    The rural-urban divide in road safety: the case of China

    Mainland China accounted for about 7% of the global road fatalities in 2008. Road crashes happening on Chinese roads were deadly. On average, one person died in every four reported traffic crashes. Despite the scarcity of data, substantial rural-urban differences were found. In the rural areas, higher-order Highways, roads with no lighting and some heavy vehicles warrant particular attention from road safety administrations. In the 2000s, the average number of road fatalities per 100 crashes on Expressways quadrupled. Furthermore, the rural-urban divide was not limited to inner provinces only but was found in a large part of the country. By 2008, nearly 70% of the provincial units were having larger shares of rural population. In the long term, only a national road safety strategy will provide the necessary holistic framework for addressing the road safety problems in China systematically.published_or_final_versio

    Cyclodextrin-PEI-Tat Polymer as a Vector for Plasmid DNA Delivery to Placenta Mesenchymal Stem Cells

    This study aims to modify a cyclodextrin-PEI-based polymer, PEI-β-CyD, with the TAT peptide for plasmid DNA delivery to placenta mesenchymal stem cells (PMSCs). By using the disulfide exchange between the SPDP-activated PEI-β-CyD and TAT peptide, the TAT-PEI-β-CyD polymer was fabricated and the success of this was confirmed by the presence of characteristic peaks for PEI (at δ 2.8-3.2 ppm), CyD (at δ 5.2, 3.8-4.0 and 3.4-3. 6 ppm) and TAT (at δ 1.6-1.9 and 6.8-7.2 ppm) in the 1H NMR spectrum of TAT-PEI-β-CyD. The polymer-plasmid-DNA polyplex could condense DNA at an N/P ratio of 7.0-8.0, and form nanoparticles with the size of 150.6±5.6 nm at its optimal N/P ratio (20/1). By examining the transfection efficiency and cytotoxicity of TAT-PEI-β-CyD, conjugation of the TAT peptide onto PEI-β-CyD was demonstrated to improve the transfection efficiency of PEI-β-CyD in PMSCs after 48 and 96 hours of post-transfection incubation. The viability of PEI-β-CyD-treated PMSCs was shown to be over 80% after 5 h of treatment and 24 h of post-treatment incubation. In summary, this study showed that the TAT-PEI-β-CyD polymer as a vector for plasmid DNA delivery to PMSCs and other cells warrants further investigations. © 2011 The Author(s).published_or_final_versionSpringer Open Choice, 21 Feb 201

    A Regression-based K nearest neighbor algorithm for gene function prediction from heterogeneous data

    BACKGROUND: As a variety of functional genomic and proteomic techniques become available, there is an increasing need for functional analysis methodologies that integrate heterogeneous data sources. METHODS: In this paper, we address this issue by proposing a general framework for gene function prediction based on the k-nearest-neighbor (KNN) algorithm. The choice of KNN is motivated by its simplicity, flexibility to incorporate different data types and adaptability to irregular feature spaces. A weakness of traditional KNN methods, especially when handling heterogeneous data, is that performance is subject to the often ad hoc choice of similarity metric. To address this weakness, we apply regression methods to infer a similarity metric as a weighted combination of a set of base similarity measures, which helps to locate the neighbors that are most likely to be in the same class as the target gene. We also suggest a novel voting scheme to generate confidence scores that estimate the accuracy of predictions. The method gracefully extends to multi-way classification problems. RESULTS: We apply this technique to gene function prediction according to three well-known Escherichia coli classification schemes suggested by biologists, using information derived from microarray and genome sequencing data. We demonstrate that our algorithm dramatically outperforms the naive KNN methods and is competitive with support vector machine (SVM) algorithms for integrating heterogenous data. We also show that by combining different data sources, prediction accuracy can improve significantly. CONCLUSION: Our extension of KNN with automatic feature weighting, multi-class prediction, and probabilistic inference, enhance prediction accuracy significantly while remaining efficient, intuitive and flexible. This general framework can also be applied to similar classification problems involving heterogeneous datasets

    Unusual Location of the Geotail Magnetopause Near Lunar Orbit: A Case Study

    The Earth's magnetopause is highly variable in location and shape and is modulated by solar wind conditions. On 8 March 2012, the ARTEMIS probes were located near the tail current sheet when an interplanetary shock arrived under northward interplanetary magnetic field conditions and recorded an abrupt tail compression at ∼(‐60, 0, ‐5) RE in Geocentric Solar Ecliptic coordinate in the deep magnetotail. Approximately 10 minutes later, the probes crossed the magnetopause many times within an hour after the oblique interplanetary shock passed by. The solar wind velocity vector downstream from the shock was not directed along the Sun‐Earth line but had a significant Y component. We propose that the compressed tail was pushed aside by the appreciable solar wind flow in the Y direction. Using a virtual spacecraft in a global magnetohydrodynamic (MHD) simulation, we reproduce the sequence of magnetopause crossings in the X‐Y plane observed by ARTEMIS under oblique shock conditions, demonstrating that the compressed magnetopause is sharply deflected at lunar distances in response to the shock and solar wind VY effects. The results from two different global MHD simulations show that the shocked magnetotail at lunar distances is mainly controlled by the solar wind direction with a timescale of about a quarter hour, which appears to be consistent with the windsock effect. The results also provide some references for investigating interactions between the solar wind/magnetosheath and lunar nearside surface during full moon time intervals, which should not happen in general

    State based model of long-term potentiation and synaptic tagging and capture

    Recent data indicate that plasticity protocols have not only synapse-specific but also more widespread effects. In particular, in synaptic tagging and capture (STC), tagged synapses can capture plasticity-related proteins, synthesized in response to strong stimulation of other synapses. This leads to long-lasting modification of only weakly stimulated synapses. Here we present a biophysical model of synaptic plasticity in the hippocampus that incorporates several key results from experiments on STC. The model specifies a set of physical states in which a synapse can exist, together with transition rates that are affected by high- and low-frequency stimulation protocols. In contrast to most standard plasticity models, the model exhibits both early- and late-phase LTP/D, de-potentiation, and STC. As such, it provides a useful starting point for further theoretical work on the role of STC in learning and memory

    Incorporating background frequency improves entropy-based residue conservation measures

    BACKGROUND: Several entropy-based methods have been developed for scoring sequence conservation in protein multiple sequence alignments. High scoring amino acid positions may correlate with structurally or functionally important residues. However, amino acid background frequencies are usually not taken into account in these entropy-based scoring schemes. RESULTS: We demonstrate that using a relative entropy measure that incorporates amino acid background frequency results in improved performance in identifying functional sites from protein multiple sequence alignments. CONCLUSION: Our results suggest that the application of appropriate background frequency information may lead to more biologically relevant results in many areas of bioinformatics
