    Balance-approach For Mechanical Properties Test of Micro Fabricated Structure

The method is proved to be very practical in testing the force-deflection behavior of silicon cantilever, in which the Young’s modulus of the material can be calculated, and in investigating the static performance of bulk micromachined capacitive accelerometers.     Genetic Diversity of Salmonella enteric serovar Typhi and Paratyphi in Shenzhen, China from 2002 through 2007

    <p>Abstract</p> <p>Background</p> <p>Typhoid and paratyphoid fever are endemic in China. The objective of this investigation was to determine the molecular features of nalidixic acid-resistant <it>Salmonella enteric </it>serovar Typhi (<it>S. typhi</it>) and Paratyphi (<it>S. paratyphi</it>) from blood isolates in Shenzhen, China.</p> <p>Results</p> <p>Twenty-five <it>S. typhi </it>and 66 <it>S. paratyphi </it>were isolated from 91 bacteriemic patients between 2002 and 2007 at a hospital in Shenzhen, Southern China. Fifty-two percent (13/25) of <it>S. typhi </it>and 95.3% (61/64) of <it>S. paratyphi </it>A were resistant to nalidixic acid. Sixty-seven isolates of nalidixic acid-resistant <it>Salmonella </it>(NARS) showed decreased susceptibility to ciprofloxacin (MICs of 0.125-1 μg/mL). All 75 NARS isolates had a single substitution in the quinolone resistance-determining region (QRDR) of GyrA (Ser83→Phe/Pro/Tyr, or Asp87→Gly/Asn), and 90.7% of these isolates carried the substitution Ser83Phe in GyrA. No mutation was found in the QRDR of <it>gyrB</it>, <it>parC</it>, or <it>parE</it>. Plasmid mediated quinolone resistance genes including <it>qnr </it>and <it>aac(6')-Ib-cr </it>were not detected in any isolate. Twenty-two distinct pulsed field gel electrophoresis (PFGE) patterns were observed among <it>S. typhi</it>. Sixty-four isolates of <it>S. paratyphi </it>A belonged to one clone. Eighty-seven investigated inpatients were infected in the community. Six patients infected by <it>S. paratyphi </it>A had a travel history before infection.</p> <p>Conclusions</p> <p>Nalidixic acid-resistant <it>S. typhi </it>and <it>S. paratyphi </it>A blood isolates were highly prevalent in Shenzhen, China. PFGE showed the variable genetic diversity of nalidixic acid-resistant <it>S. typhi </it>and limited genetic diversity of nalidixic acid -resistant <it>S. paratyphi </it>A.</p

    De novo assembly and transcriptome characterization: novel insights into the natural resistance mechanisms of Microtus fortis against Schistosoma japonicum

    BACKGROUND: Microtus fortis is a non-permissive host of Schistosoma japonicum. It has natural resistance against schistosomes, although the precise resistance mechanisms remain unclear. The paucity of genetic information for M. fortis limits the use of available immunological methods. Thus, studies based on high-throughput sequencing technologies are required to obtain information about resistance mechanisms against S. japonicum. RESULTS: Using Illumina single-end technology, a de novo assembly of the M. fortis transcriptome produced 67,751 unigenes with an average length of 868 nucleotides. Comparisons were made between M. fortis before and after infection with S. japonicum using RNA-seq quantification analysis. The highest number of differentially expressed genes (DEGs) occurred two weeks after infection, and the highest number of down-regulated DEGs occurred three weeks after infection. Simultaneously, the strongest pathological changes in the liver were observed at week two. Gene ontology terms and pathways related to the DEGs revealed that up-regulated transcripts were involved in metabolism, immunity and inflammatory responses. Quantitative real-time PCR analysis showed that patterns of gene expression were consistent with RNA-seq results. CONCLUSIONS: After infection with S. japonicum, a defensive reaction in M. fortis commenced rapidly, increasing dramatically in the second week, and gradually decreasing three weeks after infection. The obtained M. fortis transcriptome and DEGs profile data demonstrated that natural and adaptive immune responses, play an important role in M. fortis immunity to S. japonicum. These findings provide a better understanding of the natural resistance mechanisms of M. fortis against schistosomes. Lateral sensitivity effect in normal accelerometer is eliminated because the beams which are thinner than 15 micrometers have been formed in the middle of the seismic mass. Based on the calculation of sensitivity and basic resonance frequency of two kinds of bulk micromachining accelerometers, the structure parameters of cantilever and double-side-supported accelerometer have been optimized by using the sensitivity-frequency product as the figure of merit of a structure. The different etching characteristics of {311} and {100} plane of silicon in KOH maskless anisotropic etching process have been investigated thoroughly and utilized in the fabrication of symmetric mass- beam structure. Special damping design has been proposed to reduce the damping ratio of the device in order to improve the dynamic performance of the accelerometer.     Tree-ring stable carbon isotope-based April-June relative humidity reconstruction since AD 1648 in Mt. Tianmu, China

    Based on accurate dating, we have determined the stable carbon isotope ratios (delta C-13) of five Cryptomeria fortunei specimens from Mt. Tianmu, a subtropical area in southern China. The five delta C-13 time series records are combined into a single representative delta C-13 time series using a &quot;numerical mix method.&quot; These are normalized to remove temporal variations of delta(13) C in atmospheric CO2 to obtain a carbon isotopic discrimination (Delta C-13) time series, in which we observe a distinct correlation between Delta C-13 and local April to June mean relative humidity (RH (AMJ) ) (n = 64, r = 0.858, p &lt; 0.0001). We use this relationship to reconstruct RH (AMJ) variations from ad 1648 to 2014 at Mt. Tianmu. The reconstructed sequence show that over the past 367 years, Mt. Tianmu area was relatively wet, but in the latter part of the twentieth century, under the influence of increasing global warming, it has experienced a sharp reduction in relative humidity. Spatial correlation analysis reveals a significant negative correlation between RH (AMJ) at Mt. Tianmu and Sea Surface Temperature (SSTs) in the western equatorial Pacific and Indian Ocean. In other words, there is a positive correlation between tree-ring delta C-13 in Mt. Tianmu and SSTs. Both observed and reconstructed RH (AMJ) show significant positive correlations with East Asian and South Asian monsoons from 1951 to 2014, which indicate that RH (AMJ) from Mt. Tianmu reflects the variability of the Asian summer monsoon intensity to a great extent. The summer monsoon has weakened since 1960. However, an increase in relative humidity since 2003 implies a recent enhancement in the summer monsoon

    Application of a novel strong promoter from Chinese fir (Cunninghamia lanceolate) in the CRISPR/Cas mediated genome editing of its protoplasts and transgenesis of rice and poplar

    Novel constitutive promoters are essential for plant biotechnology. Although in angiosperms, a number of promoters were applied in monocots or dicots genetic engineering, only a few promoters were used in gymnosperm. Here we identified two strong promoters (Cula11 and Cula08) from Chinese fir (C. lanceolate) by screening the transcriptomic data and preliminary promoter activity assays in tobacco. By using the newly established Chinese fir protoplast transient expression technology that enables in vivo molecular biology studies in its homologous system, we compared the activities of Cula11 and Cula08 with that of the commonly used promoters in genetic engineering of monocots or dicots, such as CaM35S, CmYLCV, and ZmUbi, and our results revealed that Cula11 and Cula08 promoters have stronger activities in Chinese fir protoplasts. Furthermore, the vector containing Cas gene driven by Cula11 promoter and sgRNA driven by the newly isolated CulaU6b polyIII promoters were introduced into Chinese fir protoplasts, and CRISPR/Cas mediated gene knock-out event was successfully achieved. More importantly, compared with the commonly used promoters in the genetic engineering in angiosperms, Cula11 promoter has much stronger activity than CaM35S promoter in transgenic poplar, and ZmUbi promoter in transgenic rice, respectively, indicating its potential application in poplar and rice genetic engineering. Overall, the novel putative constitutive gene promoters reported here will have great potential application in gymnosperm and angiosperm biotechnology, and the transient gene expression system established here will serve as a useful tool for the molecular and genetic analyses of Chinese fir genes

    Prevalence of Common Respiratory Viral Infections and Identification of Adenovirus in Hospitalized Adults in Harbin, China 2014 to 2017

    Background: Respiratory infections pose a great challenge in global health, and the prevalence of viral infection in adult patients has been poorly understood in northeast China. Harbin is one of the major cities in northeast China, and more than half of any given year in Harbin is occupied by winter. To reveal the viral etiology and seasonality in adult patients from Harbin, a 4-year consecutive survey was conducted in Harbin, China.Methods: From January 2014 to December 2017, specimens were obtained from adult patients admitted to the Second Affiliated Hospital of Harbin Medical University with lower respiratory tract infections. Sputum samples were examined by direct immunofluorescence assays to detect seven common respiratory viruses, including influenza virus (type A and B), parainfluenza virus (type 1 to 3), respiratory syncytial virus and adenovirus. Adenovirus positive samples were seeded onto A549 cells to isolate viral strains. Phylogenetic analysis was conducted on the highly variable region of adenoviral hexon gene.Results: A total of 1,300 hospitalized adult patients with lower respiratory tract infections were enrolled, in which 189 patients (14.5%) were detected as having at least one viral infection. The co-infection rate in this study was 25.9% (49/189). The dominant viral pathogen from 2014 to 2017 was parainfluenza virus, with a detection rate of 7.2%, followed by influenza virus, respiratory syncytial virus and adenovirus. Based on the climate seasons determined by daily average temperature, the highest overall viral detection rate was detected in spring (22.0%, 52/236), followed by winter (13.4%, 109/813), autumn (11.4%, 13/114) and summer (10.9%, 15/137). Adenovirus type 3 strains with slight variations were isolated from positive cases, which were closely related to the GB strain from the United States, as well as the Harbin04B strain isolated locally.Conclusion: This study demonstrated that common respiratory viruses were partially responsible for hospitalized lower respiratory tract infections in adult patients from Harbin, China, with parainfluenza virus as the dominant viral pathogen. Climate seasons could be rational indicators for the seasonality analysis of airborne viral infections. Future surveillance on viral mutations would be necessary to reveal the evolutionary history of respiratory viruses
