    A pharmacological approach using optical recording

    ATP, acting through P2X 2/P2X 3 receptor-channel complexes, plays an important role in carotid body chemoexcitation in response to natural stimuli in the rat. Since the channels are permeable to calcium, P2X activation by ATP should induce changes in intracellular calcium ([Ca 2+] i). Here, we describe a novel ex vivo approach using fluorescence [Ca 2+] i imaging that allows screening of retrogradely labeled chemoafferent neurons in the petrosal ganglion of the rat. ATP-induced [Ca 2+] i responses were characterized at postnatal days (P) 5-8 and P19-25. While all labeled cells showed a brisk increase in [Ca 2+] i in response to depolarization by high KCl (60 mM), only a subpopulation exhibited [Ca 2+] i responses to ATP. ATP (250 -1,000 μM) elicited one of three temporal response patterns: fast (R1), slow (R2), and intermediate (R3). At P5-8, R2 predominated and its magnitude was attenuated 44% by the P2X 1 antagonist, NF449 (10 μM), and 95% by the P2X 1/P2X 3/P2X 2/3 antagonist, TNP-ATP (10 μM). At P19-25, R1 and R3 predominated and their magnitudes were attenuated 15% by NF449, 66% by TNP-ATP, and 100% by suramin (100 μM), a nonspecific P2 purinergic receptor antagonist. P2X 1 and P2X 2 protein levels in the petrosal ganglion decreased with development, while P2X 3 protein levels did not change significantly. We conclude that the profile of ATP-induced P2X-mediated [Ca 2+] i responses changes in the postnatal period, corresponding with changes in receptor isoform expression.     Scalable low-latency optical phase sensor array

    Optical phase measurement is critical for many applications, and traditional approaches often suffer from mechanical instability, temporal latency, and computational complexity. In this paper, we describe compact phase sensor arrays based on integrated photonics, which enable accurate and scalable reference-free phase sensing in a few measurement steps. This is achieved by connecting multiple two-port phase sensors into a graph to measure relative phases between neighboring and distant spatial locations. We propose an efficient post-processing algorithm, as well as circuit design rules to reduce random and biased error accumulations. We demonstrate the effectiveness of our system in both simulations and experiments with photonics integrated circuits. The proposed system measures the optical phase directly without the need for external references or spatial light modulators, thus providing significant benefits for applications including microscope imaging and optical phased arrays

    Enteric helminths promote Salmonella co-infection by altering the intestinal metabolome

    Intestinal helminth infections occur pre dominantly in regions where exposure to enteric bacterial pathogens is also common. Helminth infections inhibit host immunity against microbial pathogens, which has largely been attributed to the induction of regulatory or type 2 (Th2) immune responses. Here we demonstrate an additional three-way interaction in which helminth infection alters the metabolic environment of the host intestine to enhance bacterial pathogenicity. We show that an ongoing helminth infection increased colonization by Salmonella independently of T regulatory or Th2 cells. Instead, helminth infection altered the metabolic profile of the intestine, which directly enhanced bacterial expression of Salmonella pathogenicity island 1 (SPI-1) genes and increased intracellular invasion. These data reveal a novel mechanism by which a helminth-modified metabolome promotes susceptibility to bacterial co-infection

    One at a time, live tracking of NGF axonal transport using quantum dots

    Retrograde axonal transport of nerve growth factor (NGF) signals is critical for the survival, differentiation, and maintenance of peripheral sympathetic and sensory neurons and basal forebrain cholinergic neurons. However, the mechanisms by which the NGF signal is propagated from the axon terminal to the cell body are yet to be fully elucidated. To gain insight into the mechanisms, we used quantum dot-labeled NGF (QD-NGF) to track the movement of NGF in real time in compartmentalized culture of rat dorsal root ganglion (DRG) neurons. Our studies showed that active transport of NGF within the axons was characterized by rapid, unidirectional movements interrupted by frequent pauses. Almost all movements were retrograde, but short-distance anterograde movements were occasionally observed. Surprisingly, quantitative analysis at the single molecule level demonstrated that the majority of NGF-containing endosomes contained only a single NGF dimer. Electron microscopic analysis of axonal vesicles carrying QD-NGF confirmed this finding. The majority of QD-NGF was found to localize in vesicles 50–150 nm in diameter with a single lumen and no visible intralumenal membranous components. Our findings point to the possibility that a single NGF dimer is sufficient to sustain signaling during retrograde axonal transport to the cell body

    SPRWeb: preserving subjective responses to website colour schemes through automatic recolouring

    Colours are an important part of user experiences on the Web. Colour schemes influence the aesthetics, first impressions and long-term engagement with websites. However, five percent of people perceive a subset of all colours because they have colour vision deficiency (CVD), resulting in an unequal and less-rich user experience on the Web. Traditionally, people with CVD have been supported by recolouring tools that improve colour differentiability, but do not consider the subjective properties of colour schemes while recolouring. To address this, we developed SPRWeb, a tool that recolours websites to preserve subjective responses and improve colour differentiability - thus enabling users with CVD to have similar online experiences. To develop SPRWeb, we extended existing models of non-CVD subjective responses to CVD, then used this extended model to steer the recolouring process. In a lab study, we found that SPRWeb did significantly better than a standard recolouring tool at preserving the temperature and naturalness of websites, while achieving similar weight and differentiability preservation. We also found that recolouring did not preserve activity, and hypothesize that visual complexity influences activity more than colour. SPRWeb is the first tool to automatically preserve the subjective and perceptual properties of website colour schemes thereby equalizing the colour-based web experience for people with CVD.Engineering and Applied Science

    Qualitative and Mixed Methods Social Media Research: A Review of the Literature

    Social media technologies have attracted substantial attention among many types of users including researchers who have published studies for several years. This article presents an overview of trends in qualitative and mixed methods social media research literature published from 2007 through 2013. A collection of 229 qualitative studies were identified through a systematic literature review process. A subset of 55 of these articles report studies involving a combination of qualitative and quantitative methods. Articles were reviewed, analyzed, and coded through a qualitative content analysis approach. Overall trends are presented with respect to the entire collection of articles followed by an analysis of mixed methods research approaches identified in the subset of 55 studies. The most commonly used research approaches involved collecting data from people through interview, focus group, and survey methodologies. Content analysis was the second most commonly used approach whereby researchers use Facebook posts, Tweets (Twitter posts), YouTube videos, or other social media content as a data source. Many of the studies involving combinations of quantitative and qualitative data followed a design resembling Creswell and Plano Clark’s basic mixed methods typology (e.g., convergent parallel, explanatory sequential, and exploratory sequential)

    Study of Zγ events and limits on anomalous ZZγ and Zγγ couplings in pp̄ collisions at s=1.96TeV

    We present a measurement of the Zγ production cross section and limits on anomalous ZZγ and Zγγ couplings for form-factor scales of Λ=750 and 1000 GeV. The measurement is based on 138 (152) candidates in the eeγ (μμγ) final state using 320(290)pb-1 of pp̄ collisions at s=1.96TeV. The 95% C.L. limits on real and imaginary parts of individual anomalous couplings are |h10,30Z|<0.23, |h20,40Z|<0.020, |h10,30γ|<0.23, and |h20,40γ|<0.019 for Λ=1000GeV. © 2005 The American Physical Society

    Reunifying from behind bars: A quantitative study of the relationship between parental incarceration, service use, and foster care reunification

    Incarcerated parents attempting to reunify with their children in foster care can find it difficult to complete the activities on their court-ordered case plans, such as drug treatment services and visitation with children. Although much has been written regarding the obstacles that are likely to interfere with reunification for incarcerated parents, very little quantitative research has examined the topic. This study uses secondary data to examine the incarceration experiences and reunification outcomes of a sample of 225 parents in one large urban California county. In multivariate analysis controlling for problems and demographics, incarcerated parents were less likely to reunify with their children; however, service use appeared to mediate this relationship, as the negative association between incarceration and reunification did not persist when service use was included as a variable in the model. Suggestions are made for policy and practice changes to improve reunification outcomes for this population of parents.
