    Recombinant luminescent bacteria for measuring bioavailable arsenite and antimonite

    Luminescent bacterial strains for the measurement of bioavailable arsenite and antimony were constructed, The expression of firefly luciferase was controlled by the regulatory unit of the ars operon of Staphylococcus aureus plasmid pI258 in recombinant plasmid pT0021, with S. aureus RN4220, Bacillus subtilis BR151, and Escherichia coli MC1061 as host strains, Strain RN4220(pT0021) was found to be the most sensitive for metal detection responding to arsenite, antimonite, and cadmium, the lowest detectable concentrations being 100, 33, and 330 nhl, respectively, Strains BR151(pT0021) and MC1061(pT0021) responded to arsenite, arsenate, antimonite, and cadmium, the lowest detectable concentrations being 3.3 and 330 mu M and 330 and 330 nM with BR151(pT0021), respectively, and 3.3, 33, 3.3, and 33 CIM with MC1061(pT0021), respectively, In the absence of the mentioned ions, the expression of luciferase was repressed and only a small amount of background light was emitted, Other ions did not notably interfere with the measurement in any of the strains tested, Freeze-drying of the cells did not decrease the sensitivity of the detection of arsenite; however, the induction coefficients were somewhat lower

    Vaihtoehtoisia korvausmalleja hammashoidon sairausvakuutusjärjestelmän kehittämiseksi

    Tässä tutkimuksessa tavoitteena oli tarjota vaihtoehtoja keskusteluun hammashoidon sairausvakuutuskorvausjärjestelmän uudistamiseksi. Vaihtoehtoisten korvausmallien, porrasmallin ja tilimallin, korvauksia verrattiin vuonna 2009 voimassa olleen taksamallin korvauksiin. Laskelmien avulla arvioitiin, ketkä korvausta saaneet hyötyisivät ja ketkä häviäisivät. Tutkimuksen aineistona oli otos henkilöistä, jotka saivat sairausvakuutuksen sairaanhoitokorvausta yksityisen hammashoidon kustannuksista vuonna 2009. Otoksen koko oli 100 000 henkilöä, joilla oli yhteensä 503 093 toimenpidettä. Aineistossa oli tiedot henkilöille tehdyistä toimenpiteistä, toimenpiteistä maksetuista palkkioista ja saaduista korvauksista. Taustamuuttujia aineistossa olivat sukupuoli, ikä, kotikunta ja valtionveronalaiset vuositulot. Laskelmissa käytettiin kahta vaihtoehtoista korvausmallia: porrasmallia ja tilimallia. Porrasmallissa korvaus nousi, kun asiakkaan kustannukset nousivat. Tilimallissa kaikilla oli käytössään vuosittain samansuuruinen korvaussumma. Tutkimusaineiston todellisille toimenpiteille laskettiin vaihtoehtoisten mallien mukaiset korvaukset. Laskelmissa oletettiin, että henkilöiden toimenpiteet ja toimenpiteiden palkkiot säilyvät ennallaan. Korvausmallit pidettiin kustannusneutraaleina, joten niiden korvaussumma oli korkeintaan vuoden 2009 tasolla. Laskelmat osoittivat, että tilimallista hyötyisi kolme neljästä korvauksen saajasta. Korvausprosentti oli taksamallia korkeampi, kun vuosikustannus eivät ylittäneet 500 euroa. Porrasmallin käyttöönotosta hyötyisi yksi kymmenestä korvauksen saajasta. Korvausprosentti kasvoi, jos vuosikustannus ylitti 800 euroa. Sitä pienemmällä vuosikustannuksella korvausprosentti pieneni taksamalliin verrattuna. Jos sairausvakuutuksen korvausjärjestelmää halutaan uudistaa kustannusneutraalisti ja samalla parantaa edellytyksiä käyttää yksityisiä hammashoitopalveluja, tulisi soveltaa tilimallia porrasmallin asemesta

    Antibody responses to nasopharyngeal carriage of Streptococcus pneumoniae in adults: A longitudinal household study

    Background. Natural immunity to Streptococcus pneumoniae is thought to be induced by exposure to S. pneumoniae or cross-reactive antigens. No longitudinal studies of carriage of and immune responses to S. pneumoniae have been conducted using sophisticated immunological laboratory techniques.Methods. We enrolled 121 families with young children into this study. Nasopharyngeal (NP) swabs were collected monthly for 10 months from all family members and were cultured in a standard fashion. Cultured S. pneumoniae isolates were serotyped. At the beginning (month 0) and end (month 10) of the study, venous blood was collected from family members 118 years old. Serotype-specific antipolysaccharide immunoglobulin G (IgG) and functional antibody and antibodies to pneumolysin, pneumococcal surface protein A (PspA), and pneumococcal surface antigen A (PsaA) were measured in paired serum samples.Results. Levels of anticapsular IgG increased significantly after carriage of serotypes 9V, 14, 18C, 19F, and 23F by an individual or family member. For serotype 14, a higher level of anticapsular IgG at the beginning of the study was associated with reduced odds of carriage (P = .0006). There was a small (similar to 20%) but significant increase in titers of antibodies to PsaA and pneumolysin but no change in titers of antibody to PspA.Conclusions. Adults respond to NP carriage by mounting anticapsular and weak antiprotein antibody responses, and naturally induced anticapsular IgG can prevent carriage

    Small GTP-binding proteins of the rab family have been implicated as regulators of membrane traffic along the biosynthetic and endocytic pathways in eukaryotic cells. We have investigated the localization and function of rab8, closely related to the yeast YPT1/SEC4 gene products. Confocal immunofluorescence microscopy and immunoelectron microscopy on filter-grown MDCK cells demonstrated that, rab8 was localized to the Golgi region, vesicular structures, and to the basolateral plasma membrane. Two-dimensional gel electrophoresis showed that rab8p was highly enriched in immuno-isolated basolateral vesicles carrying vesicular stomatitis virus-glycoprotein (VSV-G) but was absent from vesicles transporting the hemagglutinin protein (HA) of influenza virus to the apical cell surface

    In simple epithelial cells, apical and basolateral proteins are sorted into separate vesicular carriers before delivery to the appropriate plasma membrane domains. To dissect the putative sorting machinery, we have solubilized Golgi-derived transport vesicles with the detergent CHAPS and shown that an apical marker, influenza haemagglutinin (HA), formed a large complex together with several integral membrane proteins. Remarkably, a similar set of CHAPS-insoluble proteins was found after solubilization of a total cellular membrane fraction. This allowed the cloning of a cDNA encoding one protein of this complex, VIP21 (Vesicular Integral-membrane Protein of 21 kD). The transiently expressed protein appeared on the Golgi-apparatus, the plasma membrane and vesicular structures. We propose that VIP21 is a component of the molecular machinery of vesicular transport

    Independent component analysis for tensor-valued data

    In preprocessing tensor-valued data, e.g., images and videos, a common procedure is to vectorize the observations and subject the resulting vectors to one of the many methods used for independent component analysis (ICA). However, the tensor structure of the original data is lost in the vectorization and, as a more suitable alternative, we propose the matrix- and tensor fourth order blind identification (MFOBI and TFOBI). In these tensorial extensions of the classic fourth order blind identification (FOBI) we assume a Kronecker structure for the mixing and perform FOBI simultaneously on each direction of the observed tensors. We discuss the theory and assumptions behind MFOBI and TFOBI and provide two different algorithms and related estimates of the unmixing matrices along with their asymptotic properties. Finally, simulations are used to compare the method's performance with that of classical FOBI for vectorized data and we end with a real data clustering example. (C) 2017 Elsevier Inc.     Acetylated and Methylated beta-Cyclodextrins as Viable Soluble Supports for the Synthesis of Short 2 '-Oligodeoxyribo-nucleotides in Solution

    Novel soluble supports for oligonucleotide synthesis 11a-c have been prepared by immobilizing a 5'-O-protected 3'-O-(hex-5-ynoyl) thymidine (6 or 7) to peracetylated or permethylated 6-deoxy-6-azido-beta-cyclodextrins 10a or 10b by Cu(I)-promoted 1,3-dipolar cycloaddition. The applicability of the supports to oligonucleotide synthesis by the phosphoramidite strategy has been demonstrated by assembling a 3'-TTT-5' trimer from commercially available 5'-O-(4,4'-dimethoxytrityl) thymidine 3'-phosphoramidite. To simplify the coupling cycle, the 5'-O-(4,4'-dimethoxytrityl) protecting group has been replaced with an acetal that upon acidolytic removal yields volatile products. For this purpose, 5'-O-(1-methoxy-1-methylethyl)-protected 3'-(2-cyanoethyl-N, N-diisopropyl-phosphoramidite) s of thymidine (5a), N-4-benzoyl-2'-deoxycytidine (5b) and N-6-benzoyl-2'-deoxyadenosine (5c) have been synthesized and utilized in synthesis of a pentameric oligonucleotide 3'-TTCAT-5' on the permethylated cyclodextrin support 11c

    Executive function subdomains are associated with post-stroke functional outcome and permanent institutionalization

    Background and purposeImpairment of executive functions (EFs) is a common cognitive symptom post-stroke and affects independence in daily activities. Previous studies have often relied on brief cognitive tests not fully considering the wide spectrum of EF subdomains. A detailed assessment of EFs was used to examine which of the subdomains and tests have the strongest predictive value on post-stroke functional outcome and institutionalization in long-term follow-up. MethodsA subsample of 62 patients from the Helsinki Stroke Aging Memory Study was evaluated with a battery of seven neuropsychological EF tests 3 months post-stroke and compared to 39 healthy control subjects. Functional impairment was evaluated with the modified Rankin Scale (mRS) and Instrumental Activities of Daily Living (IADL) scale at 3 months, and with the mRS at 15 months post-stroke. Institutionalization was reviewed from the national registers of permanent hospital admissions in up to 21-year follow-up. ResultsThe stroke group performed more poorly than the control group in multiple EF tests. Tests of inhibition, set shifting, initiation, strategy formation and processing speed were associated with the mRS and IADL scale in stroke patients. EF subdomain scores of inhibition, set shifting and processing speed were associated with functional outcome. In addition, inhibition was associated with the risk for earlier institutionalization. ConclusionsExecutive function was strongly associated with post-stroke functional impairment. In follow-up, poor inhibition was related to earlier permanent institutionalization. The results suggest the prognostic value of EF subdomains after stroke.Peer reviewe
