes. A amostra foi constituída por 20 agressores conjugais e 20 não agressores. Os instrumentos utilizados foram: Roteiro de Entrevista Individual Semi-Estruturada da Família de Origem do Agressor, Inventário de Estilos Parentais e o Inventário de Ansiedade Beck (BAI). Os grupos não se diferenciaram estatisticamente no que se refere à idade, escolaridade, renda e estado civil. Os grupos se diferiram estatisticamente nas duas variáveis analisadas: estilo parental de origem e ansiedade. Pode-se sugerir que a ansiedade, quando combinada com práticas parentais inapropriadas na família de origem, contribua para a agressão do parceiro íntimo.     Pollen‐mediated gene flow from a commercial potato cultivar to the wild relative S. chacoense Bitter under experimental field conditions in Argentina

    The common potato, Solanum tuberosum ssp. tuberosum (tbr, 2n = 4x = 48; 4EBN), has many closely related wild tuber‐bearing species. Around 28 to 35 of them spontaneously grow in Argentina overlapping, in some areas, with the crop and/or experimental transgenic potatoes. Although it is well proven that hybridization barriers in potatoes can be incomplete, information on gene flow between cultivated and wild germplasm is scarce. Thus, a gene flow field experiment with a circular array was set up in Balcarce, Argentina, in 2009, and evaluated over two seasons. The tetraploid tbr cultivar Huinkul MAG and one compatible cloned genotype of the related wild potato S. chacoense Bitter (chc, 2n = 2x = 24; 2EBN), which produced 2n eggs, were used, respectively, as pollen donor and receptor. Berries with hybrid seeds – as revealed by ploidy and RAPD profiles – were obtained in one season, at 30 m from the pollen donor. These results reinforce others previously obtained with the same pollen donor and a male sterile tbr cultivar in a similar array, pointing out to the need of increasing isolation distances in areas of overlap between cultivated and wild potato germplasm to prevent or minimize undesirable pollen‐mediated gene flow.EEA BalcarceFil: Capurro, Mauricio Antonio.     Evaluación in vitro de filtración hacia conducto radicular de dos sistemas de reconstrucción post endodóncica

    ResumenEl objetivo de este estudio fue el de evaluar el grado de microfiltración en dos sistemas de reconstrucción postendodóncica, valorando la penetración de un colorante a lo largo de las restauraciones realizadas con endopostes y cementos duales de resina. En este estudio se emplearon 30 dientes premolares unirradiculares extraídos a los que se les realizó tratamiento de conductos y almacenándolos en agua bidestilada a 37 °C. Se distribuyeron los dientes en tres grupos de forma aleatoria, con diez dientes por cada grupo. La cementación de los postes se realizó en dos de los grupos, usando los sistemas RelyX Fiber post (3M ESPE dental products) y Parapost (Coltene-Whaledent). Las muestras fueron sometidas al proceso de termociclado y posterior inmersión en azul de metileno por siete días a 36 °C. Posteriormente se diafanizaron las muestras y fueron analizadas bajo microscopio, obteniendo los valores de penetración en milímetros. Los resultados mostraron que los dientes tratados con el sistema Parapost presentaron menor microfiltración. El grupo control mostró el mayor índice de microfiltración. El análisis estadístico con la prueba de Donett mostró diferencia estadísticamente significativa entre los sistemas Parapost y RelyX.ConclusiónSe demostró que el sistema de reconstrucción Parapost (Coltene-Whaledent), el cual tiene una técnica de grabado ácido, reportó menores valores de microfiltración, siendo los resultados estadísticamente significativos con respecto a los obtenidos con el sistema de reconstrucción RelyX (3M ESPE dental products).AbstractThe aim of the present study was to assess the degree of microleakage observed when using two different post-endodontic reconstruction systems; this was achieved assessing penetration of a dye along restorations performed with endodontic posts and dual-cure resin cements. For the present study, 30 extracted, single rooted premolars were used. Root canal treatment was performed on the teeth which were later stored in bi-distilled water at 37 °C. Teeth were randomly allotted to three groups, all groups were comprised of 10 teeth. In two groups, post cementation was performed using the systems RelyX Fiber Post (3M ESPE dental products) and Parapost (Coltene-Whaledent). Samples were subjected to thermo-cycling and later immersed in methylene blue for seven days at 36 °C. After this, samples were subjected to a diaphanization process and were then analyzed under a microscope; penetration in millimeters was thus obtained. Results revealed that teeth treated with Parapost system exhibited lesser microleakage. Control group experienced the greater amounts of microleakage. Statistical analysis conducted with Donett test showed statistically significant difference between Parapost and RelyX systems.ConclusionParapost reconstruction system (Coltene-Whaledent) with acid-etch technique exhibited lower microleakage values. Results were statistically significant when compared to those obtained with the RelyX reconstruction system (3M ESPE dental products)
