Studie navrhuje prostorové uspořádání pozemků pro bydlení s ohledem na limity využití území a veškeré urbanistické a technické podmínky. Součástí řešení je napojení na dopravní a technickou infrastrukturu města a vymezení ploch veřejných prostranství. Výsledkem této bakalářské práce je návrh variantního řešení rozvržení území, přičemž pro vybranou variantu je zpracován návrh dopravní a technické infrastruktury. Nedílnou součástí práce je orientační ekonomické zhodnocení.The aim of this bachelor thesis is to make a territorial study of housing estate of location „Nad Bažantnicí“. The study proposes a spatial arrangement of land for housing with respect to land use limits and all urban and technical conditions. The solution includes connections to transport and technical infrastructure of the city and the delimitation of areas of public spaces. The result of this bachelor thesis is the proposal of the variant solution, where for the selected variant there is a proposal of transport and technical infrastructure.     Import 13/01/2017Obsahem práce je vypracování návrhu zástavby rozvojové plochy v obci Darkovice. Práce zahrnuje návrh variantní řešení rozvržení zástavby této plochy s ohledem na zachování veškerých technických a urbanistických zásad, respektování všech limit a dalších legislativních omezení. Součástí práce je návrh na vybudování a napojení dopravní a technické infrastruktury, vymezení ploch veřejných prostranství a návrh zeleně. Nedílnou součástí návrhu je studie jednoho z objektů a celkové orientační ekonomické zhodnocení. Výsledkem práce je vytvoření komplexní studie, která naváže na charakter okolního venkovského bydlení a scelí tuto plochu se stávající zástavbou v obci.The thesis contains proposal development area in the village Darkovice. Work includes design of alternative solutions layout development of this area in order to preserve all the technical and urban policy, respecting all limits and other legislative constraints. The work includes a proposal to build and connection of transport and technical infrastructure, defining the areas of public spaces and green design. Important parts of proposal are the study of one house and the indicative overall economic evaluation.     Between-Tactor Display Using Dynamic Tactile Stimuli

    Display of illusory vibration locations between physical vibrotactile motors (tactors) placed on the skin has the potential to reduce the number of tactors in distributed tactile displays. This paper presents a between-tactor display method that uses dynamic tactile stimuli to generate illusory vibration locations. A belt with only 6 vibration motors displays 24 targets consisting of on-tactor and between-tactor locations. On-tactor locations are represented by simply vibrating the relevant single tactor. Between-tactor locations are displayed by adjusting the relative vibration amplitudes of two adjacent motors, with either (1) constant vibration amplitudes or (2) perturbed vibration amplitudes (creating local illusory motion). User testing showed that perturbations improve recognition accuracy for in-between tactor localization

    Teachers in concordance for pseudo-labeling of 3D sequential data

    Automatic pseudo-labeling is a powerful tool to tap into large amounts of sequential unlabeled data. It is specially appealing in safety-critical applications of autonomous driving, where performance requirements are extreme, datasets are large, and manual labeling is very challenging. We propose to leverage sequences of point clouds to boost the pseudolabeling technique in a teacher-student setup via training multiple teachers, each with access to different temporal information. This set of teachers, dubbed Concordance, provides higher quality pseudo-labels for student training than standard methods. The output of multiple teachers is combined via a novel pseudo label confidence-guided criterion. Our experimental evaluation focuses on the 3D point cloud domain and urban driving scenarios. We show the performance of our method applied to 3D semantic segmentation and 3D object detection on three benchmark datasets. Our approach, which uses only 20% manual labels, outperforms some fully supervised methods.     Investigation of stacking faults in MOVPE-grown zincblende GaN by XRD and TEM

    X-ray diffraction and bright-field transmission electron microscopy are used to investigate the distribution and density of {111}-type stacking faults (SFs) present in a heteroepitaxial zincblende GaN epilayer with high phase purity, grown on a 3C-SiC/Si (001) substrate by metalorganic vapour-phase epitaxy. It is found that the 4° miscut towards the [110] direction of the substrate, that prevents the formation of undesirable antiphase domains, has a profound effect on the relative densities of SFs occurring on the different {111} planes. The two orientations of SFs in the [-110] zone, where the SF inclination angle with the GaN/SiC interface is altered by the 4° miscut, show a significant difference in density, with the steeper (111) SFs being more numerous than the shallower (-1-11) SFs by a factor of ~ 5 at 380 nm from the GaN/SiC interface. In contrast, the two orientations of SFs in the [110] zone, which is unaffected by the miscut, have densities comparable with the (-1-11) SFs in the [-110] zone. A simple model, simulating the propagation and annihilation of SFs in zincblende GaN epilayers, reproduces the presence of local SF bunches observed in TEM data. The model also verifies that a difference in the starting density at the GaN/SiC interface of the two orientations of intersecting {111} SFs in the same zone reduces the efficiency of SF annihilation.     Semisynthetic flavonoid 7-O-galloylquercetin activates Nrf2 and induces Nrf2-dependent gene expression in RAW264.7 and Hepa1c1c7 cells

    The natural flavonoid quercetin is known to activate the transcription factor Nrf2, which regulates the expression of cytoprotective enzymes such as heme oxygenase-1 (HO-1) and NAD(P)H:quinone oxidoreductase 1 (NQO1). In this study, a novel semisynthetic flavonoid 7-O-galloylquercetin (or quercetin-7-gallate, 3) was prepared by direct galloylation of quercetin, and its effect on the Nrf2 pathway was examined. A luciferase reporter assay showed that 7-O-galloylquercetin, like quercetin, significantly activated transcription via the antioxidant response element in a stably transfected human AREc32 reporter cell line. In addition, 7-O-galloylquercetin caused the accumulation of Nrf2 and induced the expression of HO-1 at both the mRNA and protein levels in murine macrophage RAW264.7 cells. The induction of HO-1 by 7-O-galloylquercetin was significantly suppressed by N-acetyl-l-cysteine and SB203580, indicating the involvement of reactive oxygen species and p38 mitogen-activated protein kinase activity, respectively. HPLC/MS analyses also showed that 7-O-galloylquercetin was not degalloylated to quercetin, but it was conjugated with glucuronic acid and/or methylated in RAW264.7 cells. Furthermore, 7-O-galloylquercetin was found to increase the protein levels of Nrf2 and HO-1, and also the activity of NQO1 in murine hepatoma Hepa1c1c7 cells. Taken together, we conclude that 7-O-galloylquercetin increases Nrf2 activity and induces Nrf2-dependent gene expression in RAW264.7 and Hepa1c1c7 cells

    Implementation of ultrasonic sensing for high resolution measurement of binary gas mixture fractions

    We describe an ultrasonic instrument for continuous real-time analysis of the fractional mixture of a binary gas system. The instrument is particularly well suited to measurement of leaks of a high molecular weight gas into a system that is nominally composed of a single gas. Sensitivity < 5 × 10−5 is demonstrated to leaks of octaflouropropane (C3F8) coolant into nitrogen during a long duration (18 month) continuous study. The sensitivity of the described measurement system is shown to depend on the difference in molecular masses of the two gases in the mixture. The impact of temperature and pressure variances on the accuracy of the measurement is analysed. Practical considerations for the implementation and deployment of long term, in situ ultrasonic leak detection systems are also described. Although development of the described systems was motivated by the requirements of an evaporative fluorocarbon cooling system, the instrument is applicable to the detection of leaks of many other gases and to processes requiring continuous knowledge of particular binary gas mixture fractions

    Effect of aerobic exercise on amyloid accumulation in preclinical Alzheimer’s: A 1-year randomized controlled trial

    Background Our goal was to investigate the role of physical exercise to protect brain health as we age, including the potential to mitigate Alzheimer’s-related pathology. We assessed the effect of 52 weeks of a supervised aerobic exercise program on amyloid accumulation, cognitive performance, and brain volume in cognitively normal older adults with elevated and sub-threshold levels of cerebral amyloid as measured by amyloid PET imaging. Methods and findings This 52-week randomized controlled trial compared the effects of 150 minutes per week of aerobic exercise vs. education control intervention. A total of 117 underactive older adults (mean age 72.9 [7.7]) without evidence of cognitive impairment, with elevated (n = 79) or subthreshold (n = 38) levels of cerebral amyloid were randomized, and 110 participants completed the study. Exercise was conducted with supervision and monitoring by trained exercise specialists. We conducted 18F-AV45 PET imaging of cerebral amyloid and anatomical MRI for whole brain and hippocampal volume at baseline and Week 52 follow-up to index brain health. Neuropsychological tests were conducted at baseline, Week 26, and Week 52 to assess executive function, verbal memory, and visuospatial cognitive domains. Cardiorespiratory fitness testing was performed at baseline and Week 52 to assess response to exercise. The aerobic exercise group significantly improved cardiorespiratory fitness (11% vs. 1% in the control group) but there were no differences in change measures of amyloid, brain volume, or cognitive performance compared to control. Conclusions Aerobic exercise was not associated with reduced amyloid accumulation in cognitively normal older adults with cerebral amyloid. In spite of strong systemic cardiorespiratory effects of the intervention, the observed lack of cognitive or brain structure benefits suggests brain benefits of exercise reported in other studies are likely to be related to non-amyloid effects

    Listeria monocytogenes is an important cause of maternal-fetal infections and serves as a model organism to study these important but poorly understood events. L. monocytogenes can infect non-phagocytic cells by two means: direct invasion and cell-to-cell spread. The relative contribution of each method to placental infection is controversial, as is the anatomical site of invasion. Here, we report for the first time the use of first trimester placental organ cultures to quantitatively analyze L. monocytogenes infection of the human placenta. Contrary to previous reports, we found that the syncytiotrophoblast, which constitutes most of the placental surface and is bathed in maternal blood, was highly resistant to L. monocytogenes infection by either internalin-mediated invasion or cell-to-cell spread. Instead, extravillous cytotrophoblasts-which anchor the placenta in the decidua (uterine lining) and abundantly express E-cadherin-served as the primary portal of entry for L. monocytogenes from both extracellular and intracellular compartments. Subsequent bacterial dissemination to the villous stroma, where fetal capillaries are found, was hampered by further cellular and histological barriers. Our study suggests the placenta has evolved multiple mechanisms to resist pathogen infection, especially from maternal blood. These findings provide a novel explanation why almost all placental pathogens have intracellular life cycles: they may need maternal cells to reach the decidua and infect the placenta
