    Inclusion complexes of β and HPβ-cyclodextrin with α, β amyrin and in vitro anti-inflammatory activity

    α, β amyrin (ABAM) is a natural mixture of pentacyclic triterpenes that has a wide range of biological activities. ABAM is isolated from the species of the Burseraceae family, in which the species Protium is commonly found in the Amazon region of Brazil. The aim of this work was to develop inclusion complexes (ICs) of ABAM and β-cyclodextrin (βCD) and hydroxypropyl-β-cyclodextrin (HPβCD) by physical mixing (PM) and kneading (KN) methods. Interactions between ABAM and the CD's as well as the formation of ICs were confirmed by physicochemical characterization in the solid state by Fourier transform infrared (FTIR), scanning electron microscopy (SEM), X-ray diffraction (XRD), thermogravimetry (TG) and differential scanning calorimetry (DSC). Physicochemical characterization indicated the formation of ICs with both βCD and HPβCD. Such ICs were able to induce changes in the physicochemical properties of ABAM. In addition, the formation of ICs with cyclodextrins showed to be an effective and promising alternative to enhance the anti-inflammatory activity and safety of ABAM

    O Largo da Banana e a presença negra em São Paulo

    The Largo da Banana relates to the history of the black population in São Paulo. Acknowledged as one of São Paulo’s “cradle” of samba, samba musicians are its main spokespersons. Largo da Banana used to be located near the former Barra Funda Railway Station. From the first decades to around half the Twentieth century, informal laborers in the railway logistics used to gather together in that space. Within a precarious and unsteady daily life, they used to play samba and tiririca. In the fifties, the municipality built the Pacaembu Viaduct in that area, aiming to extend the homonym avenue beyond the railway road. In the sixties, samba musician Geraldo Filme wrote two songs in which he paid homage to Largo da Banana and regrettedits disappearance after the viaduct’s inauguration. In these and other of his songs, the artist recorded his perceptions on the life conditions of São Paulo’s blacks and samba musicians, their sociabilities, and also the urban changes he witnessed. Considering that his artistic work contributes to enlighten aspects of social reality, I analyze some of his songs to interpret the sociabilities at Largo da Banana, as well as the urban intervention in that location.O Largo da Banana está associado à história da população negra em São Paulo. Reconhecido como um dos “berços” do samba paulista, é sobretudo através dos próprios sambistas que essa história pode hoje ser conhecida. Localizado junto à antiga estação da Barra Funda, desde as primeiras décadas até meados do século passado, aquele espaço concentrou trabalhadores informais vinculados às atividades da ferrovia. Em meio a um cotidiano precário e instável, essa população realizava rodas de samba e de tiririca. Nos anos 1950, foi construído naquele local o Viaduto Pacaembu, que prolongava a avenida homônima para além davia férrea. A partir da década de 1960, o sambista negro Geraldo Filme compôs duas canções em que homenageava o Largo da Banana e lamentava seu desaparecimento após a inauguração do viaduto. Nessas e em outras composições, o artista registrou suas percepções sobre as condições de vida da população negra e sambista na cidade, suas sociabilidades, bem como sobre as transformações urbanas que testemunhou. Considerando que sua obra artística contribui para iluminar aspectos da realidade social, ela é adotada como fonte para analisar as formas de sociabilidade no Largo da Banana e a intervenção urbanística naquele local

    Catálogo Taxonômico da Fauna do Brasil: setting the baseline knowledge on the animal diversity in Brazil

    The limited temporal completeness and taxonomic accuracy of species lists, made available in a traditional manner in scientific publications, has always represented a problem. These lists are invariably limited to a few taxonomic groups and do not represent up-to-date knowledge of all species and classifications. In this context, the Brazilian megadiverse fauna is no exception, and the Catálogo Taxonômico da Fauna do Brasil (CTFB) (http://fauna.jbrj.gov.br/), made public in 2015, represents a database on biodiversity anchored on a list of valid and expertly recognized scientific names of animals in Brazil. The CTFB is updated in near real time by a team of more than 800 specialists. By January 1, 2024, the CTFB compiled 133,691 nominal species, with 125,138 that were considered valid. Most of the valid species were arthropods (82.3%, with more than 102,000 species) and chordates (7.69%, with over 11,000 species). These taxa were followed by a cluster composed of Mollusca (3,567 species), Platyhelminthes (2,292 species), Annelida (1,833 species), and Nematoda (1,447 species). All remaining groups had less than 1,000 species reported in Brazil, with Cnidaria (831 species), Porifera (628 species), Rotifera (606 species), and Bryozoa (520 species) representing those with more than 500 species. Analysis of the CTFB database can facilitate and direct efforts towards the discovery of new species in Brazil, but it is also fundamental in providing the best available list of valid nominal species to users, including those in science, health, conservation efforts, and any initiative involving animals. The importance of the CTFB is evidenced by the elevated number of citations in the scientific literature in diverse areas of biology, law, anthropology, education, forensic science, and veterinary science, among others

    Exploring Scoring Function Space. Developing Computational Models for Drug Discovery

    Background: The idea of scoring function space established a systems-level approach toaddress the development of models to predict binding affinity faced by those interested in drugdiscovery.Objective: Our goal here is to review the concept of scoring function space and how to explore it todevelop machine learning models to address protein-ligand binding affinity.Method: We searched the articles available in PubMed about the scoring function space. We alsoutilized crystallographic structures found in the protein data bank (PDB). We used datasets available atthe PDBbind, BindingDB, and Binding MOAD to illustrate how to integrate structural and binding data.Results: The application of systems-level approaches to address receptor-drug interactions allows us tohave a holistic view of the process of drug discovery. The scoring function space added flexibility tothe process since it makes it possible to see the drug discovery as a relationship involving mathematicalspaces.Conclusion: The application of the concept of scoring function space gave us an integrated view ofdrug discovery methods.     Characterization and Antiproliferative Activity of a Novel 2-Aminothiophene Derivative-β-Cyclodextrin Binary System

    The novel 2-aminothiophene derivative 2-amino-4,5,6,7-tetrahydrobenzo[b]thiophene-3-carbonitrile (6CN) has shown potential anti-proliferative activity in human cancer cell lines. However, the poor aqueous solubility of 6CN impairs its clinical use. This work aimed to develop binary 6CN-β-cyclodextrin (βCD) systems with the purpose of increasing 6CN solubility in water and therefore, to improve its pharmacological activity. The 6CN-βCD binary systems were prepared by physical mixing, kneading and rotary evaporation methods and further characterized by FTIR, XRD, DSC, TG and SEM. In addition, molecular modeling and phase solubility studies were performed. Finally, MTT assays were performed to investigate the cytostatic and anti-proliferative effects of 6CN-βCD binary systems. The characterization results show evident changes in the physicochemical properties of 6CN after the formation of the binary systems with βCD. In addition, 6CN was associated with βCD in aqueous solution and the solid state, which was confirmed by molecular modeling and the aforementioned characterization techniques. Phase solubility studies indicated that βCD forms stable 1:1 complexes with 6CN. The MTT assay demonstrated the cytostatic and anti-proliferative activities of 6CN-βCD binary systems and therefore, these might be considered as promising candidates for new anticancer drugs

    The Brazilian Registry of Adult Patient Undergoing Cardiovascular Surgery, the BYPASS Project: Results of the First 1,722 Patients

    Abstract Objective: To report the early results of the BYPASS project - the Brazilian registrY of adult Patient undergoing cArdiovaScular Surgery - a national, observational, prospective, and longitudinal follow-up registry, aiming to chart a profile of patients undergoing cardiovascular surgery in Brazil, assessing the data harvested from the initial 1,722 patients. Methods: Data collection involved institutions throughout the whole country, comprising 17 centers in 4 regions: Southeast (8), Northeast (5), South (3), and Center-West (1). The study population consists of patients over 18 years of age, and the types of operations recorded were: coronary artery bypass graft (CABG), mitral valve, aortic valve (either conventional or transcatheter), surgical correction of atrial fibrillation, cardiac transplantation, mechanical circulatory support and congenital heart diseases in adults. Results: 83.1% of patients came from the public health system (SUS), 9.6% from the supplemental (private insurance) healthcare systems; and 7.3% from private (out-of -pocket) clinic. Male patients comprised 66%, 30% were diabetics, 46% had dyslipidemia, 28% previously sustained a myocardial infarction, and 9.4% underwent prior cardiovascular surgery. Patients underwent coronary artery bypass surgery were 54.1% and 31.5% to valve surgery, either isolated or combined. The overall postoperative mortality up to the 7th postoperative day was 4%; for CABG was 2.6%, and for valve operations, 4.4%. Conclusion: This first report outlines the consecution of the Brazilian surgical cardiac database, intended to serve primarily as a tool for providing information for clinical improvement and patient safety and constitute a basis for production of research protocols
