Regulation of heterogenous lexA expression in staphylococcus aureus by an antisense RNA originating from transcriptional read-through upon natural mispairings in the sbrB intrinsic terminator
Bacterial genomes are pervasively transcribed, generating a wide variety of antisense RNAs (asRNAs). Many of them originate from transcriptional read-through events (TREs) during the transcription termination process. Previous transcriptome analyses revealed that the lexA gene from Staphylococcus aureus, which encodes the main SOS response regulator, is affected by the presence of an asRNA. Here, we show that the lexA antisense RNA (lexA-asRNA) is generated by a TRE on the intrinsic terminator (TTsbrB) of the sbrB gene, which is located downstream of lexA, in the opposite strand. Transcriptional read-through occurs by a natural mutation that destabilizes the TTsbrB structure and modifies the efficiency of the intrinsic terminator. Restoring the mispairing mutation in the hairpin of TTsbrB prevented lexA-asRNA transcription. The level of lexA-asRNA directly correlated with cellular stress since the expressions of sbrB and lexA-asRNA depend on the stress transcription factor SigB. Comparative analyses revealed strain-specific nucleotide polymorphisms within TTsbrB, suggesting that this TT could be prone to accumulating natural mutations. Sensory deprivation in Staphylococcus aureus
Bacteria use two-component systems (TCSs) to sense and respond to environmental changes. The core genome of the major human pathogen Staphylococcus aureus encodes 16 TCSs, one of which (WalRK) is essential. Here we show that S. aureus can be deprived of its complete sensorial TCS network and still survive under growth arrest conditions similarly to wild-type bacteria. Under replicating conditions, however, the WalRK system is necessary and sufficient to maintain bacterial growth, indicating that sensing through TCSs is mostly dispensable for living under constant environmental conditions. Characterization of S. aureus derivatives containing individual TCSs reveals that each TCS appears to be autonomous and self-sufficient to sense and respond to specific environmental cues, although some level of cross-regulation between non-cognate sensor-response regulator pairs occurs in vivo. This organization, if confirmed in other bacterial species, may provide a general evolutionarily mechanism for flexible bacterial adaptation to life in new niches
Synthesis, microstructure and volumetry of novel metal thiocyanate ionic liquids with [BMIM] cation
We present a new family of ionic liquids (ILs) with a common cation, 1-butyl-3-methyl imidazolium, the popular [BMIM]+ (also written C4C1Im+) and a variety of anionic complexes (also called adducts) based in thiocyanate (SCN)?: one blank compound, BMIM(SCN), and ten doped with metals having different oxidation states: Al+3, Mn+2, Fe+3, Cr+3, Ni+2, Hg+2, Zn+2, Co+2, Cd+2 and Cu+, forming, respectively, [BMIM]3[Al(SCN)6], [BMIM]4 Mn(SCN)6, [BMIM]3 Fe(SCN)6, [BMIM]3 Cr(SCN)6, [BMIM]4 Ni(SCN)6, [BMIM]2 Hg(SCN)4, [BMIM]2 Zn(SCN)4, [BMIM]2 Co(SCN)4, [BMIM]2 Cd(SCN)4 and [BMIM]3 Cu(SCN)4. All of them were synthesized by us, except the blank IL and the Co thiocyanate, which are commercial. Obtained products have been characterized by NMR, and also by electrospray ionization, MS-ES, which allows the determination of the new ILs purities. Then, compounds have been analyzed using FT-IR and Raman spectroscopy. In addition, magnetic susceptibility and refractive index measurements were performed in some of the compounds studied, as well as thermal characterization using DSC and TGA. Finally, experimental measurements of density on all those ILs have been performed, and for some of the samples in a broad temperature range (about 100 K). In spite of being very similar compounds from the chemical point of view, they present quite different physical properties, including optical, thermal and magnetic ones? Also, they show different oxidation states (one with +1, six with +2 and other three with +3). We guess that this family of ILs will have interesting applications, mainly for photonic devices
Impact of cross-section uncertainties on supernova neutrino spectral parameter fitting in the Deep Underground Neutrino Experiment
A primary goal of the upcoming Deep Underground Neutrino Experiment (DUNE) is to measure the O(10) MeV neutrinos produced by a Galactic core-collapse supernova if one should occur during the lifetime of the experiment. The liquid-argon-based detectors planned for DUNE are expected to be uniquely sensitive to the νe component of the supernova flux, enabling a wide variety of physics and astrophysics measurements. A key requirement for a correct interpretation of these measurements is a good understanding of the energy-dependent total cross section σ(Eν) for charged-current νe absorption on argon. In the context of a simulated extraction of supernova νe spectral parameters from a toy analysis, we investigate the impact of σ(Eν) modeling uncertainties on DUNE's supernova neutrino physics sensitivity for the first time. We find that the currently large theoretical uncertainties on σ(Eν) must be substantially reduced before the νe flux parameters can be extracted reliably; in the absence of external constraints, a measurement of the integrated neutrino luminosity with less than 10% bias with DUNE requires σ(Eν) to be known to about 5%. The neutrino spectral shape parameters can be known to better than 10% for a 20% uncertainty on the cross-section scale, although they will be sensitive to uncertainties on the shape of σ(Eν). A direct measurement of low-energy νe-argon scattering would be invaluable for improving the theoretical precision to the needed level

El Proyecto de Aplicación Profesional nombrado “Desarrollo de Clústers de Turismo Regional” tiene como principal objetivo la aplicación de conocimientos y habilidades obtenidos durante la carrera profesional, por medio del diseño de una planeación turística de la región productora de Raicilla, como resultado, se contribuirá al desarrollo social de la misma por medio de una propuesta innovadora así como un modelo turístico sostenible. Tomando como referencia el desarrollo del proyecto del periodo anterior (Primavera 2022), se busca la creación de un diagnóstico de la zona indicada que compone a pueblos que cuentan con la denominación de origen, esto por medio de las visitas de campo guiadas tanto por el Mtro. Mauricio Margulles como por los raicilleros dueños y trabajadores de las tabernas seleccionadas. Posteriormente se busca la creación y el desarrollo de una propuesta que sea implementable y trabaje en conjunto con el Consejo Promotor de la Raicilla, siendo benéfica para la sociedad. Entre los principales resultados a destacar están la definición de problemáticas de las localidades de Mascota, la Yerbabuena y Navidad, la creación de una guía genérica dirigida hacia los taberneros que facilite el proceso de las visitas guiadas a turistas y la guía del contenido de los videos entregables posteriores a las visitas de campo, orientado para futuros alumnos que opten por llevar el PAP, entre otros productos que se desarrollarán a lo largo del documento.ITESO, A.C
Evidence for a Chandrasekhar-mass explosion in the Ca-strong 1991bg-like type la supernova 2016hnk
Aims. We present a comprehensive dataset of optical and near-infrared photometry and spectroscopy of type Ia supernova (SN) 2016hnk, combined with integral field spectroscopy (IFS) of its host galaxy, MCG-01-06-070, and nearby environment. Our goal with this complete dataset is to understand the nature of this peculiar object. Methods. Properties of the SN local environment are characterized by means of single stellar population synthesis applied to IFS observations taken two years after the SN exploded. We performed detailed analyses of SN photometric data by studying its peculiar light and color curves. SN 2016hnk spectra were compared to other 1991bg-like SNe Ia, 2002es-like SNe Ia, and Ca-rich transients. In addition, we used abundance stratification modeling to identify the various spectral features in the early phase spectral sequence and also compared the dataset to a modified non-LTE model previously produced for the sublumnious SN 1999by. Results. SN 2016hnk is consistent with being a subluminous (MB = -16.7 mag, sBV=0.43 ± 0.03), highly reddened object. The IFS of its host galaxy reveals both a significant amount of dust at the SN location, residual star formation, and a high proportion of old stellar populations in the local environment compared to other locations in the galaxy, which favors an old progenitor for SN 2016hnk. Inspection of a nebular spectrum obtained one year after maximum contains two narrow emission lines attributed to the forbidden [Ca ii] λλ7291,7324 doublet with a Doppler shift of 700 km s-1. Based on various observational diagnostics, we argue that the progenitor of SN 2016hnk was likely a near Chandrasekhar-mass (MCh) carbon-oxygen white dwarf that produced 0.108 M of 56Ni. Our modeling suggests that the narrow [Ca ii] features observed in the nebular spectrum are associated with 48Ca from electron capture during the explosion, which is expected to occur only in white dwarfs that explode near or at the MCh limit
Base Pairing Interaction between 5'- and 3'-UTRs Controls icaR mRNA Translation in Staphylococcus aureus.
The presence of regulatory sequences in the 3' untranslated region (3'-UTR) of eukaryotic mRNAs controlling RNA stability and translation efficiency is widely recognized. In contrast, the relevance of 3'-UTRs in bacterial mRNA functionality has been disregarded. Here, we report evidences showing that around one-third of the mapped mRNAs of the major human pathogen Staphylococcus aureus carry 3'-UTRs longer than 100-nt and thus, potential regulatory functions. We selected the long 3'-UTR of icaR, which codes for the repressor of the main exopolysaccharidic compound of the S. aureus biofilm matrix, to evaluate the role that 3'-UTRs may play in controlling mRNA expression. We showed that base pairing between the 3'-UTR and the Shine-Dalgarno (SD) region of icaR mRNA interferes with the translation initiation complex and generates a double-stranded substrate for RNase III. Deletion or substitution of the motif (UCCCCUG) within icaR 3'-UTR was sufficient to abolish this interaction and resulted in the accumulation of IcaR repressor and inhibition of biofilm development. Our findings provide a singular example of a new potential post-transcriptional regulatory mechanism to modulate bacterial gene expression through the interaction of a 3'-UTR with the 5'-UTR of the same mRNA
Acceptability and feasibility of a virtual community of practice to primary care professionals regarding patient empowerment: A qualitative pilot study
Background: Virtual communities of practice (vCoPs) facilitate online learning via the exchange of experiences and knowledge between interested participants. Compared to other communities, vCoPs need to overcome technological structures and specific barriers. Our objective was to pilot the acceptability and feasibility of a vCoP aimed at improving the attitudes of primary care professionals to the empowerment of patients with chronic conditions. Methods: We used a qualitative approach based on 2 focus groups: one composed of 6 general practitioners and the other of 6 practice nurses. Discussion guidelines on the topics to be investigated were provided to the moderator. Sessions were audio-recorded and transcribed verbatim. Thematic analysis was performed using the ATLAS-ti software. Results: The available operating systems and browsers and the lack of suitable spaces and time were reported as the main difficulties with the vCoP. The vCoP was perceived to be a flexible learning mode that provided up-to-date resources applicable to routine practice and offered a space for the exchange of experiences and approaches. Conclusions: The results from this pilot study show that the vCoP was considered useful for learning how to empower patients. However, while vCoPs have the potential to facilitate learning and as shown create professional awareness regarding patient empowerment, attention needs to be paid to technological and access issues and the time demands on professionals. We collected relevant inputs to improve the features, content and educational methods to be included in further vCoP implementation. Trial registration: ClinicalTrials.gov, NCT02757781.
