    Aerosynthesis: Growth of Vertically-aligned Carbon Nanofibres with Air DC Plasma

    Vertically-aligned carbon nanofibres (VACNFs) have been synthesized in a mixture of acetone and air using catalytic DC plasma-enhanced chemical vapour deposition. Typically, ammonia or hydrogen is used as an etchant gas in the mixture to remove carbon that otherwise passivates the catalyst surface and impedes growth. Our demonstration of the use of air as the etchant gas opens up the possibility that ion etching could be sufficient to maintain the catalytic activity state during synthesis. It also demonstrates a path toward growing VACNFs in the open atmosphere

    Enhanced self-field critical current density of nano-composite YBa(2)Cu(3)O(7) thin films grown by pulsed-laser deposition

Copyright @ EPLA, 2008.Enhanced self-field critical current density Jc of novel, high-temperature superconducting thin films is reported. Layers are deposited on (001) MgO substrates by laser ablation of YBa2Cu3O7−ή(Y-123) ceramics containing Y2Ba4CuMOx (M-2411, M=Ag, Nb, Ru, Zr) nano-particles. The Jc of films depends on the secondary-phase content of the ceramic targets, which was varied between 0 and 15 mol%. Composite layers (2 mol% of Ag-2411 and Nb-2411) exhibit Jc values at 77 K of up to 5.1 MA/cm2, which is 3 to 4 times higher than those observed in films deposited from phase pure Y-123 ceramics.     Three novel rice genes closely related to the <em>Arabidopsis</em> <sub>9<em>IRX</em></sub>, <sub>9<em>IRXL</em></sub>, and<sub>14<em> IRX</em></sub> genes and their roles in xylan biosynthesis

    Xylan is the second most abundant polysaccharide on Earth, and represents a major component of both dicot wood and the cell walls of grasses. Much knowledge has been gained from studies of xylan biosynthesis in the model plant, Arabidopsis. In particular, the irregular xylem (irx) mutants, named for their collapsed xylem cells, have been essential in gaining a greater understanding of the genes involved in xylan biosynthesis. In contrast, xylan biosynthesis in grass cell walls is poorly understood. We identified three rice genes Os07g49370 (OsIRX9), Os01g48440 (OsIRX9L), and Os06g47340 (OsIRX14), from glycosyltransferase family 43 as putative orthologs to the putative ÎČ-1,4-xylan backbone elongating Arabidopsis IRX9, IRX9L, and IRX14 genes, respectively. We demonstrate that the over-expression of the closely related rice genes, in full or partly complement the two well-characterized Arabidopsis irregular xylem (irx) mutants: irx9 and irx14. Complementation was assessed by measuring dwarfed phenotypes, irregular xylem cells in stem cross sections, xylose content of stems, xylosyltransferase (XylT) activity of stems, and stem strength. The expression of OsIRX9 in the irx9 mutant resulted in XylT activity of stems that was over double that of wild type plants, and the stem strength of this line increased to 124% above that of wild type. Taken together, our results suggest that OsIRX9/OsIRX9L, and OsIRX14, have similar functions to the Arabidopsis IRX9 and IRX14 genes, respectively. Furthermore, our expression data indicate that OsIRX9 and OsIRX9L may function in building the xylan backbone in the secondary and primary cell walls, respectively. Our results provide insight into xylan biosynthesis in rice and how expression of a xylan synthesis gene may be modified to increase stem strength

    International audienceWe propose a method for multi-view reconstruction from videos adapted to dynamic cluttered scenes under uncontrolled imaging conditions. Taking visibility into account, and being based on a global optimization of a true spatio-temporal energy, it oilers several desirable properties: no need for silhouettes, robustness to noise, independent from any initialization, no heuristic force, reduced flickering results, etc. Results on real-world data proves the potential of what is, to our knowledge, the only globally optimal spatio-temporal multi-view reconstruction method

    Decreasing NF-ÎșB Expression Enhances Odontoblastic Differentiation and Collagen Expression in Dental Pulp Stem Cells Exposed to Inflammatory Cytokines

    Inflammatory response in the dental pulp can alter the collagen matrix formation by dental pulp stem cells and lead to a delay or poor healing of the pulp. This inflammatory response is mediated by cytokines, including interleukin-1ÎČ and tumor necrosis factor-α. In this study, it is hypothesized that suppressing the actions of these inflammatory cytokines by knocking down the activity of transcription factor Nuclear Factor–ÎșB will lead to dental pulp stem cell differentiation into odontoblasts and the production of collagen. Here, the role of Nuclear Factor–ÎșB signaling and its reduction was examined during odontogenic behavior in the presence of these cytokines. The results showed a significant increase in Nuclear Factor–ÎșB gene expression and p65 protein expression by interleukin-1ÎČ and tumor necrosis factor-α. Nuclear Factor–ÎșB activation in the presence of these cytokines decreased significantly in a dose-dependent manner by a Nuclear Factor–ÎșB inhibitor (MG132) and p65 siRNA. Down-regulation of Nuclear Factor–ÎșB activity also enhanced the gene expression of the odontoblastic markers (dentin sialophosphoprotein, Nestin, and alkaline phosphatase) and displayed an odontoblastic cell morphology indicating the promotion of odontogenic differentiation of dental pulp stem cells. Finally, dental pulp stem cells exposed to reduced Nuclear Factor–ÎșB activity resulted in a significant increase in collagen (I)-α1 expression in the presence of these cytokines. In conclusion, a decrease in Nuclear Factor-ÎșB in dental pulp stem cells in the presence of inflammatory cytokines enhanced odontoblastic differentiation and collagen matrix formation.The open access fee for this work was funded through the Texas A&M University Open Access to Knowledge (OAK) Fund

    Skeletal Site-Related Variation in Human Trabecular Bone Transcriptome and Signaling

    BACKGROUND: The skeletal site-specific influence of multiple genes on bone morphology is recognised, but the question as to how these influences may be exerted at the molecular and cellular level has not been explored. METHODOLOGY: To address this question, we have compared global gene expression profiles of human trabecular bone from two different skeletal sites that experience vastly different degrees of mechanical loading, namely biopsies from iliac crest and lumbar spinal lamina. PRINCIPAL FINDINGS: In the lumbar spine, compared to the iliac crest, the majority of the differentially expressed genes showed significantly increased levels of expression; 3406 transcripts were up- whilst 838 were down-regulated. Interestingly, all gene transcripts that have been recently demonstrated to be markers of osteocyte, as well as osteoblast and osteoclast-related genes, were markedly up-regulated in the spine. The transcriptome data is consistent with osteocyte numbers being almost identical at the two anatomical sites, but suggesting a relatively low osteocyte functional activity in the iliac crest. Similarly, osteoblast and osteoclast expression data suggested similar numbers of the cells, but presented with higher activity in the spine than iliac crest. This analysis has also led to the identification of expression of a number of transcripts, previously known and novel, which to our knowledge have never earlier been associated with bone growth and remodelling. CONCLUSIONS AND SIGNIFICANCE: This study provides molecular evidence explaining anatomical and micro-architectural site-related changes in bone cell function, which is predominantly attributable to alteration in cell transcriptional activity.     Effect of cell immobilization and pH on Scheffersomyces stipitis growth and fermentation capacity in rich and inhibitory media

    Background A wide range of value-added products can potentially be produced by bioprocessing hardwood spent sulfite liquors (HSSLs) that are by-products of pulp and paper industry with a high pentose sugar content. However, besides sugars, HSSLs contain considerable amounts of sulfonated lignin derivatives and acetic acid that inhibit the metabolic activity of most microorganisms. Scheffersomyces stipitis is a yeast with high capacity to ferment the pentose sugar xylose under appropriate microaerophilic conditions but it has limited tolerance to HSSL inhibitors. In the present study, cultivations of suspended and immobilized S. stipitis were compared in terms of growth capacity and by-product formation using rich medium and HSSL to investigate whether the immobilization of cells in calcium alginate beads could be a protection against inhibitors while favoring the presence of microaerophilic conditions. Results Whereas cell immobilization clearly favored the fermentative metabolism in rich medium, pH control was found to play a more important role than cell immobilization on the ethanol production efficiency from bio-detoxified HSSL (bdHSSL), leading to an improvement of 1.3-fold on the maximum ethanol productivity than using suspended cells. When immobilization and pH control were applied simultaneously, the ethanol yield improved by 1.3-fold with unchanged productivity, reaching 0.26 g ethanol.(g glucose\&#8201;+\&#8201;xylose)\&#8722;1. Analysis of the immobilized beads inside revealed that the cells had grown in the opposite direction of the cortex. Conclusions Immobilization and pH control at 5.5, when applied simultaneously, have a positive impact on the fermentative metabolism of S. stipitis, improving the ethanol production efficiency. For the first time light microscopic analysis of the beads suggested that the nutrient and mass transfer limitations played a more important role in the fermentation than a possible protective role against inhibitors.
