    A retrospective study of cryptorchidectomy in horses: Diagnosis, treatment, outcome and complications in 70 cases

    The aim of the study was to investigate the breed predisposition and the diagnostic and surgical management of horses referred for cryptorchidism. The breed, localization of retained testis, diagnosis, type of surgical treatment and complications were analyzed. Seventy horses were included in the study; the Western Riding horse breeds were the most affected (Quarter Horse 34/70, 48.5%; Appaloosa 9/70, 12.8%). In unilateral cryptorchids (65/70, 92.8%) the most common location for a retained testis was the left abdomen (28/65, 43%), while in bilateral cryptorchids (5/70, 7.1%), bilateral abdominal retention was the most frequent (3/5, 6%). Information about testis localization was achieved through transabdominal ultrasound (30/49 cases, 61.2%), through per rectum palpation (21/49 cases, 42.9%) and through inguinal palpation (14/49 cases, 28.9%). Cryptorchidectomy was achieved with standing laparoscopy (44/70 cases, 62.8%), or with open inguinal orchiectomy in general anesthesia (26/70 cases, 37.2%). Complications during laparoscopy were spleen puncture (1/44, 2.2%), a self-limiting bleeding from the spermatic cord (10/44 cases, 22.7%), hyperthermia (3/44 cases, 6.8%), and emphysema (15/44, 34%). During inguinal open cryptorchidectomy difficulties with identifying the inguinal testis during surgery (8/26 cases, 30.8%) and a moderate and self-limiting swelling of the inguinal region after surgery (17/26, 65.4%) were observed. For orchiectomy, a standing laparoscopy was confirmed as the preferred procedure for an abdominally retained testis with almost no complications

    Mules are hybrids bred from the mating of a jack donkey and a horse mare, known for their strength and resistance and still used to work in agriculture. Although they have been for long considered sterile, evidence of estrus cycle has been demonstrated together with abnormal behavior related to ovarian activity. In this study, a bilateral standing laparoscopic ovariectomy technique using the LigaSure technology was applied in 10 mare mules for treating unwanted behavioral patterns. The technique was effectively performed on these animals avoiding the risk of general anesthesia, and the use of the LigaSure technology allowed good hemostasis and reduced surgical time.     Hybrid machine learning approach for gully erosion mapping susceptibility at a watershed scale

    Gully erosion is a serious threat to the state of ecosystems all around the world. As a result, safeguarding the soil for our own benefit and from our own actions is a must for guaranteeing the longterm viability of a variety of ecosystem services. As a result, developing gully erosion susceptibility maps (GESM) is both suggested and necessary. In this study, we compared the effectiveness of three hybrid machine learning (ML) algorithms with the bivariate statistical index frequency ratio (FR), named random forest-frequency ratio (RF-FR), support vector machine-frequency ratio (SVM-FR), and naïve Bayes-frequency ratio (NB-FR), in mapping gully erosion in the GHISS watershed in the northern part of Morocco. The models were implemented based on the inventory mapping of a total number of 178 gully erosion points randomly divided into 2 groups (70% of points were used for training the models and 30% of points were used for the validation process), and 12 conditioning variables (i.e., elevation, slope, aspect, plane curvature, topographic moisture index (TWI), stream power index (SPI), precipitation, distance to road, distance to stream, drainage density, land use, and lithology). Using the equal interval reclassification method, the spatial distribution of gully erosion was categorized into five different classes, including very high, high, moderate, low, and very low. Our results showed that the very high susceptibility classes derived using RF-FR, SVM-FR, and NB-FR models covered 25.98%, 22.62%, and 27.10% of the total area, respectively. The area under the receiver (AUC) operating characteristic curve, precision, and accuracy were employed to evaluate the performance of these models. Based on the receiver operating characteristic (ROC), the results showed that the RF-FR achieved the best performance (AUC = 0.91), followed by SVM-FR (AUC = 0.87), and then NB-FR (AUC = 0.82), respectively. Our contribution, in line with the Sustainable Development Goals (SDGs), plays a crucial role for understanding and identifying the issue of “where and why” gully erosion occurs, and hence it can serve as a first pathway to reducing gully erosion in this particular area

    Lung adenocarcinoma originates from retrovirus infection of proliferating type 2 pneumocytes during pulmonary post-natal development or tissue repair

    Jaagsiekte sheep retrovirus (JSRV) is a unique oncogenic virus with distinctive biological properties. JSRV is the only virus causing a naturally occurring lung cancer (ovine pulmonary adenocarcinoma, OPA) and possessing a major structural protein that functions as a dominant oncoprotein. Lung cancer is the major cause of death among cancer patients. OPA can be an extremely useful animal model in order to identify the cells originating lung adenocarcinoma and to study the early events of pulmonary carcinogenesis. In this study, we demonstrated that lung adenocarcinoma in sheep originates from infection and transformation of proliferating type 2 pneumocytes (termed here lung alveolar proliferating cells, LAPCs). We excluded that OPA originates from a bronchioalveolar stem cell, or from mature post-mitotic type 2 pneumocytes or from either proliferating or non-proliferating Clara cells. We show that young animals possess abundant LAPCs and are highly susceptible to JSRV infection and transformation. On the contrary, healthy adult sheep, which are normally resistant to experimental OPA induction, exhibit a relatively low number of LAPCs and are resistant to JSRV infection of the respiratory epithelium. Importantly, induction of lung injury increased dramatically the number of LAPCs in adult sheep and rendered these animals fully susceptible to JSRV infection and transformation. Furthermore, we show that JSRV preferentially infects actively dividing cell in vitro. Overall, our study provides unique insights into pulmonary biology and carcinogenesis and suggests that JSRV and its host have reached an evolutionary equilibrium in which productive infection (and transformation) can occur only in cells that are scarce for most of the lifespan of the sheep. Our data also indicate that, at least in this model, inflammation can predispose to retroviral infection and cancer

    Novel NPM1 exon 5 mutations and gene fusions leading to aberrant cytoplasmic nucleophosmin in AML

    Nucleophosmin (NPM1) mutations in acute myeloid leukemia (AML) affect exon 12, but also sporadically affect exons 9 and 11, causing changes at the protein C-terminal end (tryptophan loss, nuclear export signal [NES] motif creation) that lead to aberrant cytoplasmic NPM1 (NPM1c+), detectable by immunohistochemistry. Combining immunohistochemistry and molecular analyses in 929 patients with AML, we found non–exon 12 NPM1 mutations in 5 (1.3%) of 387 NPM1c+ cases. Besides mutations in exons 9 (n = 1) and 11 (n = 1), novel exon 5 mutations were discovered (n = 3). Another exon 5 mutation was identified in an additional 141 patients with AML selected for wild-type NPM1 exon 12. Three NPM1 rearrangements (NPM1/RPP30, NPM1/SETBP1, NPM1/CCDC28A) were detected and characterized among 13 979 AML samples screened by cytogenetic/fluorescence in situ hybridization and RNA sequencing. Functional studies demonstrated that in AML cases, new NPM1 proteins harbored an efficient extra NES, either newly created or already present in the fusion partner, ensuring its cytoplasmic accumulation. Our findings support NPM1 cytoplasmic relocation as critical for leukemogenesis and reinforce the role of immunohistochemistry in predicting AML-associated NPM1 genetic lesions. This study highlights the need to develop new assays for molecular diagnosis and monitoring of NPM1-mutated AML

    Laparoscopic cryptorchidectomy in standing horse using the ligasure™

    A standing laparoscopic technique for cryptorchidectomy of horses using theLigaSure ™ device for division of the spermatic cord isdescribed. Twenty horses were referred for suspected unilateral cryptorchidism.After preparation, all horses underwent standing laparoscopy. In every horse thefirst trocar was positioned in the right flank; if a retained testis waseventually detected, two other laparoscopic portals were made in the same flankvertically. In case of left cryptorchidism three portals were created on theleft flank in a specular position. The first instrument portal was used to graspthe testicle with traumatic forceps while the second portal was used to insertthe LigaSure ™ device and dissect the spermatic cord. Thetesticle was removed through an enlarged portal; in three cases an Endopouch Probasket (Ethicon Endosurgery) was used for withdrawal of the testis. Nocomplications occurred during or after surgery. Mean time for dieresis of thespermatic cord was 8 minutes; 60 minutes was the mean time required for theoverall procedure from sedation to the exit of the horse from the stock (range40 - 80 min.). In our opinion the use of the LigaSure ™ systemseems to reduce surgical time, tissue damage and warrants good haemostasis
