    A streamlined, automated protocol for the production of milligram quantities of untagged recombinant rat lactate dehydrogenase A using ÄKTAxpress™

    We developed an efficient, automated 2-step purification protocol for the production of milligram quantities of untagged recombinant rat lactate dehydrogenase A (rLDHA) from E. coli, using the ÄKTAxpress™ chromatography system. Cation exchange followed by size exclusion results in average final purity in excess of 93% and yields ~ 14 milligrams per 50 ml of original cell culture in EnPresso B media, in under 8 hrs, including all primary sample processing and column equilibration steps. The protein is highly active and coherent biophysically and a viable alternative to the more problematic human homolog for structural and ligand-binding studies; an apo structure of untagged rLDHA was solved to a resolution 2.29 Å (PDB ID 5ES3). Our automated methodology uses generic commercially available pre-packed columns and simple buffers, and represents a robust standard method for the production of milligram amounts of untagged rLDHA, facilitating a novel fragment screening approach for new inhibitors

    Cost of capital to the hospital sector.

    This paper provides estimates of the cost of equity and debt capital to for-profit and non-profit hospitals in the U.S. for the years 1972-83. The cost of equity is estimated using, alternatively, the Capital Asset Pricing Model and Arbitrage Pricing Theory. We find that the cost of equity capital, using either model, substantially exceeded anticipated inflation. The cost of debt capital was much lower. Accounting for the corporate tax shield on debt and capital paybacks by cost-based insurers lowered the net cost of capital to hospitals

    Ecosmart reinforcement for a masonry polycentric pavilion vault

    In the cultural life of modern societies great importance has acquired the preservation of existing and, in particular, ancient architectural heritage. With the inherent historical aspects, the economic implications have to be taken into account as well. Indeed, especially European cities and countries receive significant economic advantages by the existence of monuments and ancient suburbs. In this context, structural maintenance, strengthening and monitoring has gained an important academic and professional impulse. The present paper aims to present the results of a real scale experimental work regarding the application of an innovative seismic retrofitting technique for masonry walls and vaults by Hydraulic Lime Mortar strengthened by Glass Fiber Reinforced Polymer textile grids (HLM-GFRP) embedding new sensing systems as fiber optical sensors. The real scale specimen is a masonry polycentric pavilion vault that was damaged during the L’Aquila earthquake of April 2009. The need of eco compatibility of bonding material with masonry support implies the use of HLM-GFRP as strengthening system. On the other hand, the use of Fiber Bragg Grating (FBG) has a large number of advantages in opposite to electrical measuring methods. Example are: small sensor dimensions, low weight as well as high static and dynamic resolution of measured values, distributed sensing feature allowing to detect anomalies in load transfer between reinforcement and substrate and the location of eventual cracking patterns. A suitable Finite Element (FE) model is developed both to assess the effectiveness of the HLM-GFRP strengthening layers in retrofitting of the masonry vault and to define the strain field essential to the design of the FBG sensors network. © Gattulli et al

    Diffusion of surgical technology. An exploratory study.

    The study presents an empirical analysis of the diffusion patterns of five surgical procedures. Roles of payer mix, regulatory policies, physician diffusion, competition among hospitals, and various hospital characteristics such as size and the spread of technologies are examined. The principal data base is a time series cross-section of 521 hospitals based on discharge abstracts sent to the Commission on Professional and Hospital Activities. Results on the whole are consistent with a framework used to study innovations in other contexts in which the decisions of whether to innovate and timing depend on anticipated streams of returns and cost. Innovation tends to be more likely to occur in markets in which the more generous payers predominate. But the marginal effects of payer mix are small compared to effects of location and hospital characteristics, such as size and teaching status. Hospital rate-setting sometimes retarded diffusion.
